Inhibition of synaptophysin ubiquitination may improve the intelligent drop due to high glucose and hypoxia.

Synaptophysin (SYP) is a synaptic vesicle membrane protein and involved in the release of neurotransmitters, synaptic plasticity, and formation and recycling of synaptic vesicles. SYP may serve as a specific marker of synaptic proteins. Our results showed high glucose and hypoxia could significantly impair the intelligence and the SYP level was positively related to the degree of intelligence in animals. Ubiquitination may influence the SYP expression. In our study, an ubiquitination inhibitor (MG-132) was injected intraperitoneally. Results showed this could increase SYP expression in the bra…

Exploring the spectrum of 3-M syndrome, a primordial short stature disorder of disrupted ubiquitination.

3-M syndrome is an autosomal recessive primordial growth disorder characterized by small birth size and post-natal growth restriction associated with a spectrum of minor anomalies (including a triangular-shaped face, flat cheeks, full lips, short chest and prominent fleshy heels). Unlike many other primordial short stature syndromes, intelligence is normal and there is no other major system involvement, indicating that 3-M is predominantly a growth-related condition. From an endocrine perspective, serum GH levels are usually normal and IGF-I normal or low, while growth response to rhGH therapy

Smad ubiquitination regulatory factor 2 expression is enhanced in hypertrophic scar fibroblasts from burned children.

Transforming growth factor-β1 (TGF-β1) plays a key role in hypertrophic scar formation. A lot of studies have shown that TGF-β1 stimulates fibroblast proliferation, collagen production, and α-smooth muscle actin (α-SMA) expression, inhibits matrix degradation and eventually leads to scar formation. Smad proteins are important intracellular mediators of TGF-β1 signaling, and Smad ubiquitination regulatory factor 2 (Smurf2), an ubiquitin ligase for Smads, plays critical roles in the regulation of TGF-β1/Smad signaling. It was reported that Smurf2 was abnormally expressed during the process of li…

Helicobacter pylori CagA activates NF-kappaB by targeting TAK1 for TRAF6-mediated Lys 63 ubiquitination.

Helicobacter pylori-initiated chronic gastritis is characterized by the cag pathogenicity island-dependent upregulation of proinflammatory c … activation through the tumour necrosis factor receptor-associated factor 6-mediated, Lys 63-linked ubiquitination of TAK1. These findings show that polyubiquitination of TAK1 regulates the activation of NF-kappaB, which in turn is used by H. pylori CagA for the H. pylori-induced inflammatory response.

Role of CYP2E1 activity in endoplasmic reticulum ubiquitination, proteasome association, and the unfolded protein response.

In an experimental model of liver cirrhosis, marked increases in ER proteasome content in rat livers were observed 5 h after acute i.p. injection of the hepatotoxicant CCl4. To confirm the role of CYP2E1 in mediating protein misfolding/damage in the ER via its metabolism of CCl4, 293T cells stably transfected with human CYP2E1 were exposed to CCl4 and cell ER fractions assessed for ubiquitination. Increases in ER ubiquitin conjugates were noted in CYP2E1/293T cells treated with CCl4 and not in controls, suggesting these effects are CYP2E1 specific. Finally, the role of CYP2E1 in ER homeostasis…

Loss of UBE3C-mediated PIM2 ubiquitination drives Helicobacter pylori-induced gastritis via JAK2/STAT3 activation.

Helicobacter pylori (H. pylori) infection is a major cause of chronic gastritis, yet its molecular mechanisms remain incompletely understood … ylori infection downregulated the expression of the E3 ubiquitin ligase UBE3C, leading to decreased ubiquitination of PIM2 at K165 and enhanced PIM2 protein stability. Accumulated PIM2 subsequently activated the JAK2/STAT3 pathway, driving inflammatory responses. Combined treatment with the JAK1/2 inhibitor Ruxolitinib further potentiated the anti-inflammatory e …

CNOT4 Suppresses Bladder Cancer Progression by Promoting Ubiquitination-Mediated Degradation of EIF2AK2 and Inhibiting Autophagy.

EIF2AK2, also known as PKR, is a stress-responsive kinase that regulates multiple cellular processes, including autophagy, apoptosis, and tu … hways. Mechanistic analyses revealed that CNOT4 interacted with EIF2AK2 and promoted its K48-linked ubiquitination and proteasomal degradation. Restoration of EIF2AK2 expression largely reversed the tumor-suppressive effects of CNOT4, indicating that EIF2AK2 acts as a key downstream effector. Consistent with these findings, CNOT4 overexpression significantly sup …

Colorectal cancer is a leading cause of cancer-related death worldwide. The role of immediate early response 5-like (IER5L) in colorectal ca … lly, IER5L bound directly to the E3 ubiquitin ligase TRAF7. This interaction facilitated K48-linked ubiquitination and proteasomal degradation of the tumor suppressor p53. Restoring p53 expression reversed IER5L-driven malignant phenotypes. TRAF7 knockdown or mutation of its lysine 451 residue, a critical IER5L binding site, abrogated IER5L effects. The IER5L-TR …

USP1 represses ferroptosis in endometrial carcinoma cells by regulating the ubiquitination and stability of ESR1.

Endometrial carcinoma (EC) is a prevalent gynecologic malignancy. Ferroptosis, a type of programmed cell death, holds promise as a therapeut … ng protein involved in ferroptosis regulation. USP1 was shown to interact with ESR1, reduce its polyubiquitination, and enhance its protein stability. Rescue experiments revealed that ESR1 overexpression counteracted the pro-ferroptotic effects of USP1 depletion in erastin-treated EC cells. Finally, in vivo studies using an EC xenograft model confirmed the anti- …

Polyglutamine-Expanded Ataxin-3 Accelerates CFTR Degradation Through K63-Linked Ubiquitination to Exacerbate Microglial Inflammation.

The deubiquitinase Ataxin-3 causes spinocerebellar ataxia type 3 (SCA3) upon polyglutamine (polyQ) expansion. While expressed in the nervous system, the function of the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel therein remains unclear, as does its potential regulation by Ataxin-3. This study reveals that Ataxin-3 interacts with and promotes CFTR degradation in human microglia by its K63-linked polyubiquitination, thereby shortening CFTR's half-life. Paradoxically, K63-linked polyubiquitin chains also promote the degradation of Ataxin-3 itself, suggesting a com…

MCCC2 stabilizes LTBP1 via suppressing SMURF1-dependent ubiquitination to drive bone metastasis in prostate cancer.

Prostate cancer (PCa) is one of the most common malignancies in men globally, with bone metastasis being a leading cause of death in advance … n vitro, and bone metastasis in vivo. Mechanistically, MCCC2 competitively inhibits SMURF1-mediated ubiquitination and degradation of LTBP1, stabilizing LTBP1 to activate TGF-β signaling. Clinically, high MCCC2 expression correlates with elevated LTBP1 levels, increased bone metastasis incidence, and poor prognosis in PCa patients. These findings reveal that MCC …

Serine ubiquitination of the SARS-CoV-2 RdRp by the non-canonical E3 ligase MYCBP2.

Post-translational modifications, including ubiquitination, have emerged as important regulators of viral infection and the host innate immune response. We screened the proteins of SARS-CoV-2 for ubiquitination and identified the RNA-dependent RNA polymerase (RdRp) NSP12 as being ubiquitinated, which has not previously been reported. Importantly, we confirmed that NSP12 is ubiquitinated during SARS-CoV-2 infection, and the NSP12 proteins of different coronaviruses are differentially associated with ubiquitin chains. SARS-CoV-2 NSP12 was primarily associated with K63-linked polyubiquitin chains…

SAE1 knockdown attenuates hepatic fibrosis by promoting ubiquitination and proteasomal degradation of TGF-β receptors.

OBJECTIVE: Liver fibrosis is characterised by excessive accumulation of extracellular matrix and can ultimately progress to cirrhosis and malignant transformation. Activation of hepatic stellate cells (HSCs), largely driven by transforming growth factor-beta (TGF-β) signalling, is a central event in fibrogenesis. SAE1 is a subunit of the small ubiquitin-like modifier-activating enzyme E1; its role in liver fibrosis and HSC activation remains unclear. This study aimed to investigate the regulatory function and underlying mechanism of SAE1 in hepatic fibrogenesis. METHODS: A carbon tetrachlorid
