Bioactive compound from the Tibetan turnip (Brassica rapa L.) elicited anti-hypoxia effects in OGD/R-injured HT22 cells by activating the PI3K/AKT pathway.

Cerebral stroke, a common clinical problem, is the predominant cause of disability and death worldwide. Its prevalence increases and infarctions exacerbate with age. A Tibetan plant, Brassica rapa L., possesses multiple medicinal effects, such as anti-altitude sickness, anti-hyperlipidemia and anti-fatigue, as mentioned in the noted ancient Tibet pharmacopeia "The Four Medical Tantras". Our preliminary studies also showed the anti-hypoxia protection mechanism of B. rapa L., implying its possible relationship with anti-ischemic neuroprotection. However, the potential molecular mechanism of the …

Role of AURKA in the hypothalamus-pituitary-testicular axis in Tibetan sheep from Tianzhu.

The mitosis-associated protein aurora kinase A (AURKA) regulates the maturation of germ cells. We have previously reported using transcriptome analysis that AURKA is expressed in yak testes. Although Tibetan sheep possess an immense economic value, their reproductive rate is low. Herein, the expression and functions of AURKA in the hypothalamus-pituitary-testicular (HPT) axis in Tibetan sheep from Tianzhu were investigated. The cDNA sequence of sheep AURKA was cloned and bioinformatics techniques were used to predict its structure. Tissue expression of AURKA was determined by qPCR, immunoblott…

The microbe-mediated mechanisms affecting topsoil carbon stock in Tibetan grasslands.

Warming has been shown to cause soil carbon (C) loss in northern grasslands owing to accelerated microbial decomposition that offsets increased grass productivity. Yet, a multi-decadal survey indicated that the surface soil C stock in Tibetan alpine grasslands remained relatively stable. To investigate this inconsistency, we analyzed the feedback responses of soil microbial communities to simulated warming by soil transplant in Tibetan grasslands. Whereas microbial functional diversity decreased in response to warming, microbial community structure did not correlate with changes in temperature…

Poisonous plants weaken plant-microbe-fauna trait networks across temperature and precipitation gradients.

Trait-based ecology provides a mechanistic framework for predicting how species and trophic interactions respond to environmental change, yet cross-trophic trait linkages remain poorly understood. We investigated how the expansion of poisonous plants reshapes functional traits of plants, soil microbes, and soil fauna along climatic gradients on the Qinghai-Tibet Plateau, and how these shifts affect multitrophic trait networks and ecosystem complexity. To address this, we quantified 36 functional traits covering plants, bacteria, fungi, nematodes, and mites from 150 plots at 15 sites spanning a…

Indigenous Sporosarcina pasteurii-driven carbonate mineralization couples lead immobilization with erosion control in contaminated slopes.

Lead (Pb) contamination in erosion-prone soils presents a dual environmental issue because it causes concurrent pollution and soil degradation. We developed a sustainable bioremediation technique utilizing microbially induced calcium carbonate precipitation (MICP) with the indigenous Pb-tolerant ureolytic bacterium Sporosarcina pasteurii, isolated from the biological soil crusts of the Qinghai-Tibet Plateau. The strain exhibited considerable urease activity and consistent growth under extreme lead stress (2000 mg/L) and effectively facilitated carbonate mineralization. The MICP treatment resul…

Phylogenomic conflict and reticulate evolution during radiative diversification of the genus Rheum.

BACKGROUND AND AIMS: The evolutionary mechanisms that underpin the remarkable morphological diversity observed in rapidly radiating plant lineages within mountainous ecosystems continue to captivate the attention of evolutionary biology. The genus Rheum (rhubarb), is an alpine genus renowned for its striking variation in growth forms and specialized bracts in the Qinghai-Tibet Plateau and adjacent regions, thus presenting an exceptional system to address this inquiry. METHODS: We employed a phylogenomic approach, integrating nuclear gene and plastid gene information derived from transcriptome…

Plateau Environment, Gut Microbiota, and Depression: A Possible Concealed Connection?

Plateau environments present unique mental health challenges owing to stressors including hypoxia, low temperatures, and intense ultraviolet (UV) radiation. These factors induce structural and functional alterations in the gut microbiota, disrupting gut-brain axis homeostasis and contributing to the higher prevalence of depression in plateau regions relative to flatland areas. For example, studies report that 28.6% of Tibetan adults and 29.2% of children/adolescents on the Qinghai-Tibet Plateau experience depression, with increasing evidence linking this trend to alterations in the gut microbi…

Whole-genome resequencing to investigate the genetic diversity and the molecular basis underlying key economic traits in indigenous sheep breeds adapted to hypoxic environments.

Under the combined effects of long-term natural selection and artificial domestication, Tibetan sheep on the Qinghai-Tibet Plateau have evolved distinct ecotypes to survive extreme high-altitude conditions, including hypoxia, cold, and low oxygen levels. These ecotypic variations not only serve as an ideal model for studying plateau livestock adaptation but also harbor valuable genetic diversity. However, the lack of comprehensive genetic analyses on their adaptive and phenotypic traits has hindered the effective conservation and utilization of these resources. Using whole-genome resequencing,…

Structure and activity alteration in adult highland residents' cerebrum: Voxel-based morphometry and amplitude of low-frequency fluctuation study.

INTRODUCTION: People living in highland areas may have factors that allow them to adapt to chronic hypoxia, but these physiological mechanisms remain unclear. This study aimed to investigate the brain mechanism in a cohort of adult residents of Tibet, a well-known plateau section in China, by observing differences in brain structure and function in non-plateau populations. METHODS: The study included 27 Tibetan and 27 non-plateau region residents who were matched in age, sex, and education. All participants underwent high-resolution three-dimensional T1 weighted imaging (3D-T1WI) and resting-…
