We report the cloning, expression and functional characterisation of a peroxidase belonging to the peroxiredoxin family from the potato cyst nematode Globodera rostochiensis, the first molecule of this type from any nematode parasitic on plants. The G. rostochiensis peroxiredoxin catalyses the breakdown of hydrogen peroxide, but not cumene or t-butyl hydroperoxide, in a trypanosomatid reducing system comprising trypanothione reductase, trypanothione and tryparedoxin. In common with its homologues from Onchocerca volvulus and Brugia malayi, the G. rostochiensis enzyme is present on the surface

Tomato fruit ripening is a critical developmental process that determines the fruit quality. However, the regulatory mechanisms underlying the ripening initiation remain incompletely understood. Here, we characterized SlVTE3(2), a tocopherol-biosynthesis-related methyltransferase, and investigated its role in tomato fruit ripening. CRISPR/Cas9-mediated mutation of SlVTE3(2) accelerated ripening initiation, resulting in earlier ethylene production, faster softening, and an earlier color transition, whereas overexpression delayed these processes. Transcriptome analysis showed that SlVTE3(2) affe

The phosphate-solubilizing bacteria (PSB) can play a key role in converting unavailable phosphate to available form for root uptake as an environmentally friendly alternative to phosphate-based fertilizers. Four Bacillus spp. strains were assessed for their potential to boost soil health, promote tomato growth and fruit quality and reduce soilborne fungal diseases. B. amyloliquefaciens SV30 was efficient in increasing by 55 and 18% the available phosphate level in soil and in tomato leaves, respectively. The soil bacterial population and alkaline phosphatase activity were significantly improve

Optimizing light environments is essential for maximizing crop productivity and resource-use efficiency in plant factories with artificial lighting. Specifically, the frequency of light-dark cycles within a 24-h period interacts with the intrinsic circadian rhythms of plants, thereby influencing carbon metabolism and photosynthetic efficiency. In this study, we examined the effects of different light-dark cycle frequencies on the growth and photosynthetic activity of cucumber and tomato seedlings. The light-dark cycle frequencies were established as follows: Cycle 1 (16/8 h), 2 (8/4 h), 4 (4/2

Polysaccharide modulation is an effective strategy for tailoring the texture and stability of plant-based fat substitutes. This study investigated the modulatory effects of κ-carrageenan, konjac gum, and pullulan at three mass ratios (emulsion:polysaccharide = 5:1, 4:1, 3:1) on the stabilization of Pickering emulsion gels containing potato-soy protein composite nanoparticles. The results indicated that the modulatory effects were dependent on the type of polysaccharide. The pullulan group (3,1) imparted the smallest oil droplet size and optimal freeze-thaw stability to the emulsion; its gel wa

Tomato quality is shaped by both pre-harvest growing conditions and post-harvest factors; however, these aspects are often studied independently. This study explored the extent to which pre-harvest melatonin treatment affects tomato yield, quality and shelf life in terms of firmness, organoleptic properties (total sugar content and acidity) and bioactive constituents, with a focus on antioxidants (lycopene and vitamins C and E). Melatonin-treated plants were slightly more productive (7.8%) and yielded fruit with greater firmness (9.5%) at harvest. When stored at room temperature, tomatoes redd

Tomatoes are vital crops globally and they provide numerous nutritional benefits, including vitamins, lycopene, and gamma-aminobutyric acid (GABA). This study utilized CRISPR/Cas9 technology to introduce mutations in two genes, HWS (Hawaiian Skirt) and GAD3 (glutamate decarboxylase 3), to enhance tomato quality, increase yield stability, and improve parthenocarpic traits. Our results demonstrated that HWS and GAD3 mutations led to higher sugar (Brix) and GABA content, as well as the induction of facultative parthenocarpy, especially at elevated temperatures. Moreover, facultative parthenocarpy

In this study, the effects of Phlebopus portentosus polysaccharides on the physicochemical properties and digestibility of potato starch (PS) were systematically evaluated. Alkali-extracted polysaccharides from Phlebopus portentosus (APBP) were fractionated into three subfractions (APBP-30, APBP-70, and APBP-80) through gradient ethanol precipitation. These subfractions differed in monosaccharide composition and molecular weight. Further, PS-APBP subfraction composites were prepared and investigated for their physicochemical and structural properties, as well as in vitro digestibility. Pasting

The credibility of nutritional research is dependent on the rigor with which studies are conducted and the ability for independent assessment to be performed. Despite the importance of these, more work is needed in the field of nutrition to buttress the trustworthiness of nutrition research. To develop and apply a process for evaluating the rigor, reproducibility, and verifiability of nutritional research, using the relationship between potato consumption and Colorectal Cancer (CRC) as a case study. We updated existing systematic reviews to include studies on potatoes and CRC, assessing their

BACKGROUND: Fusarium wilt of tomato (Solanum lycopersicum L.), caused by Fusarium oxysporum f. sp. lycopersici (FOL), is a devastating soil-borne disease. Due to the limitations of conventional methods, sustainable biocontrol strategies are required. Bacillus velezensis is considered a promising biocontrol agent; however, the strain-specific nature of its antifungal and plant-growth-promoting mechanisms demands further investigation. RESULTS: We demonstrated the efficacy of B. velezensis BV25 against tomato Fusarium wilt. In vitro assays showed that FOL growth was significantly inhibited by bo…

This study investigated the effects of the hydrogel-to-oleogel ratio and potato protein isolate (PPI) concentration on the printing and post-heating properties of food-grade bigels. Bigels with hydrogel-to-oleogel ratios of 10:0, 7:3, 5:5, 3:7, and 0:10 were evaluated for physical stability, rheological behavior, structural recovery, and 3D printing performance. Among the biphasic formulations, H7O3 exhibited the highest liquid-binding capacity (99.43%) and structural recovery ratio (88.7%) and produced lateral dimensions closest to those of the CAD model. H7O3 was therefore selected for PPI i

Tomato spotted wilt virus (TSWV) is a formidable plant pathogen, inflicting severe economic losses in agriculture due to its broad host range and insect vector transmission. In this study, TSWV was identified in tomato plants in Muş province, Turkey, using molecular methods, and its coat protein (CP) gene was sequenced. The Muş 49 strain shared a remarkable 97% nucleotide similarity with global TSWV isolates and exhibited close phylogenetic relationships with strains from Turkey, Hungary, Bulgaria, and Serbia. Notably, the findings suggest that TSWV genetic diversity is independent of host pla

The increase of plant parasitic nematode resistance poses a great threat to agricultural and forestry production. To search for nematicides with novel mechanism of action, nematicidal activity of the 29 pyrazine compounds were tested. Compound C2 showed excellent nematicidal activity against Bursaphelenchus xylophilus, Meloidogyne incongnita, and Aphelenchoides besseyi, with LC50 values were 2.3, 3.3, and 10.6 mg/L, respectively, and the pot experiment showed that C2 reduced the number of tomato roots knots to a certain extent. In addition, C2 significantly inhibited egg hatching, locomotion a

BACKGROUND: Recently, oleogel has been proposed as an excellent substitute for conventional vegetable oils for deep-frying applications. Waxes are the most used oleogelators and are generally recognized as safe (GRAS) by the Food and Drug Administration (FDA). The properties of oleogels prepared from different types of waxes vary in their physiochemical composition, which may affect their deep-frying performance.
RESULTS: In this study, soybean oil-based oleogels prepared from four different types of waxes (i.e., beeswax, carnauba wax, candelilla wax, and rice bran wax) were developed to eval

A nuclear-localized cysteine desulfhydrase, LCD1, plays a crucial role in mediating endogenous hydrogen sulfide production in tomatoes. However, the mechanism underlying the nuclear localization of SlLCD1 is not yet fully understood. In this study, it was found that SlLCD1 specifically interacted with nuclear import receptor importin α3 (SlIMPA3). Furthermore, it was demonstrated that silencing SlIMPA3 through virus-induced gene silencing or introducing mutations in SlIMPA3 via CRISPR/Cas9 significantly accelerated fruit ripening. Moreover, enhanced chlorophyll degradation, carotenoid accumula

Nitric oxide (NO), a pivotal redox signaling molecule, coordinates plant development and immune responses through S-nitrosylation-mediated protein modification. While NO-dependent S-nitrosylation fine-tunes immune responses, whether pathogens hijack this process to subvert plant immunity remains unclear. Here it is shown that S-nitrosoglutathione reductase (GSNOR), which maintains NO homeostasis by degrading S-nitrosoglutathione (GSNO), positively regulates tomato resistance to Phytophthora capsici. Active-site mutations in GSNOR abolished its function in plant defense. Remarkably, GSNOR is ma

This study investigates the adsorption of Congo red (CR) dye onto laser-induced graphene derived from potato skins (LIGPS), a graphene-like material produced via rapid and cost-effective CO2 direct laser writing. Raman spectroscopy and X-ray diffraction (XRD) confirmed the graphitic nature of LIGPS. Activation with ZnCl₂ (LIGPS50) altered the morphology of the material, as observed in SEM images. Additionally, ZnCl₂ activation significantly increased the surface area from 11.256 to 126.696 m2 g⁻1, total pore volume from 0.016 to 0.253 cm3 g⁻1, and average pore diameter from 4.82 to 7.99 nm. The processes of morphogenesis and differentiation are crucial for leaf development, with the duration of the morphogenetic window influencing final leaf shape. Leaves at different developmental stages exhibit distinct morphological and physiological characteristics that may influence their ability to resist pathogens, and disease resistance has been linked to developmental stage in many plant species. To understand how leaf development impacts disease resistance, we examined the immunity of leaves at distinct developmental stages, exploring the role of hormonal pathways and the impact of leaf

The European Farm-to-Fork strategy partially focuses on a shift towards alternative protein. Among these, potato protein is promising, because it is nutritious, low allergenic and highly techno-functional. However, research on the impact of temperature on potato protein gel properties under salt conditions typical for food systems is lacking. Therefore, the effect of temperature (50 °C, 60 °C, 70 °C or 90 °C, selected based on differential scanning calorimetry) of isothermal treatment on potato protein aggregation and gelation was studied in a model system containing 250 mM NaCl. Two new analy
