Decursin ameliorates diabetic kidney disease by attenuating renal epithelial-mesenchymal transition via inhibition of the PI3K/Akt pathway.

OBJECTIVE: This study aims to elucidate the therapeutic mechanisms of Decursin (DE) against diabetic kidney disease (DKD), focusing on its effects on renal epithelial-mesenchymal transition (EMT) and the underlying signaling pathways. METHODS: A streptozotocin (STZ)-induced DKD model was established in rats and treated with DE for 12 weeks. Metabolic and renal function indices were assessed using blood and urine samples, while kidney tissues were subjected to histopathological examination. Network pharmacology and disease database analyses were employed to predict the core target genes of DE

Marek's disease virus infection alters the composition of exosomes in chicken fibroblasts and promotes viral replication.

Marek's disease (MD) is a highly contagious lymphoproliferative disease caused by Marek's disease virus (MDV), typically characterized by severe immunosuppression. In this study, exosomes were isolated and purified from chicken embryo fibroblasts (CEFs) infected with the virulent MDV strain Md5 (Exo-Md5) and uninfected controls (Exo-NC), respectively. Combining small RNA sequencing, mass spectrometry, and Western blot analysis, we confirmed that MDV infection significantly remodels the exosomal cargo composition. Exo-Md5 exhibited a predominant downregulation of miRNAs. Subsequent GO and KEGG

The prevalence of protein misfolding as a mechanism for hereditary deafness.

Hearing loss is the most common sensory deficit impacting ~5% of the world's population. The Deafness Variation Database (DVD) is a public resource of deafness variants, containing 381,924 missense variants across 224 genes, with 303,577 classified as a variant of uncertain significance (VUS). To address the challenge of evaluating each deafness associated VUS, we evaluate a family of probabilistic frameworks to quantify the strength of computational evidence based on ACMG/AMP recommendations. First, CADD and REVEL are compared using Bayesian models parameterized using either a ClinVar 2019 da

CD47 as a prognostic biomarker and potential immunotherapy target in penile squamous cell carcinoma.

Penile squamous cell carcinoma (PSCC) remains a global healthcare concern, characterized by poor outcomes and limited therapeutic options. CD47, a “don’t-eat-me” signal that enables tumor immune evasion, has not been extensively evaluated in PSCC. This study aims to explore the expression and significance of CD47 in PSCC. RNA-sequencing was performed on eight paired PSCC and adjacent normal tissues. Protein expression was assessed by immunohistochemistry (IHC) in an independent of 131 PSCC cases, and associations with clinicopathological factors and prognosis was evaluated. In addition, the in

Recombinant multiepitope proteins for hepatitis C virus diagnosis: Systematic review.

Hepatitis C virus (HCV) infection remains as major global health concern given the more than 50 million chronic cases recorded and the significant barriers to its early diagnosis, mainly in low-resource settings. Conventional serological assays rely on recombinant or synthetic antigens accounting for higher production costs and for limiting diagnostic accessibility. Recombinant multiepitope proteins have emerged as promising alternative for incorporating conserved antigenic regions from multiple HCV proteins, such as Core, NS3, NS4 and NS5, into a single construct capable of broadly detecting

CYLD inhibits ubiquitin-mediated degradation of IRE1 to promote inflammatory injury in ulcerative colitis.

BACKGROUND: Previous studies have shown that elevated inositol-requiring enzyme 1 (IRE1) protein levels exacerbate ulcerative colitis (UC). However, the mechanism underlying the increase in IRE1 protein levels in UC remains unclear. Deubiquitinating enzymes (DUBs) have been demonstrated to play a key role in regulating intestinal inflammation. This study aimed to investigate the DUB that influenced IRE1 levels in UC and its specific regulatory mechanisms. METHODS: The Ubibrowser 2.0 database was used to screen out IRE1-related DUBs. The combination of DUBs and IRE1 was verified by Co-IP detec

Integrating bioinformatics and machine learning analyses to identify immune-related secretory proteins and therapeutic small-molecule drugs in calcific aortic valve disease with type 2 diabetes.

INTRODUCTION: Type 2 diabetes mellitus (T2DM) is a globally prevalent metabolic disease, and emerging studies have revealed its strong association with calcific aortic valve disease (CAVD). Chronic inflammation, oxidative stress, and immune dysregulation induced by hyperglycemia in T2DM may accelerate CAVD progression, although the molecular mechanisms remain unclear. METHODS: We integrated and analyzed four CAVD and two T2DM gene expression datasets from the GEO database. Through differential gene expression analysis, weighted gene co-expression network analysis (WGCNA), and secretory protei

Advanced Paternal Age and Sperm Proteome Dynamics: A Possible Explanation for Age-Associated Male Fertility Decline.

Male fertility is strongly influenced by environmental exposures, lifestyle, and advancing age. While advanced paternal age (APA) has been linked with a progressive decline in male fertility, poor reproductive outcomes, and decreased offspring health, the molecular mechanisms underlying these alterations remain unclear. In this work, we investigated the impact of men's age on human sperm protein expression and phosphorylation to identify molecular alterations possibly responsible for the age-associated decline in male fertility. Semen samples from volunteers attending fertility consultations a

Ursolic acid ameliorates ocular surface dysfunction in dry eye via targeting EGFR/RAS/RAF/MAP2K1/MAPK1 pathway.

Dry eye (DE), a multifactorial ocular surface disease, is predominantly characterized by inflammation as a central pathological factor. Ursolic acid (UA), a pentacyclic triterpenoid with well-documented anti-inflammatory properties, was evaluated in this study for its therapeutic effects on ocular surface dysfunction associated with DE and its underlying mechanisms. A hyperosmotic stress model (500 mOsM) using human corneal epithelial cells (HCEs) and an animal model of DE was established to assess UA's protective effects on both cellular and organismal levels. Comprehensive assessments, inclu

Quantifying tumor specificity using Bayesian probabilistic modeling for drug target discovery and prioritization.

In diseases such as cancer, the design of new therapeutic strategies requires extensive, costly, and unfortunately sometimes deadly testing to reveal life threatening "off target" effects. A crucial first step in predicting toxicity are analyses of normal RNA and protein tissue expression, which are now possible using comprehensive molecular tissue atlases. However, no standardized approaches exist for target prioritization, which instead rely on ad-hoc thresholds and manual inspection. Such issues are compounded, given that genomic and proteomic data detection sensitivity and accuracy are oft
