Linkage and association of successful aging to the 6q25 region in large Amish kindreds.

Successful aging (SA) is a multidimensional phenotype involving living to older age with high physical function, preserved cognition, and continued social engagement. Several domains underlying SA are heritable, and identifying health-promoting polymorphisms and their interactions with the environment could provide important information regarding the health of older adults. In the present study, we examined 263 cognitively intact Amish individuals age 80 and older (74 SA and 189 "normally aged") all of whom are part of a single 13-generation pedigree. A genome-wide association study of 630,309

Novel autism subtype-dependent genetic variants are revealed by quantitative trait and subphenotype association analyses of published GWAS data.

The heterogeneity of symptoms associated with autism spectrum disorders (ASDs) has presented a significant challenge to genetic analyses. Even when associations with genetic variants have been identified, it has been difficult to associate them with a specific trait or characteristic of autism. Here, we report that quantitative trait analyses of ASD symptoms combined with case-control association analyses using distinct ASD subphenotypes identified on the basis of symptomatic profiles result in the identification of highly significant associations with 18 novel single nucleotide polymorphisms

An infectious aetiology for childhood acute leukaemia: a review of the evidence.

There are three current hypotheses concerning infectious mechanisms in the aetiology of childhood leukaemia: exposure in utero or around the time of birth, delayed exposure beyond the first year of life to common infections and unusual population mixing. No specific virus has been definitively linked with childhood leukaemia and there is no evidence to date of viral genomic inclusions within leukaemic cells. The case-control and cohort studies have revealed equivocal results. Maternal infection during pregnancy has been linked with increased risk whilst breast feeding and day care attendance i

High Patient Willingness to Grant Broad Consent for Real-World Data Use in Rheumatology-Implications for Real-World Data Platform Governance: Cross-Sectional Study.

BACKGROUND: Medical real-world data (RWD) are often siloed across organizations, making them inaccessible for research. Unlocking these data could advance clinical research and patient care. The pan-European Data Nexus platform (DNP) links RWD, facilitating its use, for example by artificial intelligence (AI) tools, to support the generation of real-world evidence. In Europe, particularly Germany, the secondary use of health data is governed by stringent regulatory requirements, including informed consent. OBJECTIVE: This study evaluated the informed broad consent form for the RWD (Data Nexus

Hantaviruses in rodents and humans, Xi'an, PR China.

Xi'an, the capital of Shaanxi province, located in north-west China, is one of the major endemic areas for haemorrhagic fever with renal syndrome (HFRS). In this study, the epidemiological data of HFRS in Xi'an from 1959 to 2010, especially in the past ten years (2001-2010), were surveyed. The features of hantavirus (HV) host carriers, the molecular characteristics of the HV S gene from hosts and patients, and the genome of the viral isolate were also investigated. Data showed that there might be a ten-year cycle of HFRS in Xi'an. Although the main population group infected over the past ten y

Genetic Etiology and Neurodevelopmental Outcomes in Children With Craniosynostosis: A 15-Year Single-Center Retrospective Study.

BACKGROUND: The traditional distinction between syndromic and nonsyndromic craniosynostosis has guided clinical management, but recent genomic advances suggest these categories represent a continuous spectrum rather than discrete entities. Data on genetic etiologies and long-term neurodevelopmental outcomes in Asian populations remain limited. METHODS: The authors conducted a retrospective cohort study of 68 consecutive children diagnosed with craniosynostosis at a single tertiary center between January 2011 and November 2025. Genetic testing included whole-genome/whole-exome sequencing, targ

Welcome to the house-warming party: social interactions and antagonism against Listeria monocytogenes in multi-species biofilms.

UNLABELLED: Listeria monocytogenes integrates into multi-species biofilms and persists in food processing facilities. Its persistence depends on the composition of the microbial communities, but those species that accommodate L. monocytogenes, or not, remain to be identified. Moreover, the decision-making on biofilm vs pellicle formation in microbial communities remains poorly understood. It was the aim of this study to identify communities that accommodate Listeria, or not, and to observe biofilm vs pellicle formation in multi-species biofilms. We quantified L. monocytogenes in six reconstitu

Review: Early and late determinants of puberty in ruminants and the role of nutrition.

This article reviews the scientific literature on puberty with a focus on ruminants and draws inference, where appropriate, from recent findings in transgenic mouse models and human pathology. Early genetic determinants of puberty have been discovered in humans suffering from hypogonadotropic hypogonadism or central precocious puberty. Transgenic mouse models selected on the basis of the causative defective genes helped in discovering the cellular and molecular mechanisms involved. Most of the genes found are involved in the development of neuroendocrine networks during embryo development and

Starter Unit Promiscuity of Dictyostelid Type III Polyketide Synthases Generates Diverse Acylphloroglucinols with Antimicrobial Activity.

Cavenderia subdiscoidea and Cavenderia ungulata are dictyostelid social amoebae found in northern Thailand. Genomic analysis identified two type III polyketide synthase genes, designated cspks and cupks. Heterologous expression of both genes in Aspergillus oryzae NSAR1 yielded a variety of acylphloroglucinols derived from different starter units. Among these were cavenderic acids A-E (1-5) and two related ester derivatives (6-7). Compounds 1 and 2 are reported here for the first time as natural products, whereas compounds 3-7 are new compounds. Structural analysis suggested that compounds 1-7

From predicted bacteriocins to ecological function.

Genome and metagenome mining have uncovered large repertoires of predicted bacteriocin loci, but ecological interpretation has lagged behind discovery. This review uses bacteriocins as a focused model for a broader problem in antimicrobial gene prediction: sequence identifies encoded potential, but it does not establish expression, product deployment, target engagement, or community-level consequence. We distinguish four forms of bacteriocin "silence": transcriptional silence, where loci are not detectably expressed under tested conditions; conditional silence, where expression or activity eme

Structural Variant and Repeat Expansion Findings Identified by Optical Genome Mapping in Complex Autism Spectrum Disorder With Concomitant Neurodevelopmental Disorders.

Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder characterized by persistent deficits in social communication and interaction, along with restricted, repetitive patterns of behavior, interests, or activities. Single-nucleotide variants (SNVs) and structural variants (SVs), including copy-number variants (CNVs), have been reported as important contributors to the genetic basis of ASD. In this study, we evaluated the diagnostic contribution of optical genome mapping (OGM) as a complementary cytogenomic approach in a selected ASD cohort enriched for complex ASD with developmental d

The Maculalactone Biosynthetic Gene Cluster, a Cryptic Furanolide Pathway Revealed in Nodularia sp. NIES-3585.

Cyanobacteria have long been recognized as a prolific source of bioactive natural products (NPs). Among these are the furanolides, a structurally diverse class of compounds first discovered in the 1980s. Furanolides are characterized by a γ-butyrolactone core bearing aromatic or aliphatic substituents at the α- and β-positions and an aromatic substituent at the γ-position. Recent advances in understanding the genetic basis of furanolide biosynthesis have enabled genome mining approaches to discover related cryptic furanolide biosynthetic gene clusters (BGCs). In this work, we identified and cl

Large Tuberculosis Outbreaks - United States, 2017-2023.

During 2017-2023, based on an analysis of national genomic and tuberculosis (TB) case surveillance data, 50 large TB outbreaks (10 or more related TB cases in a 3-year period) involving 1,092 cases were identified in 23 states. Compared with 61,993 other persons who received a diagnosis of TB during this period, persons included in large outbreaks were more frequently U.S.-born (79% versus 26%), and a higher percentage reported substance use (27% versus 12%), homelessness (9% versus 5%), and incarceration (11% versus 3%). Approximately one fourth of these large outbreak-related cases were iden

Investigation of the transcriptome and metabolome of the cerebral cortex and testes in Cntnap4-deficient mice.

BACKGROUND: Autism spectrum disorder (ASD) involves challenges in social interaction and communication and repetitive behaviours. CNTNAP4 is implicated in neuronal signalling, and its deficiency plays a role in ASD. Transcriptomic analyses revealed similar gene expression between the brain and in humans as well as in mice. However, the relationships between the brain and testicular gene expression profiles and metabolism in ASD remain unclear. In this study, the effects of Cntnap4 deletion on gene expression and metabolic profiles in the cerebral cortex and testes were investigated to better u

Genomic Evidence for Dual Introductions, Limited Gene Flow and Niche Preferences in the Invasive Wasp Vespula germanica in South Africa.

Biological invasions are major drivers of recent biodiversity changes, yet the genetic structure and ecological mechanisms underlying invasion dynamics remain poorly resolved in invasive social insects. In South Africa, the European wasp Vespula germanica (Hymenoptera: Vespidae), introduced in the 1970s, has spread remarkably more slowly than in other regions and its geographical distribution in the country remains limited. Although two mitochondrial haplotypes have been reported in South Africa, the species' range expansion and fine-scale population structure remain poorly understood. Using 2
