Imaging dopamine transmission parameters in cannabis dependence.

Low striatal dopamine D2/3 receptor (D2/3) availability and low ventrostriatal dopamine release have been observed in alcoholism, cocaine and heroin dependence. Multiple studies to date have examined D2 availability in cannabis dependence and have consistently failed to demonstrate alterations. In addition, the response of the dopamine system to an amphetamine challenge and to a stress challenge has also been examined, and did not show alterations. We review these studies here and conclude that cannabis dependence is an exception among commonly abused drugs in that it is not associated with bl

Estrogen effects on the brain: actions beyond the hypothalamus via novel mechanisms.

From its origins in how the brain controls the endocrine system via the hypothalamus and pituitary gland, neuroendocrinology has evolved into a science that now includes hormone action on many aspects of brain function. These actions involve the whole central nervous system and not just the hypothalamus. Advances in our understanding of cellular and molecular actions of steroid hormones have gone beyond the important cell nuclear actions of steroid hormone receptors to include signaling pathways that intersect with other mediators such as neurotransmitters and neuromodulators. This has, in tur

Neuromodulation in Tourette syndrome: dopamine and beyond.

Almost since the beginning of research on Tourette syndrome (TS), tics have been linked to a dysfunction of the dopamine (DA) system. At first, this assumption was mainly based on clinical findings of DA antagonists being the most effective drug in treating tics, but in recent years nuclear imaging has enabled a much deeper understanding of DA neurotransmission in TS. Based on the findings of various PET and SPECT studies the first part of the review discusses four hypotheses on DA dysfunctions in TS: (i) DA hyperinnervation, (ii) supersensitive DA receptors, (iii) pre-synaptic DA abnormality

The dual role of serotonin in defense and the mode of action of antidepressants on generalized anxiety and panic disorders.

Antidepressants are widely used to treat several anxiety disorders, among which generalized anxiety disorder (GAD) and panic disorder (PD). Serotonin (5-HT) is believed to play a key role in the mode of action of these agents, a major question being which pathways and receptor subtypes are involved in each type of anxiety disorder. The dual role of 5-HT in defense hypothesis assumes that 5-HT facilitates defensive responses to potential threat, like inhibitory avoidance, related to anxiety, whereas it inhibits defensive responses to proximal danger, like one-way escape, related to panic. The f

How social experiences influence the brain.

Social experiences throughout life influence gene expression and behavior, however, early in development these influences have a particularly profound effect. In mammals, mother-infant interactions are the primary source of social stimulation and result in long-term changes in offspring phenotype. This has previously been demonstrated in rodents and primates, however, recent studies in rats have advanced our understanding of how these influences are achieved at a mechanistic level, through epigenetic modification, and provide a model for studying the transmission of social behavior across gene

Treatment of dementia with neurotransmission modulation.

The prevalence of dementia is growing in developed countries where elderly patients are increasing in numbers. Neurotransmission modulation is one approach to the treatment of dementia. Cholinergic precursors, anticholinesterases, nicotine receptor agonists and muscarinic M(2) receptor antagonists are agents that enhance cholinergic neurotransmission and that depend on having some intact cholinergic innervation to be effective in the treatment of dementia. The cholinergic precursor choline alfoscerate may be emerging as a potential useful drug in the treatment of dementia, with few adverse eff

General anaesthetics and their effects on GABA(A) receptor desensitization.

1. Intravenous anaesthetics, including propofol and thiopental have at least three distinct effects on GABA(A) receptor function. 2. Low concentrations of these drugs enhance the amplitude of currents evoked by sub-saturating concentrations of GABA whereas higher concentrations directly activate the receptor in the absence of GABA. 3. Propofol and some barbiturates also decrease the rate and extent of desensitization as indicated by a prolongation in the decay of currents evoked by saturating concentrations of GABA. 4. In contrast, sedative benzodiazepines that lack general anaesthetic propert

Amino acids in neurotransmission and disease.

BACKGROUND: The amino acids, glutamate, GABA and glycine are involved in normal cellular function as well as in neuro-transmission. Glutamate in particular may be linked to neuron development and an oversupply may be the major cause of many acute and chronic neurological diseases. Such diseases include retinopathy of prematurity, glaucoma, hypertension, diabetes and ischemic damage secondary to vessel occlusion. CONCLUSIONS: This paper provides a review of the localization, proposed function and the mechanisms of manufacture and deactivation of the amino acid neurotransmitters. Current mechan

Neurotransmission in epilepsy.

Some evidence indicates that in some types of focal epilepsy the enhanced excitability is due in part to impaired gamma-aminobutyric acid (GABA)ergic inhibitory feedback. One form that this can take is impaired excitatory input to GABAergic interneurons. Enhanced excitatory receptor sensitivity, most characteristically involving N-methyl-D-aspartate (NMDA) receptors, has been identified in kindled rodents and in focal epilepsy in humans. Drugs that enhance GABA-mediated inhibition are anticonvulsant in many syndromes of generalized and focal epilepsy. Mechanisms through which this occurs inclu

Inflammatory sensory polyganglionopathies.

The inflammatory sensory polyganglionopathies are a group of uncommon neurologic disorders primarily affecting dorsal root ganglion cells and their processes. Subdivided into malignant and nonmalignant inflammatory sensory polyganglionopathy, these conditions are important in the differential diagnosis of sensory neuropathy. The clinical significance of malignant inflammatory sensory polyganglionopathy, a paraneoplastic syndrome, rests in the discovery of subclinical cancer, which may produce antineuronal nuclear antibodies directed against the dorsal root ganglion cell and other neurons resul

[Myasthenia gravis and myasthenic syndromes in children].

Myasthenia gravis is the most frequent abnormality of the neuromuscular junction in adults and children. In its milder forms satisfactory management may be obtained with oral neostigmine, but in more severely affected patients steroid therapy and thymus resection may be necessary. Congenital myasthenic syndrome is rather infrequent but timely recognition an identification are very important because treatment and prognosis vary widely in its different forms. Hypermagnesemia, botulism, organophosphate insecticides, are also highlighted among toxic and metabolic causes of neuromuscular junction a

Laser microbeam axotomy and conduction block show that electrical transmission at a central synapse is distributed at multiple contacts.

Touch (T) sensory neurons in the leech innervate defined regions of skin and synapse on other neurons, including other T cells, within the ganglionic neuropil. The cells' receptive fields in the periphery are comprised of a central region, innervated by thick axons, and adjoining regions (minor fields) innervated by thinner axons. Secondary branches, known to be sites of synapses, emerge from the thinner and thicker axons. Pairs of T cells appear to make up to 200 separate contacts distributed within the neuropil. When the T cell is hyperpolarized, as occurs during natural stimulation of the c

Neuropharmacology of tics.

In pharmacological treatment of Gilles de la Tourette's syndrome neuroleptic (antipsychotic) drugs have been the most effective. This has led to the hypothesis of a relative dopaminergic overactivity in the brain. Animal studies with neuroleptic drugs different in their affinity to different dopamine areas as well as studies with drugs different in their selectivity to different receptors have, however, not shown direct correlations to the clinical response in the treatment of hyperkinetic syndromes. There is a trend that drugs with high affinity to striatal D2-receptors might be the most pote

Synaptic and intrinsic plasticity in the ventral tegmental area after chronic cocaine.

Cocaine exposure induces persistent changes in synaptic transmission and intrinsic properties of ventral tegmental area (VTA) dopamine neurons. Despite significant progress in understanding cocaine-induced plasticity, an effective treatment of cocaine addiction is lacking. Chronic cocaine potentiates excitatory and alters inhibitory transmission to dopamine neurons, induces dopamine neuron hyperexcitability, and reduces dopamine release in projection areas. Understanding how intrinsic and synaptic plasticity interact to control dopamine neuron firing and dopamine release could prove useful in …
