Identification of Bacitracin Resistance Mechanisms in Streptococcus sobrinus.

Streptococcus sobrinus is a member of the Streptococcus mutans group and is a cariogenic bacterium similar to S. mutans. One of the characteristics of these bacteria is bacitracin resistance, and BceAB (MbrAB), an ABC transporter of S. mutans, has been reported to be involved in bacitracin resistance. However, the factors responsible for bacitracin resistance in S. sobrinus are unknown. In this study, 11 S. sobrinus strains were isolated from the oral cavity, and their bacitracin susceptibility was assessed. The results showed that 9 strains were low-susceptible to bacitracin (MIC = 128-512 µg…

Selection of streptococcal glucan-binding protein C specific DNA aptamers to inhibit biofilm formation.

Streptococcus mutans is a commensal oral bacterium, yet its capacity for extensive biofilm formation is a major contributor to dental caries. This study presents a novel biofilm inhibition strategy by targeting GbpC, a cornerstone protein in S. mutans biofilm architecture, with specific DNA aptamers. Using SELEX (Systematic Evolution of Ligands by EXponential enrichment), we selectively targeted the extracellular domain of GbpC while incorporating structurally similar antigen I/II protein and a GbpC-deficient S. mutans strain as counter-targets to ensure high specificity. Aptamer selection was…

Probiotics for caries prevention: A narrative review.

Dental caries, a widespread global health problem, results from oral microbiome dysbiosis dominated by acidogenic pathogens like Streptococcus mutans. Conventional preventive methods, such as fluoride and antimicrobial rinses, often lack specificity and can disrupt beneficial microbes. Probiotics are live, health-promoting bacteria or yeasts commonly found in foods such as yogurt or in dietary supplements. They help restore microbial balance and have emerged as a promising strategy for caries management. This review explores the mechanisms, efficacy, and clinical applications of probiotics in …
