Axonal ER Ca2+ Release Selectively Enhances Activity-Independent Glutamate Release in a Huntington Disease Model.

Action potential (AP)-independent (miniature) neurotransmission occurs at all chemical synapses but remains poorly understood, particularly in pathologic contexts. Axonal endoplasmic reticulum (ER) Ca2+ stores are thought to influence miniature neurotransmission, and aberrant ER Ca2+ handling is implicated in progression of Huntington disease (HD). Here, we report elevated mEPSC frequencies in recordings from YAC128 mouse (HD-model) neurons (from cortical cultures and striatum-containing brain slices, both from male and female animals). Pharmacological experiments suggest that this is mediated

Engineering and Delivery of cGAS-STING Immunomodulators for the Immunotherapy of Cancer and Autoimmune Diseases.

The cyclic GMP-AMP synthase-stimulator interferon gene (cGAS-STING) pathway is an emerging therapeutic target for the prophylaxis and therapy of a variety of diseases, ranging from cancer, infectious diseases, to autoimmune disorders. As a cytosolic double stranded DNA (dsDNA) sensor, cGAS can bind with relatively long dsDNA, resulting in conformational change and activation of cGAS. Activated cGAS catalyzes the conversion of adenosine triphosphate (ATP) and guanosine triphosphate (GTP) into cGAMP, a cyclic dinucleotide (CDN). CDNs, including 2'3'-cGAMP, stimulate adapter protein STING on the

HMGB1-mediated autophagy regulates sodium/iodide symporter protein degradation in thyroid cancer cells.

BACKGROUND: Sodium/iodide symporter (NIS)-mediated iodide uptake plays an important physiological role in regulating thyroid gland function, as well as in diagnosing and treating Graves' disease and thyroid cancer. High-mobility group box 1 (HMGB1), a highly conserved nuclear protein, is a positive regulator of autophagy conferring resistance to chemotherapy, radiotherapy and immunotherapy in cancer cells. Here the authors intended to identify the role of HMGB1 in Hank's balanced salt solution (HBSS)-induced autophagy, explore NIS protein degradation through a autophagy-lysosome pathway in thy

Effect of Ciprofloxacin on Susceptibility to Aortic Dissection and Rupture in Mice.

IMPORTANCE: Fluoroquinolones are among the most commonly prescribed antibiotics. Recent clinical studies indicated an association between fluoroquinolone use and increased risk of aortic aneurysm and dissection (AAD). This alarming association has raised concern, especially in patients with AAD with risk of rupture and in individuals at risk for developing AAD. OBJECTIVE: To examine the effect of ciprofloxacin on AAD development in mice. DESIGN, SETTING, AND PARTICIPANTS: In a mouse model of moderate, sporadic AAD, 4-week-old male and female C57BL/6J mice were challenged with a high-fat diet

Enhanced binding of calmodulin to RyR2 corrects arrhythmogenic channel disorder in CPVT-associated myocytes.

AIMS: Calmodulin (CaM) plays a key role in modulating channel gating in ryanodine receptor (RyR2). Here, we investigated (a) the pathogenic role of CaM in the channel disorder in CPVT and (b) the possibility of correcting the CPVT-linked channel disorder, using knock-in (KI) mouse model with CPVT-associated RyR2 mutation (R2474S). METHODS AND RESULTS: Transmembrane potentials were recorded in whole cell current mode before and after pacing (1-5 Hz) in isolated ventricular myocytes. CaM binding was assessed by incorporation of exogenous CaM fluorescently labeled with HiLyte Fluor(®) in saponin

Mitochondrial JNK phosphorylation as a novel therapeutic target to inhibit neuroinflammation and apoptosis after neonatal ischemic brain damage.

Neonatal encephalopathy is associated with high mortality and life-long developmental consequences. Therapeutic options are very limited. We assessed the effects of D-JNKi, a small peptide c-Jun N-terminal kinase (JNK) MAP kinase inhibitor, on neuroinflammation, mitochondrial integrity and neuronal damage in a neonatal rat model of ischemic brain damage. Hypoxic-ischemic (HI) brain injury was induced in postnatal-day 7 rats by unilateral carotid artery occlusion and hypoxia, and was followed by intraperitoneal D-JNKi treatment. We demonstrate here for the first time that a single intraperitone

Targeting of human catalase to peroxisomes is dependent upon a novel COOH-terminal peroxisomal targeting sequence.

We have identified a novel peroxisomal targeting sequence (PTS) at the extreme COOH terminus of human catalase. The last four amino acids of this protein (-KANL) are necessary and sufficient to effect targeting to peroxisomes in both human fibroblasts and Saccharomyces cerevisiae, when appended to the COOH terminus of the reporter protein, chloramphenicol acetyl transferase. However, this PTS differs from the extensive family of COOH-terminal PTS tripeptides collectively termed PTS1 in two major aspects. First, the presence of the uncharged amino acid, asparagine, at the penultimate residue of

A rapid and efficient in planta assay for assessment of RNA editing factor performance.

Land plant mitochondria and plastids are endosymbiotic organelles that possess their own, reduced genomes. Gene expression is largely regulated at the transcript level, predominantly by nuclear-encoded proteins. Notably, up to thousands of cytidines in organellar RNAs are specifically edited to uridines, mostly restoring conserved genetic information. Key players in this process are nuclear-encoded pentatricopeptide repeat (PPR) proteins, which bind target RNAs in a one-PPR:one-nucleotide manner, following a two-amino-acid-based code. Cytidine deamination is catalyzed by intrinsic or trans-act

Intracellular Fate of a Dual-Fluorescent Hydrophobic Ion Pair: Comparison of Lipid-Based Nanocarriers.

Effective intracellular trafficking and delivery of hydrophilic drugs remain challenging due to poor membrane permeability and limited encapsulation in lipid-based nanocarriers. To address this, we developed a dual-fluorescent hydrophobic ion pair (HIP) by pairing a model fluorescent hydrophilic drug, Cascade Blue hydrazide, with the lipophilic probe DiA. The HIP was subsequently incorporated into three lipid-based nanocarriers─self-emulsifying drug delivery systems (SEDDS), nanoemulsions, and liposomes─to enable visualization and comparison of how formulation composition influences intracellu

Genome-wide analysis of FATA associated with drought tolerance in tetraploid potato (Solanum tuberosum).

The cuticle represents the outer most protective barrier against biotic and abiotic stresses. It is composed of cutin and waxes and protects plants from desiccation, UV, cold, mechanical stresses, and pathogens. GWAS/BSAseq combined with SeqSNP analyses in an association panel of 34 potato cultivars had revealed that the acyl-ACP thioesterase FATA (Soltu.DM.06G033680.1) is significantly associated with drought tolerance in potato. Apart from three FATB genes, only one FATA gene is present in potato that has the highest homology to FATA2 in Arabidopsis. FATA is responsible for the export of C18

A novel photosensitizer (hE26) for photodynamic therapy enhances anticancer efficacy via reactive oxygen species-induced mitochondrial apoptosis in non-small cell lung cancer.

INTRODUCTION: Non-small cell lung cancer (NSCLC) remains a leading cause of mortality and thus requires novel therapeutic interventions. Although our previously synthesized phosphorus tetraphenylporphyrin (E26) effectively induces cell death via photodynamic therapy (PDT), its 610-nm excitation wavelength limits deep-tissue penetration. To address this limitation, we developed a novel hydrogenated derivative, hE26, in which excitation is shifted to an optimized longer wavelength of 660 nm. METHODS: The anticancer potential of the novel hE26-mediated PDT and the former compound E26 was compare

Vesicular nucleotide transporter (VNUT)-dependent ATP secretion by hepatic stellate cells promotes liver fibrosis.

Although liver fibrosis presents a substantial global health challenge, therapeutic options that directly target liver fibrosis remain limited. Hepatic stellate cells (HSCs) are key contributors to fibrosis through extracellular matrix production. This study uncovers a previously unrecognized function of HSCs: ATP secretion. We found that HSCs express the vesicular nucleotide transporter (VNUT) on secretory vesicles and actively release ATP. In mouse HSCs, VNUT is localized to the cytosol and around lipid droplets. In a thioacetamide-induced liver fibrosis model, VNUT inhibition with clodronat

The parasitophorous vacuole membrane of Toxoplasma gondii: A custom structural framing with smart interior plumbing for resource allocation.

In essence, the parasitophorous vacuole (PV) of Toxoplasma is a "Do-It-Yourself" renovation project led by the parasite after invasion of a guest house (the host cell) to install itself surreptitiously in a private and custom-built room, reinforced by drywall (the PV membrane). The function of the PV membrane (PVM) is to keep away the host security guards (immune response, lysosomes) from the parasite while allowing necessary and controlled access to host resources. Inside the PV, Toxoplasma relies on a crew of creative interior designers (the GRA proteins or GRA) to remodel and transform the

Neuronal TLR4 upregulation activates the cGAS-STING pathway to induce ferroptosis in EAE mice.

INTRODUCTION: Progressive neurofunctional impairment in multiple sclerosis (MS) is largely driven by neuronal damage and loss, yet the underlying molecular mechanisms remain poorly understood. This study aimed to investigate the role of neuronal Toll-like receptor 4 (TLR4) in promoting ferroptosis, an iron-dependent cell death pathway, during experimental autoimmune encephalomyelitis (EAE). METHODS: We leveraged a MOG35-55-induced EAE mouse model (n = 10 per group) alongside in vitro LPS-stimulated SH-SY5Y mono- and co-culture systems (n = 3 biological replicates) to interrogate the crosstalk

Focal adhesion assembly and cell migration require myoferlin in PDAC cell lines.

Pancreatic ductal adenocarcinoma (PDAC) has a dire prognosis due to late diagnosis and ineffective treatments. Myoferlin, a transmembrane protein, is overexpressed in PDAC and correlates with poor patient survival, making it a potential therapeutic target. Several reports indicated that myoferlin depletion in cancer cells reduced migration or metastatic dissemination, but they failed to provide a consistent mechanism. Understanding the biology behind the dissemination of PDAC is of outmost importance, as this disease is often diagnosed at an advanced stage. This study aims to further elucidate

Intratumoral microbiome composition and its role in tumor recurrence in primary liver cancer.

BACKGROUND: The human microbiome is increasingly recognized as a factor in cancer development, though its role in primary liver cancer (PLC) remains unclear. This retrospective cohort study examines tissue-specific microbiota differences between hepatocellular carcinoma (HCC) and cholangiocarcinoma (CCA), and explores their association with tumor recurrence in PLC. METHODS: Clinical data from 116 patients (85 HCC and 31 age-matched CCA) were analyzed. Fresh frozen samples underwent RNAscope™ ISH assay for microbial RNA detection, while the MiSeq platform targeted 16 S ribosomal RNA. Microbiom

Cells Dying via the AP-3 Complex-Dependent Regulated Death Pathway Support the Surviving Cells under Amino Acid Deficiency Conditions.

Starvation can initiate a programmed death in unicellular organisms. It is believed that cells committing suicide via this mechanism support surviving cells with their nutrients. It was recently discovered that heating Saccharomyces cerevisiae yeast cells to 51°C kills the cells though a special pathway that starts with the decomposition of the lysosomal membrane followed by the loss of the plasma membrane integrity a few hours later. Since lysosomes contain hydrolases of nitrogen-containing biopolymers, this programmed death pathway could help the survivors during nitrogen starvation. The los

Nitric oxide-induced Na+/H+ exchange activity confers salt tolerance in pea (Pisum sativum L.) mesophyll cells.

Salinity stress severely hinders global agricultural productivity, and the issue will only increase under current climate scenarios. Due to complexity of salinity tolerance traits, crop breeding for salt tolerance remains a highly challenging task. The exogenous application of growth regulators, such as nitric oxide (NO), is considered a viable practical alternative to boost crop yield and quality under conditions of soil salinity. Numerous papers reported beneficial role of exogenous NO application on plant growth under salt stress, but very few explored the mechanistic basis of this process.

Activation of toll-like receptor 2 promotes the expression of inflammatory mediators and cell proliferation of human polycystic kidney disease cells.

Autosomal dominant polycystic kidney disease (ADPKD) is characterized by the progressive enlargement of fluid-filled cysts, leading to a decline in renal function. Toll-like receptors (TLR)-2 and TLR4 are pattern recognition receptors and components of the innate immune response. We found that mRNA levels for TLR2 and TLR4, an adaptor protein MyD88, and the transcription factor NF-κB were elevated in the kidneys of ADPKD patients and PKD mice. There was decreased expression of IκBα, an inhibitory protein sequestering NF-κB in the cytosol, and increased NF-κB nuclear translocation in human ADPK

Mitochondrial calcium shapes B cell signaling and mitochondrial function.

INTRODUCTION: Calcium (Ca2+) signaling plays a pivotal role in determining B cell fate, shaping processes such as activation, differentiation, anergy or apoptosis. Upon B cell antigen receptor activation, Ca2+ is rapidly mobilized from the endoplasmic reticulum and supplemented by Ca2+ influx from the extracellular space, ultimately driving activation of various signaling pathways required for appropriate B cell responses. Although mitochondria also harbor significant levels of Ca2+, how mitochondrial Ca2+ dynamics are regulated in B cells in response to activation or other cues remains unknow
