Microinjection of the dopamine D2-receptor antagonist Raclopride into the medial preoptic area reduces REM sleep in lactating rats.

The medial preoptic area (mPOA) is a brain structure classically related to both non-REM (NREM) sleep and maternal behavior. Although the dopaminergic system is known to play a role in the control of the states of sleep and wakefulness, its effects within the mPOA on sleep are still not clear. Microinjection of the dopamine D2 receptor antagonist Raclopride into the mPOA has been shown to promote nursing postures in lactating dams with no effects on active maternal behavior. We hypothesized that the facilitation of nursing postures may be also associated with the promotion of NREM sleep. In or

Slow wave and rem sleep mechanisms are differently altered in hereditary pick disease associated with the TAU G389R mutation.

Sleep disturbances are found in the course of most dementing syndromes. We report a longitudinal polysomnographic and 18FDG-PET study in a 38-year-old male with FTDP17 carrying the Tau gene mutation G389R. All-night sleep EEG and wake cerebral glucose metabolism at rest (eyes/ears covered) of the preceding day were studied twice, eight months (Night 1; PET 1) and sixteen months (Night 2; PET 2) after the initial neurological evaluation. The Night 1 study showed sleep fragmentation associated to a short REM latency and a severe reduction of slow wave sleep, with relatively preserved NREM-REM sl

Carbachol microinjection into the caudal peribrachial area induces long-term enhancement of PGO wave activity but not REM sleep.

This study presents new findings of carbachol-induced long-term ponto-geniculo-occipital (PGO) enhancement lasting five days, but without REM sleep enhancement. A quantitative analysis of the number and types of bilateral PGO wave events during slow wave sleep with PGO activity (SP) and REM was performed in each of four cats over a period of six days following a single unilateral microinjection of carbachol nanospheres into the caudolateral peribrachial area. The results demonstrate increases in the summed total of all PGO wave events to continue for five days postcarbachol reaching a peak six

Spatiotemporal deep learning for early detection of isolated REM sleep behavior disorder and Parkinson's disease using functional MRI data.

This study aimed to develop and evaluate a spatiotemporal deep-neural-network (stDNN) using resting-state fMRI (rs-fMRI) data to identify brain biomarkers associated with isolated REM sleep behavior disorder (iRBD) and Parkinson's disease (PD) and to differentiate these conditions from controls. The final sample included 771 subjects, comprising 423 patients with PD, 144 with iRBD, and 204 healthy controls. stDNN model was applied to mean timeseries extracted for each subject from rs-fMRI data. By integrating spatio-temporal features, the network classified subjects based on distinct neural pa

Progression of Motor and Cognitive Functions in Isolated REM Sleep Behavior Disorder: A 7-Year Prospective Matched Cohort Study.

BACKGROUND: Although clinical markers (eg, motor and cognitive impairment) in isolated rapid eye movement sleep behavior disorder (iRBD) are associated with faster phenoconversion, their longitudinal trajectory patterns (linear or nonlinear) remain unclear. Additionally, evidence regarding the magnitude of neurodegenerative risk in iRBD compared to non-RBD individuals remains limited. OBJECTIVES: The aim was to investigate longitudinal changes in clinical markers and assess the magnitude of neurodegenerative risk in iRBD versus non-RBD subjects. METHODS: In this prospective matched cohort st

Focal ischemic stroke impairs REM sleep and inhibits c-Fos expression of SST neuron in EP of male rats.

PURPOSE: The basal ganglia contribute to sleep-wake regulation and are vulnerable to ischemic stroke. Specifically, the striatum, as an upstream input region, appears critically involved, since rapid eye movement (REM) sleep disruption occurs in both extensive (cortex and striatum) and focal striatal infarcts. Nonetheless, the post-stroke dynamics of REM sleep disturbances and their neural targets remain unclear, largely due to the technical limitations in animal models and complexity of stroke outcomes. METHODS: We modified the transient middle cerebral artery occlusion model in rats to indu

Cognitive training and promoting a healthy lifestyle in isolated REM sleep behavior disorder: The randomized controlled trial CogTrAiL-RBD.

INTRODUCTION: Isolated rapid eye movement (REM) sleep behavior disorder (iRBD) is a prodromal phase of Lewy body diseases and linked to a high risk for cognitive decline. Therefore, iRBD provides a suitable window for early intervention. METHODS: In the monocentric randomized controlled trial "Cognitive Training & a Healthy, Active Lifestyle Program for People with Isolated REM Sleep Behaviour Disorder" (CogTrAiL-RBD), the feasibility and behavioral effects of a 5-week digital, adaptive, multidomain cognitive training alongside a module promoting a healthy lifestyle (INT) were compared to a w

Decoding Prodromal Lewy Body Disease: Clinical Differences Between Isolated REM Sleep Behavior Disorder and Hyposmia With Dopamine Transporter Deficit.

BACKGROUND: Lewy body diseases (LBDs) are heterogeneous, and this variability is already evident in the prodromal phase. Several frameworks propose that prodromal heterogeneity reflects distinct phenotypic and biological subtypes, supported by differences in clinical profiles, imaging, and α-synuclein biomarkers. We provide a detailed clinical characterization of two enriched prodromal cohorts: isolated REM Sleep Behavior Disorder (iRBD) and Hyposmia with Dopamine Transporter Deficit. METHODS: This cross-sectional study included 360 participants with iRBD, 1101 with hyposmia and abnormal dopa

Self-Reported REM Sleep Behavior Disorder in Patients With Progressive Supranuclear Palsy: Clinical and 18F-Florzolotau PET Imaging Findings.

BACKGROUND AND OBJECTIVES: Rapid eye movement sleep behavior disorder (RBD) is increasingly recognized in patients with tauopathies, but its significance and underpinnings remain unclear. To address this gap, we investigated the prevalence of self-reported RBD in patients with progressive supranuclear palsy (PSP) and explored its clinical and imaging correlates using 18F-florzolotau PET imaging. METHODS: We consecutively enrolled patients meeting the 2017 Movement Disorder Society clinical criteria for PSP at a Chinese tertiary hospital between May 2019 and March 2022. Patients underwent comp

REM sleep behavior disorder: Relation between aggressiveness and emotions expressed in dream enactment and cognitive and anxiety/depression status.

Aggressiveness and negative emotions in dreams reports of patients with alfa-synucleinopathies have been associated with cognitive dysfunction. Observation of dream enactment episodes could be a more precise method to capture dream content in patients with REM sleep behavior disorder (RBD). Our objective was to assess the relation between aggressive and emotional dream enactment episodes in patients with RBD and cognition and depression/anxiety. Motor events (ME) during REM sleep were classified by visual inspection of video-PSG files. Cognition and anxiety/depression were assessed with MoCA a

Systematic review: REM sleep, dysphoric dreams and nightmares as transdiagnostic features of psychiatric disorders with emotion dysregulation - Clinical implications.

BACKGROUND: Fragmented rapid eye movement (REM) sleep disrupts the overnight resolution of emotional distress, a process crucial for emotion regulation. Emotion dysregulation, which is common across psychiatric disorders, is often associated with sleep disturbances. This systematic review explores how REM sleep and nightmares affect emotion processing and regulation in individuals with psychiatric disorders where emotion dysregulation is a key concern, suggesting novel sleep-related treatment pathways. METHODS: We performed a PRISMA-compliant systematic search of the PUBMED, Web of Science, a

Downregulation of microRNA-330-5p induces manic-like behaviors in REM sleep-deprived rats by enhancing tyrosine hydroxylase expression.

AIM: In our pilot study, we found an increase in tyrosine hydroxylase (Th) mRNA expression in the prefrontal cortex of 72-h REM sleep-deprived (SD) rats, a mania model. Additionally, the expression levels of miR-325-3p, miR-326-3p, and miR-330-5p, the predicted target miRNAs on TH, were significantly decreased. Based on these results, in this study, we investigated whether miRNA-325-3p, miR-326-3p, and miR-330-5p modulate TH and manic-like behaviors in SD rats. METHODS: Manic-like behaviors were assessed using the open field test (OFT) and elevated plus-maze (EPM) test. The direct binding act

Twenty Years After: Another Case Report of Melatonin Effects on REM Sleep Behavior Disorder, Using Serial Dopamine Transporter Imaging.

INTRODUCTION: REM sleep behavior disorder (RBD) is considered a prodromal phase of α-synucleinopathy like Parkinson disease (PD). PD is characterized by a progressive decline of dopamine (DA) in the striatum. Here we report the surprising increase in DA transporter density over successive years in an RBD patient treated with melatonin. CASE PRESENTATION: A then 72-year-old man was clinically suspected to suffer from PD in 2011. DA transporter scintigraphy (DaTSCAN) revealed reduced DA transporter density, and the patient was diagnosed with developing PD. Because of outacting dreams every nigh

[Revised 2017 Clinical Diagnostic Criteria for Dementia with Lewy Bodies: Inclusion of REM Sleep Behavior Disorder and Indicative Biomarkers].

Although dementia with Lewy bodies (DLB) is the second most common neurodegenerative disorder after Alzheimer disease, its diagnostic sensitivity in clinical practice is sub-optimal. In 2017, the DLB clinical diagnostic criteria were modified to include the additional core feature of REM sleep behavior disorder (RBD) and three new indicative biomarkers. With this revision, clinically probable DLB is represented by two or more core features (fluctuations, parkinsonism, visual hallucinations, or RBD) or by one core feature plus one indicative biomarker (low striatal dopamine transporter uptake,

Evening-type military veterans report worse lifetime posttraumatic stress symptoms and greater brainstem activity across wakefulness and REM sleep.

Evening chronotypes exhibit increased rates of affective dyregulation and sleep disturbances (e.g., insomnia and nightmares). Such symptoms are common to military veterans with posttraumatic stress disorder (PTSD); however, the influence of chronotype on this population remains unknown. We examined behavioral, psychological, and neural correlates of chronotype in 36 combat-exposed military veterans with varying degrees of posttraumatic stress symptomatology. We employed FDG-PET to assess neural activity across wakefulness and rapid eye movement (REM) sleep. We used polysomnography and diaries

The neuronal network responsible for paradoxical sleep and its dysfunctions causing narcolepsy and rapid eye movement (REM) behavior disorder.

Rapid eye movement (REM) sleep behavior disorder (RBD) is a parasomnia characterized by the loss of muscle atonia during paradoxical (REM) sleep (PS). Conversely, cataplexy, one of the key symptoms of narcolepsy, is a striking sudden episode of muscle weakness triggered by emotions during wakefulness, and comparable to REM sleep atonia. The neuronal dysfunctions responsible for RBD and cataplexy are not known. In the present review, we present the most recent results on the neuronal network responsible for PS. Based on these results, we propose an updated integrated model of the mechanisms res

Deciphering the heterogeneity of prodromal α-synucleinopathy: linking brain biotyping and gut dysbiosis in isolated REM sleep behaviour disorder.

BACKGROUND: Isolated/idiopathic REM sleep behaviour disorder (iRBD) is a specific prodrome of α-synucleinopathies. We previously identified two distinct clinical-neuroimaging biotypes in iRBD: Biotype 1, marked by widespread cortical-subcortical-cerebellar atrophy with motor and cognitive deficits, and Biotype 2, with preserved brain structure and neurocognition. Converging evidence implicates the gut-brain axis in early α-synucleinopathy, yet its role in these divergent iRBD phenotypes remains unexplored. METHODS: We analysed faecal microbiota using shotgun metagenomic sequencing and structu

Extracellular water-to-total body water ratio predicts phenoconversion in REM sleep behavior disorder: A bioimpedance-based cohort study.

BACKGROUND: REM sleep behavior disorder (RBD) is a prodromal manifestation of α-synucleinopathies, with most patients converting to Parkinson's disease (PD), dementia with Lewy bodies (DLB), or multiple system atrophy (MSA) within 10-15 years. Practical, non-invasive biomarkers for predicting phenoconversion remain limited. OBJECTIVE: To evaluate whether bioelectrical impedance analysis (BIA) parameters, specifically extracellular water-to-total body water ratio (ECW/TBW) and phase angle (PhA), are associated with phenoconversion risk and motor symptom severity in patients with RBD. METHODS:

Chronic REM sleep deprivation leads to manic- and OCD-related behaviors, and decreases hippocampal BDNF expression in female rats.

BACKGROUND: Rapid-eye movement (REM) sleep deprivation (SD) can induce manic-like behaviors in rodents. On the other hand, lithium, as one of the oldest drugs used in neuropsychiatric disorders, is still one of the best drugs for the treatment and control of bipolar disorder. In this study, we aimed to investigate the role of chronic short-term REM SD in the induction of manic-like behaviors in female rats. METHODS: The rats were exposed to REM SD for 14 days (6 hours/day). Lithium was intraperitoneally injected at the doses of 10, 50, and 100 mg/kg. RESULTS: REM SD induced hyperactivity and

BACKGROUND AND PURPOSE: Migraine aura, near-death experiences (NDEs), and rapid eye movement (REM) sleep intrusions might share common mechanisms. Here, we investigated the prevalence of NDEs and REM sleep intrusions in people with migraine. We hypothesized that NDEs and REM sleep intrusions are more prevalent in migraine patients with aura than in those without. METHODS: We conducted a prospective cross-sectional cohort study at a tertiary headache center, based on a prespecified sample size (n = 808).
