To blink or not to blink, that is the question: Reassessing rabbit eyeblink conditioning for learning and memory evaluation in nonclinical safety studies.

Eyeblink conditioning (EBC) has been explored as a cognitive endpoint in rabbit pre- and postnatal development (PPND) and juvenile toxicity (JAS) studies; however, species-specific behavioral constraints, procedural demands, and the basic nature of the task may limit its ability to address complex neurobehavioral objectives in regulatory contexts. While EBC provides a measure of associative learning, it may lack the sensitivity and relevance required for regulatory toxicology studies designed to evaluate human health risk through complex neurobehavioral assessment paradigms. In addition, given

A tool to assess the internal validity of toxicokinetic studies.

Toxicological animal studies are essential to risk assessment. The reliability of the studies depends on the extent to which study design, conduct, and analysis have a high internal validity. Several tools exist for assessing the internal validity of human and animal studies, however, a dedicated tool for animal toxicokinetic studies is lacking. We developed a questionnaire to systematically assess toxicokinetic animal studies. The questionnaire consists of seven main domains with questions for which points are given by the assessor multiplied by fixed weighting factors (WF). The scores are th

Organobodies: a robust and size-controllable system for generating scalable hiPSC-derived liver organoids for drug toxicity screening.

Human hepatic organoids derived from pluripotent or adult stem cells offer powerful platforms for disease modeling and drug discovery. However, developing robust and scalable organoids capable of sustaining long-term functionality remains challenging. Here, we developed a novel, semi-defined approach using a self-assembling peptide and collagen I to create highly uniform human induced pluripotent stem cell-derived hepatic organoids in droplet format, which we term hepatic organobodies (OBs). This method enabled rapid, reproducible production of threedimensional (3D) liver tissues, which remain

Redefining ocular safety assessment: retinal organoids as platforms for predicting human ocular toxicology.

Despite the substantial progress in drug discovery and precision therapeutics, the predictive power of current ocular safety assessments remains limited owing to the lack of human experimental models. Conventional two-dimensional cell cultures lack the complex laminar organization, multicellular interactions, and functional electrophysiological properties of the human retina. Additionally, animal models frequently exhibit species-specific differences in retinal development, metabolism, and stress responses that hinder translational accuracy. Human induced pluripotent stem cell-derived retinal

In vitro models of the male reproductive system: applications for developmental and reproductive toxicology.

There is a paucity of in vitro models to study the male reproductive system. Proper function of the reproductive system is critical for endocrine function, growth and development, and fertility. Without practical in vitro screening models, reproductive toxicities can be missed in early drug development or standard toxicological batteries. Successful in vitro models of the male reproductive system need to recapitulate the dynamic nature of the testis, considering the formation of the testicular niches from gonadal differentiation through puberty and the post-pubertal activity of the paracrine a

Developmental Toxicity Assay Based on Real-Time Monitoring of Fibroblast Growth Factor Signal Disruption in Human Induced Pluripotent Stem Cells.

Developmental toxicity (Devtox) is typically evaluated in animal models. However, there are challenges in ensuring accuracy, such as false negatives due to interspecies differences and ethical concerns regarding animal welfare. Therefore, there is a need for new test systems to evaluate human Devtox and help address these issues. We hypothesized that teratogens disrupt Devtox-related signaling pathways essential for morphogenesis, irrespective of their molecular targets. To test this hypothesis, we developed a fibroblast growth factor (FGF) reporter system using human induced pluripotent stem

Rupture of Brumadinho dam (Minas Gerais, Brazil): embryotoxicity in zebrafish induced by metal mixture-contaminated water.

Rupture of dam B-1 at the Córrego do Feijão mine in Brumadinho, state of Minas Gerais, Brazil (25/Jan/2019) contaminated the Paraopeba River with mine waste. To identify the adverse effects of this event on public and environmental health, we conducted the Fish Embryo Acute Toxicity (FET) test (OECD No. 236). During the tests, zebrafish embryos were exposed for 144 hours to solutions containing realistic concentrations of dissolved iron and aluminum and total manganese at the following analysis points upstream or downstream of the dam: 10 km, upstream; 19.7 km, downstream, at the point where w

Acute and sub-acute toxicity study of ethanol extract from Nectandra leucantha Nees & Mart. (Lauraceae) barks.

Nectandra leucantha has been used in traditional medicine. Several metabolites isolated from N. leucantha extracts displayed immunomodulatory, antileishmanial properties, but the determination of the toxicological profile in mammals has not previously been performed. In this study, the ethanol extract from N. leucantha barks (EENl) was characterized by HPLC/HRESIMS. To study acute toxicity, female mice received EENl in a single dose of 100, 300, 1000, or 2000 mg/kg bw. Later, sub-acute toxicity was introduced in female and male mice by oral gavage at 100, 500 or 1000 mg/kg bw for 28 consecutiv

Investigating the Joint Effects of Pesticides and Ultraviolet B Radiation in Xenopus laevis and Other Amphibians.

Exposure to multiple stressors often results in higher toxicity than one stressor alone. Examining joint effects of multiple stressors could provide more realistic exposure scenarios and a better understanding of the combined effects. In amphibian toxicology, simultaneous exposure to some pesticides and ultraviolet B (UVB) radiation has been suggested to be detrimental and more harmful in amphibian early-life stages than either stressor alone. Therefore, it is important to investigate the joint effects of these two stressors and provide data that could lead to more informed risk assessment. He

Tetrodotoxins Secretion and Voltage-Gated Sodium Channel Adaptation in the Ribbon Worm Kulikovia alborostrata (Takakura, 1898) (Nemertea).

Nemertea is a phylum of marine worms whose members bear various toxins, including tetrodotoxin (TTX) and its analogues. Despite the more than 30 years of studying TTXs in nemerteans, many questions regarding their functions and the mechanisms ensuring their accumulation and usage remain unclear. In the nemertean Kulikovia alborostrata, we studied TTX and 5,6,11-trideoxyTTX concentrations in body extracts and in released mucus, as well as various aspects of the TTX-positive-cell excretion system and voltage-gated sodium (Nav1) channel subtype 1 mutations contributing to the toxins' accumulation

AOP and IATA applied to ocular surface toxicity.

Until now, the Draize test on rabbits has been the only test performed to anticipate ocular toxicity of pharmaceutical compounds, mainly irritation. The OECD is urging the scientific community to develop and validate alternative methods to reduce the need for animal testing. Since the models and tests used cannot reflect the entire biologic response, it is necessary to combine them into integrated approaches to testing and assessment (IATA) to obtain robust data. IATAs, along with adverse outcome pathways (AOP) that encompass molecular cascades and key events, require the best combinations of

Invited review: human air-liquid-interface organotypic airway tissue models derived from primary tracheobronchial epithelial cells-overview and perspectives.

The lung is an organ that is directly exposed to the external environment. Given the large surface area and extensive ventilation of the lung, it is prone to exposure to airborne substances, such as pathogens, allergens, chemicals, and particulate matter. Highly elaborate and effective mechanisms have evolved to protect and maintain homeostasis in the lung. Despite these sophisticated defense mechanisms, the respiratory system remains highly susceptible to environmental challenges. Because of the impact of respiratory exposure on human health and disease, there has been considerable interest i

Antineoplastic Agents: Environmental Prevalence and Adverse Outcomes in Aquatic Organisms.

Cancer is the second leading cause of death worldwide, with 9.6 million cancer-related deaths in 2018. Cancer incidence has increased over time, and so has the prescription rate of chemotherapeutic drugs. These pharmaceuticals, known as antineoplastic agents, enter the aquatic environment via human excretion and wastewater. The objectives of the present critical review were to investigate the risk of antineoplastics to aquatic species and to summarize the current state of knowledge regarding their levels in the environment, because many antineoplastics are not adequately removed during wastewa

Screening for Enhancement of Permethrin Toxicity by Plant Essential Oils Against Adult Females of the Yellow Fever Mosquito (Diptera: Culicidae).

Aedes aegypti L. (Diptera: Culicidae) is one of the most medically important mosquito species, due to its ability to spread viruses of yellow fever, dengue fever, and Zika in humans. In this study, the insecticidal activity of 17 plant essential oils was evaluated via topical application against two strains of Ae. aegypti mosquito, Orlando (insecticide-susceptible) and Puerto Rico (pyrethroid-resistant). Initial screens with the Orlando strain showed that cucumber seed oil (2017 sample) was the most toxic, followed by sandalwood and thyme oil. When the essential oils were mixed with permethrin

Heterogeneous photocatalytic degradation of diuron on zinc oxide: Influence of surface-dependent adsorption on kinetics, degradation pathway, and toxicity of intermediates.

Heterogeneous photocatalytic reaction has been generally applied for degradation of toxic contaminants. Degradations of a compound using the same kind of catalyst that was synthesized differently are commonly found in literature. However, the reported degradation intermediates are normally inconsistent. This issue is especially important for the degradation of toxic compounds because intermediates may be more toxic than their parent compounds and understanding the reason is necessary if appropriate catalysts are to be designed. This work systematically compares the photocatalytic degradation o
