Epidermal growth factor and transforming growth factor-alpha: role in protection and healing of gastric mucosal lesions.

The maintenance of the integrity of the gastrointestinal mucosa and the repair of acute and chronic mucosal lesions are under the influence of various growth factors, especially epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-alpha). EGF originates mainly from salivary glands, whereas TGF-alpha is released locally in the gastric mucosa, particularly when the mucosa is exposed to topical irritants. EGF and TGF-alpha have similar spectra of biological activity, which include the stimulation of the restitution and proliferation of mucosal cells, gastroprotection, vasodilat

The PRKN/METTL3/CLDN2 axis promotes colorectal cancer development through epigenetic mechanisms.

BACKGROUND: Dysregulation of N6-methyladenosine (m6A) RNA methylation plays a crucial role in colorectal cancer (CRC) development. Despite the established oncogenic role of methyltransferase-like 3 (METTL3) in CRC, the mechanisms behind its tumor-promoting functions are not fully understood. METHODS: Functional impacts were assessed by measuring cell viability, invasion, colony formation, migration, apoptosis, and M2 macrophage polarization in vitro, as well as in vivo tumorigenicity. The interaction between METTL3 and claudin 2 (CLDN2) was validated via RNA immunoprecipitation and luciferase

α7nAChR inhibits pyroptosis of rheumatoid arthritis synovial fibroblasts via the NLRP3/GSDMD pathway.

The pyroptosis of synovial fibroblasts (SFs) plays a pivotal role in the pathogenesis of rheumatoid arthritis (RA) and serves as a key driver of joint inflammation. Prior studies have elucidated the biological role of the α7 nicotinic acetylcholine receptor (α7nAChR) in RA; however, its influence on pyroptosis in RA-SFs remains to be fully understood. In this study, the human synovial fibroblast cell line MH7A was stimulated with TNF-α to mimic the characteristics of RA-SFs. The extent of pyroptosis was assessed by measuring the levels of lactate dehydrogenase (LDH) and pro-inflammatory cytoki

Recessive Mutations in SYNPO2 as a Candidate of Monogenic Nephrotic Syndrome.

INTRODUCTION: Most of the approximately 60 genes that if mutated cause steroid-resistant nephrotic syndrome (SRNS) are highly expressed in the glomerular podocyte, rendering SRNS a "podocytopathy." METHODS: We performed whole-exome sequencing (WES) in 1200 nephrotic syndrome (NS) patients. RESULTS: We discovered homozygous truncating and homozygous missense mutation in SYNPO2 (synaptopodin-2) (p.Lys1124∗ and p.Ala1134Thr) in 2 patients with childhood-onset NS. We found SYNPO2 expression in both podocytes and mesangial cells; however, notably, immunofluorescence staining of adult human and ra

Alkylindole-sensitive receptors modulate microglial cell migration and proliferation.

Ligands targeting G protein-coupled receptors (GPCR) expressed by microglia have been shown to regulate distinct components of their activation process, including cell proliferation, migration and differentiation into M1 or M2 phenotypes. Cannabinoids, including the active component of the Cannabis plant, tetrahydrocannabinol (THC), and the synthetic alkylindole (AI) compound, WIN55212-2 (WIN-2), activate two molecularly identified GPCRs: CB1 and CB2 . Previous studies reported that WIN-2 activates an additional unknown GPCR that is not activated by plant-derived cannabinoids, and evidence ind

Inhibition of endometrial cancer by n-3 polyunsaturated fatty acids in preclinical models.

Although preclinical and epidemiologic studies have shown the importance of n-3 polyunsaturated fatty acids (PUFA) in the prevention of hormone-responsive cancers such as breast cancer, evidence of the association between n-3 PUFAs and endometrial cancer risk is limited and no previous study has examined the effect of n-3 PUFAs on endometrial cancer in cellular and animal models. In this study, we demonstrated that docosahexenoic acid (DHA) dose- and time-dependently inhibited endometrial cancer cell proliferation, colony formation, and migration and promoted apoptosis.
