Comparative analysis of advanced therapy regulatory frameworks in Latin America: perspectives from the ISCT South and Central America Legal and Regulatory Affairs Committee.

Advanced therapies, including gene therapies, somatic cell therapies, and tissue-engineered products, pose distinctive regulatory challenges because of their scientific complexity, manufacturing variability, clinical uncertainty, and long-term safety considerations. Recent clinical advances have heightened the need for regulatory pathways capable of supporting safe, effective, and sustainable access. Across Latin America, regulatory approaches for these products have evolved unevenly, reflecting differences in legislative development, institutional capacity, and regulatory specialization among

When multimodal fusion fails: contrastive alignment as a necessary stabilizer for TCR-peptide binding prediction.

MOTIVATION: Multimodal learning is often assumed to improve predictive performance by combining complementary views, yet in biological applications auxiliary modalities are frequently imperfect, incomplete, or derived from upstream predictors and heuristics. We study this issue in TCR-peptide binding prediction, where sequence embeddings from pretrained protein language models are strong and transferable, but structure-derived residue graphs must be built from predicted folds and discretized contacts. These structural views can therefore be noisy, inconsistent across proteins, and sensitive to

Antitumour effects of single or combined monoclonal antibodies directed against membrane antigens expressed by human B cells leukaemia.

BACKGROUND: The increasing availability of different monoclonal antibodies (mAbs) opens the way to more specific biologic therapy of cancer patients. However, despite the significant success of therapy in breast and ovarian carcinomas with anti-HER2 mAbs as well as in non-Hodkin B cell lymphomas with anti-CD20 mAbs, certain B cell malignancies such as B chronic lymphocytic leukaemia (B-CLL) respond poorly to anti-CD20 mAb, due to the low surface expression of this molecule. Thus, new mAbs adapted to each types of tumour will help to develop personalised mAb treatment. To this aim, we analyse t

Biologics in pediatric chronic rhinosinusitis with nasal polyps: A systematic review of efficacy and safety.

Chronic rhinosinusitis with nasal polyps (CRSwNP) is uncommon in pediatric populations but may significantly impair quality of life. Adult data support biologics targeting type 2 inflammatory pathways, whereas pediatric evidence remains sparse and largely indirect. This systematic review aimed to summarize the available evidence on both efficacy and safety, rather than to estimate a precise treatment effect. A systematic review was conducted in accordance with the PRISMA 2020 guidelines. PubMed, Scopus, and Cochrane Library databases were searched from inception to March 2026 for studies inves

Living donor liver transplantation in transplant oncology: A disease-specific framework for access, timing, and donor risk.

Living donor liver transplantation (LDLT) should not only be evaluated as an alternative graft source in transplant oncology; it should also be evaluated as a disease-specific intervention on access and oncologic time. By removing dependence on deceased-donor availability, LDLT can convert an uncertain wait into a planned operation-but that temporal advantage is not biologically neutral, and its consequences differ sharply by tumor type. In hepatocellular carcinoma, an abbreviated wait can reduce waitlist dropout but may also shorten the interval during which aggressive biology would otherwise

Comparative Efficacy of Platelet-Rich Fibrin and Enamel Matrix Derivative As Adjuncts to Non-Surgical Periodontal Therapy: A Split-Mouth Randomised Clinical Trial.

BACKGROUND: Periodontitis is a chronic multifactorial inflammatory condition resulting in the gradual breakdown of periodontal supporting structures. Although non-surgical periodontal therapy (NSPT) is the gold standard first therapeutic option, its effectiveness can be limited in moderate-to-severe cases, hence necessitating the application of adjunctive therapies. Platelet-rich fibrin (PRF) and enamel matrix derivative (EMD) are among the most extensively utilised regenerative biomaterials; nevertheless, direct comparative evidence related to their adjunctive advantage in NSPT is scarce. Molecular Pathogenesis, Tumor Microenvironment and Health Disparities in Select Pediatric Solid Tumors: An Integrative Narrative Review.

BACKGROUND/OBJECTIVES: Pediatric solid tumors (PST) are a biologically distinct group of malignancies whose developmental origins and molecular drivers differ substantially from those of adult cancers, with direct implications for therapeutic strategy and clinical outcome. This review synthesizes current evidence on molecular pathogenesis, tumor microenvironment biology, and the structural conditions that shape access to care across select PST. METHODS: A narrative review of peer-reviewed literature was conducted primarily using PubMed, supplemented by Google Scholar, covering publications fr

Temporal Trends in Surgery and Hospitalization Rates for Crohn's Disease in Brazil: A Population-Based Study.

INTRODUCTION: Biological therapy has transformed the natural course of inflammatory bowel disease, but there are still controversies regarding its potential to reduce surgical rates for Crohn's disease (CD). This study, conducted with the support of the Brazilian National Healthcare System, aimed to analyze temporal trends in surgery and hospitalization rates among patients with CD and to correlate these data with the dispensing of azathioprine (AZA), infliximab (IFX), and adalimumab (ADA). METHODOLOGY: This retrospective observational study used data from the National Public Healthcare Depar

Ecological drivers of movement for two sympatric marine predators in the California current large marine ecosystem.

BACKGROUND: An animal's movement reflects behavioral decisions made to address ecological needs; specifically, that movement will become less directional in regions with high prey availability, indicating foraging behavior. In the marine realm, animal behavior occurs below the sea surface and is difficult to observe. We used an extensive satellite tagging dataset to explore how physical and biological habitat characteristics influence blue (Balaenoptera musculus) and fin (B. physalus) whale movement and foraging behavior in the California Current Ecosystem across four known bioregions. Amyloid-related imaging abnormalities (ARIA): radiological, biological and clinical characteristics.

Excess accumulation and aggregation of toxic soluble and insoluble amyloid-β species in the brain are a major hallmark of Alzheimer's disease. Randomized clinical trials show reduced brain amyloid-β deposits using monoclonal antibodies that target amyloid-β and have identified MRI signal abnormalities called amyloid-related imaging abnormalities (ARIA) as possible spontaneous or treatment-related adverse events. This review provides a comprehensive state-of-the-art conceptual review of radiological features, clinical detection and classification challenges, pathophysiology, underlying biologic

Appetite control and energy balance regulation in the modern world: reward-driven brain overrides repletion signals.

Powerful biological mechanisms evolved to defend adequate nutrient supply and optimal levels of body weight/adiposity. Low levels of leptin indicating food deprivation and depleted fat stores have been identified as the strongest signals to induce adaptive biological actions such as increased energy intake and reduced energy expenditure. In concert with other signals from the gut and metabolically active tissues, low leptin levels trigger powerful activation of multiple peripheral and brain systems to restore energy balance. It is not just neurons in the arcuate nucleus, but many other brain s

Surveillance of acute flaccid paralysis in the Marches region (Italy): 1997-2007.

BACKGROUND: The last case of poliomyelitis due to transmission of indigenous wild poliovirus occurred in Italy in 1982, however, it is important to guarantee a high quality surveillance as there is a risk of importation of cases from areas where polio is endemic. Stopping poliovirus transmission is pursued through a combination of high infant immunization coverage and surveillance for wild poliovirus through reporting and laboratory testing of all cases of acute flaccid paralysis (AFP) among children under fifteen years of age. The aim of this study was to describe and to evaluate 11 years of

New cytokine-dependent acute myeloid leukemia cell line MUTZ-11 with disomic chromosome rearrangement t(16;17).

Continuous human leukemia-lymphoma (LL) cell lines represent a rich resource of abundant, accessible and manipulable living cells contributing significantly to a better understanding of the pathophysiology of hematopoietic tumors. In particular, classical and molecular cytogenetics have benefitted enormously from the availability of LL cell lines with specific chromosomal abnormalities. Such aberrations may be the portal to the discovery of novel oncogene rearrangements for which positive cell lines provide a resource for both discovery and functional studies. The new continuous leukemia cell

A new method of antitumor therapy with a high dose of TNF perfusion for unresectable liver tumors.

There are primary and secondary malignant liver tumors for which principal treatment is surgical resection. There is no established treatment for unresectable malignant liver tumors, however, and the prognosis for these is quite poor. An effective treatment for malignant liver tumors is thus urgently needed. Recent advances in molecular biology have uncovered the structures and/or functions of many cytokines thought to have a strong relation with the mechanisms of the antitumor effect of biological therapies. Availability of those cytokines in large amounts and homogeneously owing to advances

Discovery of an orally bioavailable pyridone-based BRD4 inhibitor with potent antitumor efficacy in colorectal cancer.

Colorectal cancer (CRC) remains a major clinical burden with limited durable responses. Given reports implicating Bromodomain-containing protein 4 (BRD4) in CRC proliferation and therapy resistance, more potent BRD4-targeting agents with proven in-vivo activity are needed. Through virtual screening followed by structure-guided chemical modification, we identified H5 as a BRD4 inhibitor. In biochemical assays, H5 shows IC50 = 7.9 ± 0.5 nM, outperforming JQ-1 (IC50 = 33.0 ± 1.0 nM) by 4-fold. Molecular dynamics simulations indicate that H5 engages BRD4 via hydrogen bonds with Asn140 and Asp88 an

ImesAgCl/TBHP Promoted Aqueous Synthesis of Isatin-Amide Derivatives: Anti-proliferative, Anti-Vascular Endothelial Growth Factor Receptor 2, Bioavailability and Molecular Docking Studies.

Cancer is a worldwide health burden that influences about every geographic location and socioeconomic class. One of the main challenges that remains is the appearance of resistance to cancer therapy. It is found that vascular endothelial growth factor receptor 2 (VEGFR-2) is overexpressed in a number of tumours. Few FDA approved drugs (such as Sorafenib, Sunitinib, Nintedanib and Cabotanzinib), which target VEGFRs, however, a significant issue with the monotherapy of these agents is drug resistance. Hence, in the present work. Some isatin-aromatic amides (5a-q) were synthesised via Ag(I)-NHC/T

IGF-I bioavailability in congenital isolated growth hormone deficiency.

BACKGROUND: The Itabaianinha cohort in Brazil carries a homozygous growth hormone-releasing hormone (GHRH) receptor (GHRH-R) gene mutation, causing congenital isolated GH deficiency (GHD). Affected individuals present with severe short stature, central obesity, hypercholesterolemia, and marked reductions in serum GH, IGF-I, and IGFBP 3 concentrations yet show no premature atherosclerosis and maintain a normal lifespan. IGF-I mostly circulates bound to IGFBPs and requires proteolytic cleavage for IGF-I receptor activation. Pregnancy-associated plasma protein A (PAPP-A) is an important IGF-depen

DoE-Optimized Chitosan-Coated pH-Sensitive Liposomes for Targeted Nose-to-Brain Delivery of Temozolomide.

Glioblastoma is an aggressive brain tumor with limited treatment efficacy due to poor blood-brain barrier (BBB) penetration and intrinsic resistance. Temozolomide (TMZ), the frontline chemotherapeutic, undergoes rapid hydrolysis at physiological pH, with low systemic bioavailability and efflux-mediated clearance. To address this, we developed chitosan-coated pH-sensitive liposomes (SpH-TMZ-CS-LIPO) that were optimized via Box-Behnken design, achieving 223.48 ± 1.93 particle size, 35.72 ± 0.51 zeta potential, and 78.85 ± 2.62% entrapment. The formulation exhibited pH-sensitive release, with cum
