From aggregate structure to disease trajectory: the role of the proteostasis network.

Neurodegenerative diseases are characterized by the accumulation of a small number of misfolded proteins, yet they differ markedly in cellular vulnerability and clinical outcome. We propose that this heterogeneity arises from the interaction between two largely orthogonal variables: the structural identities of protein aggregates and the identities of the cellular proteostasis networks (PNs) that engage them. Protein aggregates exist as structurally diverse assemblies with distinct intrinsic properties. At the same time, PNs vary between cell types, disease states and ageing in their compositi

A novel intrarenal multimodal 2D/3D registration algorithm and preliminary phantom study.

Retrograde intrarenal surgery (RIRS) is a widely utilized diagnostic and therapeutic tool for multiple upper urinary tract pathologies. The image-guided navigation system can assist the surgeon to perform precise surgery by providing the relative position between the lesion and the instrument after the intraoperative image is registered with the preoperative model. However, due to the structural complexity and diversity of multi-branched organs such as kidneys, bronchi, etc., the consistency of the intensity distribution of virtual and real images will be challenged, which makes the classical

New insights on the role of the insular cortex and habenula in OSA.

PURPOSE: Abnormal structure or function in the central nervous system (CNS) can also affect obstructive sleep apnea (OSA). Because human afferent and motor pathways that regulate apnea are still poorly understood, it is not possible to modify the behavior of motor neurons to control airway function. The purpose of this article is to clear the central control mechanism of genioglossus (GG) and to discuss how altered activity in the limbic system and its related structures might affect OSA development, in order to provide help for the treatment of this disease. METHODS: Functional magnetic reso

Exploration of biomaterial-tissue integration in heterogeneous microporous annealed particle scaffolds in subcutaneous implants over 12 months.

Microporous annealed particle (MAP) scaffolds are comprised of hydrogel microparticles with inter- and intra-particle cross-links that provide structure and cell-scale porosity, making them an increasingly attractive option for injectable tissue augmentation. Many current injectable biomaterials create a substantial foreign body response (FBR), while MAP scaffolds mitigate this response and have the potential to facilitate the formation of new tissue, though this de novo tissue formation is poorly understood. Here, we leverage a subcutaneous implant model to explore the maturation of MAP impla

An Exploratory Study into the Implantation of Arytenoid Cartilage Scaffold in the Horse.

Respiratory function in the horse can be severely compromised by arytenoid chondritis, or arytenoid chondropathy, a pathologic condition leading to deformity and dysfunction of the affected cartilage. Current treatment in cases unresponsive to medical management is removal of the cartilage, which can improve the airway obstruction, but predisposes the patient to other complications like tracheal penetration of oropharyngeal content and dynamic collapse of the now unsupported soft tissue lateral to the cartilage. A tissue engineering approach to reconstructing the arytenoid cartilage would repr

Invasion of Clavibacter michiganensis into the Xylem and Surrounding Tissues Drives Histological Alterations and Disease Development in Tomato.

Clavibacter michiganensis is a xylem-colonizing and seed-borne phytopathogen that causes bacterial canker disease in tomato plants. In this study, we aimed to determine the colonization patterns of C. michiganensis, as well as the histological changes in tomato plants during infection. To achieve these, GFP-tagged C. michiganensis was inoculated into tomato plants using three methods: stem injection, soil drenching with or without prior root cutting, and cotton-pad inoculation. After inoculation, bacterial colonization was observed using GFP signals, and histological changes were examined usin

Bedinvetmab (Librela/Beransa) in dogs raises safety concerns, including rapidly progressive osteoarthritis, and warrants vigilant adverse event reporting.

Bedinvetmab (Librela/Beransa) is a canine anti-nerve growth factor monoclonal antibody (aNGFmAb) approved for the management of osteoarthritis pain. By neutralizing NGF, it reduces nociceptive signaling and improves comfort. However, NGF plays critical roles in neural, bone, and cartilage homeostasis across species. Multiple studies indicate that NGF contributes to bone remodeling and cartilage repair by modulating osteoblast, osteoclast, and chondrocyte activity, processes that are highly active in osteoarthritic joints. In humans, rodents, and rabbits, inhibition of NGF is associated with ra

Bone Toxicity Case Report Combining Encorafenib, Cetuximab and WNT974 in a Phase I Trial.

BACKGROUND/AIM: More than 90% of colorectal cancers (CRC) have alterations in WNT signaling. Eight to ten percent of patients with metastatic Kirsten rat sarcoma virus - wild type (KRAS-WT) CRC have B-Raf Proto-Oncogene, Serine/Threonine Kinase (BRAF) mutations and do not benefit from epidermal growth factor receptor (EGFR) antibodies. The addition of a porcupine inhibitor could increase the response rate in patients with BRAFV600E-mutated KRAS-WT metastatic CRC (mCRC) with WNT pathway alterations. PATIENTS AND METHODS: We report two cases of severe bone toxicities during treatment with the B

A comparison of an absorbable nasal implant versus functional rhinoplasty for nasal obstruction.

PURPOSE: An absorbable nasal implant for the treatment lateral nasal wall collapse was approved for use in patients with nasal obstruction. It remains to be seen whether this treatment is equivalent to open techniques for the treatment of nasal valve incompetence from collapsibility. MATERIALS AND METHODS: Two groups were analyzed for the study. One group had surgery which included the implant, septoplasty, and inferior turbinate submucous reduction and the other group had a variety of functional rhinoplasty techniques for lateral wall insufficiency in addition to septoplasty and inferior tur

Ultrastructural Characteristics of DHA-Induced Pyroptosis.

Microglial cells are resident macrophages of the central nervous system (CNS) that respond to bioactive lipids such as docosahexaenoic acid (DHA). Low micromolar concentrations of DHA typically promote anti-inflammatory functions of microglia, but higher concentrations result in a form of pro-inflammatory programmed cell death known as pyroptosis. This study used scanning electron microscopy (SEM) and transmission electron microscopy (TEM) to investigate the morphological characteristics of pyroptosis in BV-2 microglial cells following exposure to 200 µM DHA. Vehicle-treated cells are characte

Impairment-driven cancer rehabilitation in patients with neoplastic spinal cord compression using minimally invasive spine stabilization.

BACKGROUND: Neoplastic spinal cord compression is a cause of severe disability in cancer patients. To prevent irreversible paraplegia, a structured strategy is required to address the various impairments present in cancer patients. In this study, we aimed to identify the status where rehabilitation with minimally invasive spine stabilization (MISt) effectively improves ADL. METHODS: We retrospectively reviewed 27 consecutive patients with neoplastic spinal compression who were treated with MISt. We classified the impairments of patients through our multidisciplinary tumor board based on spine

Adult scoliosis can be reduced through specific SEAS exercises: a case report.

BACKGROUND: It has been known since many years that scoliosis can continue to progress after skeletal maturity: the rate of progression has shown to be linear, and it can be used to establish an individual prognosis. Once there is progression there is an indication for treatment: usually it is proposed a surgical one. There are very few papers on an alternative rehabilitation approach; since many years we propose specific SEAS exercises and the aim of this study is to present one case report on this approach. CASE PRESENTATION: All radiographs have been measured blindly twice using the same p

Effects of chronic intermittent asphyxia on rat diaphragm and limb muscle contractility.

OBJECTIVE: In obstructive sleep apnea (OSA), there is intermittent upper airway (UA) collapse due to an imbalance between the collapsing force generated by the diaphragm and the stabilizing force of the UA muscles. This results in chronic intermittent asphyxia (CIA). We have previously shown that CIA affects UA muscle fatigue, but little is known about the effects of chronic hypoxia on diaphragm or on limb muscle contractile properties and structure. DESIGN: Rats were exposed to asphyxia and normoxia twice per minute for 8 h/d for 5 weeks to simulate the intermittent asphyxia of OSA in humans

Equine thoracoscopy: normal anatomy and surgical technique.

Six normal, healthy horses age 3-10 years underwent left and right thoracoscopic examination using a rigid telescope. A minimum of 30 days was allowed between procedures. Horses were restrained in stocks and sedated with a continuous detomidine infusion. After surgical preparation of the hemithorax elected for surgery, and administration of local or regional anaesthesia of the surgery sites, thoracoscopy was completed during two 15 min pneumothorax periods. During the procedures, the thoracic structures were viewed using a 57 cm, 10 mm diameter, 30 degrees rigid telescope connected to a digita

Retinal axon regeneration in the lizard Gallotia galloti in the presence of CNS myelin and oligodendrocytes.

Retinal ganglion cell (RGC) axons in lizards (reptiles) were found to regenerate after optic nerve injury. To determine whether regeneration occurs because the visual pathway has growth-supporting glia cells or whether RGC axons regrow despite the presence of neurite growth-inhibitory components, the substrate properties of lizard optic nerve myelin and of oligodendrocytes were analyzed in vitro, using rat dorsal root ganglion (DRG) neurons. In addition, the response of lizard RGC axons upon contact with rat and reptilian oligodendrocytes or with myelin proteins from the mammalian central nerv

Biomechanical research of medial femoral circumflex vascularized bone-grafting in the treatment of early-to-mid osteonecrosis of the femoral head: a finite element analysis.

PURPOSE: Hip preservation therapy of early ONFH (Osteonecrosis of the femoral head) has emerged as one of the hot areas of research. We have optimized the procedure of traditional MFCVBG (medial femoral circumflex vascularized bone grafting) by using specialized surgical tools and used the finite element analysis to guide the implantation position of the bone flap during surgery and validate the biological mechanical stability of the modified MFCVBG. METHODS: This study was based on the data of a male patient with left hip (ARCO stage IIB, JIC type C) hormonal ONFH. Harris score (HHS), antero

Structures of the procapsid and mature virion of enterovirus 71 strain 1095.

Enterovirus 71 (EV71) is an important emerging human pathogen with a global distribution and presents a disease pattern resembling poliomyelitis with seasonal epidemics that include cases of severe neurological complications, such as acute flaccid paralysis. EV71 is a member of the Picornaviridae family, which consists of icosahedral, nonenveloped, single-stranded RNA viruses. Here we report structures derived from X-ray crystallography and cryoelectron microscopy (cryo-EM) for the 1095 strain of EV71, including a putative precursor in virus assembly, the procapsid, and the mature virus capsid

Sensorimotor coordination and the structure of space.

Embedded in neural and behavioral organization is a structure of sensorimotor space. Both this embedded spatial structure and the structure of physical space inform sensorimotor control. This paper reviews studies in which the gravitational vertical and horizontal are crucial. The mathematical expressions of spatial geometry in these studies indicate methods for investigating sensorimotor control in freefall. In freefall, the spatial structure introduced by gravitation - the distinction between vertical and horizontal - does not exist. However, an astronaut arriving in space carries the physio
