The requirement of glycoprotein C (gC) for interindividual spread is a conserved function of gC for avian herpesviruses.

We have formerly shown that glycoprotein C (gC) of Gallid alphaherpesvirus 2, better known as Marek's disease (MD) alphaherpesvirus (MDV), is required for interindividual spread in chickens. Since gC is conserved within the Alphaherpesvirinae subfamily, we hypothesized gC was important for interindividual spread of other alphaherpesviruses. To test this hypothesis, we first generated a fluorescent protein tagged clone of Gallid alphaherpesvirus 3 MD vaccine strain 301B/1 to track virus replication in cell culture and chickens using fluorescent microscopy. Following validation of this system, w

Molecular identification and subtype distribution of Blastocystis sp. in farm and pet animals in Turkey.

A total of 1340 fresh fecal samples from farm and pet animals in Central Anatolia and the Middle Black Sea Region of Turkey were investigated using a PCR assay targeting the SSU rRNA of Blastocystis sp. An overall Blastocystis sp. prevalence of 19.4% (183/940) was found in farm animals, including cattle, sheep, water buffaloes, and chickens. Fecal samples of dogs, cats, and horses were negative. The highest prevalence of Blastocystis sp. was found in sheep (38.2%) among the farm animals. The SSU rRNA sequence analysis revealed two animal-specific subtypes, including ST10 in cattle and sheep an

Growth, Carcass Composition, Haematology and Immunity of Broilers Supplemented with Sumac Berries (Rhus coriaria L.) and Thyme (Thymus vulgaris).

Alternatives to antibiotics as growth promoters for broilers could reduce bacterial resistance to antibiotics, while at the same time maintaining growth and improving carcass composition. We investigated the benefits of adding the medicinal plants sumac and thyme at 1, 2 or 3% of the diet for male Ross broiler chicks, with four replicates of ten birds in each treatment group and a Control. Feed intake was reduced for chickens fed the sumac supplements, and, at the two higher doses, defeathered body weight was also reduced. Abdominal fat was reduced by 41% in chickens fed thyme and 62% in those

Experimental infection with avian leukosis virus isolated from Marek's disease vaccines.

Recently, avian leukosis virus (ALV) was isolated from four lots of Marek's disease vaccine produced by two laboratories. The ALVs isolated were characterized by examination of their interactions with cells of two phenotypes (C/E and C/A,E), subgroup-specific polymerase chain reaction (PCR), virus neutralization, envelope gene sequencing, and phylogenetic analysis. All four ALVs are exogenous, belong to subgroup A, and appear to be virtually identical to each other based on PCR and envelope gene nucleotide sequences. We describe herein the characterization of the contaminant viruses in vivo by

The pathogenesis of infectious bursal disease: serologic, histopathologic, and clinical chemical observations.

Infectious bursal disease in 35-day-old specific-pathogen-free (SPF) chickens was characterized clinically by its acute onset and brief duration. Clinical signs included depression, anorexia, diarrhea, and polyuria. A detectable precipitin antibody response occurred between 3 and 5 days postinoculation. Evaluation of pooled serum samples obtained from infectious bursal disease virus (IBDV)-infected chickens revealed transient changes in potassium, cholesterol, uric acid, lactate dehydrogenase, serum glutamic-oxalic transaminase, and serum proteins. Individual serum samples analyzed for uric ac

Oral administration of recombinant Lactobacillus plantarum co expressing FimB, CnaA, NetB and FBA antigens targeting chicken dendritic cells provides effective protection against necrotizing enteritis in broilers.

Necrotic enteritis (NE), an acute intestinal disease of broilers caused by Clostridium perfringens, poses a significant threat to global poultry production. The ban on antibiotic growth promoters in feed, particularly in the EU since 2006, has exacerbated this problem, leading to a resurgence of NE and substantial economic losses. To address this challenge, we developed a novel probiotic-based oral vaccine strategy. We engineered three recombinant Lactobacillus plantarum NC8 strains, NC8(pYL560) expressing three secreted C. perfringens antigens (FimB, NetB, CnaA), NC8(pYL594) expressing a four

Effect of dietary heated hemp seed cake and phytase as soybean meal substitution on broiler chicken performance, carcass yield, visceral organ weight, intestinal health, and serum biochemical parameters.

Hemp seed is a potential candidate to replace soybean meal dependency due to its rich protein and balanced amino acid profile content despite containing anti-nutrients. This study purposed to evaluate dietary heated hemp seed cake (HSC) and the addition of phytase as a substitution for soybean meal on broiler productive performances, carcass characteristics, visceral organ weight, serum biochemical and intestinal health of broiler chickens. A total of 210 unsexed Ross broiler chicks were randomly divided into 7 treatments and 6 replicates using a completely randomized design and reared for 42

Aerogel-based oil sorbents derived from pomelo (Citrus grandis L.) peel powder as fat replacers: effects on physicochemical properties and volatile profiles of semi-dried chicken sausages.

Traditional oleogels require thermal treatment, which promotes lipid oxidation and diminishes health benefits in meat products. This study developed aerogels containing 5-6 % pomelo (Citrus grandis L.) peel powder (125 or 250 μm), which were converted into aerogel-based oleogel-like sorbents (ABOS) by immersion in rice bran oil at room temperature. ABOS were used to replace 20 % pork fat in semi-dried chicken sausages over four days of drying. The reformulated sausages maintained desirable pH (∼5.3) and water activity (∼0.93), while showing 33-46 % reduction in lipid content and 2.8-3.9-fold i

Promotion of Th1 and Th2 responses over Th17 in Riemerella anatipestifer stimulation in chicken splenocytes: Correlation of gga-miR-456-3p and gga-miR-16-5p with NOS2 and CCL5 expression.

Riemerella (R.) anatipestifer poses a significant threat to ducks, resulting in mortality rates ranging from 5-75%. This disease is highly infectious and economically consequential for domestic ducks. Although other avian species, such as chickens, also display susceptibility, the impact is comparatively less severe than in ducks. IL-17A has a pronounced correlation with R. anatipestifer infection in ducks, which is less in chickens. This study performed an in vitro transcriptome analysis using chicken splenic lymphocytes collected at 4-, 8-, and 24-hour intervals following R. anatipestifer st

(-)-Hydroxycitric Acid Alleviates Oleic Acid-Induced Steatosis, Oxidative Stress, and Inflammation in Primary Chicken Hepatocytes by Regulating AMP-Activated Protein Kinase-Mediated Reactive Oxygen Species Levels.

Nonalcoholic fatty liver disease (NAFLD) is one of the most complex liver diseases in the world, which is characterized by hepatic steatosis, oxidative stress, inflammation, and apoptosis. (-)-Hydroxycitric acid [(-)-HCA] can regulate obesity in different animals, while whether this beneficial effect of (-)-HCA can alleviate the NAFLD and its mechanism is unclear. Hence, this study aimed to determine the potential actions and mechanisms of (-)-HCA on NAFLD in oleic acid (OA)-induced hepatocytes. We found that (-)-HCA effectively improved OA-induced hepatic steatosis by regulating the expressio

Dynamic profiles, biodistribution and integration evaluation after intramuscular/intravenous delivery of a novel therapeutic DNA vaccine encoding chicken type II collagen for rheumatoid arthritis in vaccinated normal rodent.

BACKGROUND: The persistence, biodistribution, and risk of integration into the host genome of any new therapeutic DNA vaccine must be established in preclinical studies. We previously developed the DNA vaccine pcDNA-CCOL2A1 encoding chicken type II collagen (CCII) for the treatment of rheumatoid arthritis (RA). In the present study, we characterized its dynamic profile, biodistribution, and potential for genomic DNA integration in normal vaccinated rodent. RESULTS: A real-time quantitative PCR analysis (RT-qPCR) of animals administered a single dose of pcDNA-CCOL2A1 (300 μg/kg by intramuscula

Valproic acid substantially downregulated genes folr1, IGF2R, RGS2, COL6A3, EDNRB, KLF6, and pax-3, N-acetylcysteine alleviated most of the induced gene alterations in chicken embryo model.

Valproic acid induced teratogenicity at genetic and somatic levels, the action mechanism is still unclear. We hypothesized that folate receptor gene (folr1) and others may be interacting to elicit neural tube defect (NTD), while N-acetylcysteine (NAC) may be beneficial for protection. In chicken embryo model, the experiment was conducted in two parts. The first part was carried out to test the optimum dose of VPA. The second part was conducted to test the protective effect of NAC at doses 10 and 20 mM. VPA induced dysvascularization, incomplete somite enclosure, histone deacetylase (HDAC) inhi

Genetic characterisation of extended-spectrum β-lactamases in Escherichia coli isolated from retail chicken products including CTX-M-9 containing isolates: a food safety risk factor.

1. Bacterial resistance to β-lactam antibiotics has risen dramatically in Escherichia coli from food animals. In a previous study, 29 randomly selected chicken products, collected in Portugal, were analysed for the presence of extended-spectrum β-lactamases (ESBLs)-producing E. coli; and during this study the genetic characterisation of ESBLs genes was investigated. 2. The presence of genes encoding TEM, OXA, SHV, and CTX-M type beta-lactamases was studied by PCR followed by sequencing. Additionally, other mechanisms of antimicrobial resistance, phylogenetic groups and the presence of virulenc

Dispersal of the northern fowl mite, Ornithonyssus sylviarum (Canestrini and Fanzago), and the chicken body louse, Menacanthus stramineus (Nitzsch), among thirty strains of egg-type hens in a caged laying house.

From September 1978 through Februrary 1979, dispersal of uncontrolled, naturally occurring populations of the northern fowl mite, Ornithonyssus sylviarum (Canestrini and Fanzago), and the chicken body louse, Menacanthus stramineus (Nitzch) was studied on 30 strains of egg-type pullets reared to 20 weeks old on four growing rations before being housed for egg production. Hens representing all 30 strains and each of the four feeding regimens had mites. The northern fowl mite spread from initial infestations down the entire length of the house in 1 month; after four months, the chicken body louse

Identification of virulence-related amino acid mutations of avian encephalomyelitis virus associated with vaccination methods.

Avian encephalomyelitis virus (AEV), a picornavirus, primarily infects the central nervous system of 1 to 2-week-old young chickens but not pullets. When wild-type AEV undergoes serial passaging in chicken embryos, it becomes to be embryo-adapted and can cause avian encephalomyelitis in chickens of all ages following intracutaneous infection through parenteral routes. This study was conducted to explore whether an outbreak of AEV in 95-day-old chickens was linked to inadvertent embryo adaptation of the AEV vaccine and its association with vaccination method. In this study, an AEV strain AEV/JS
