Targeting the ubiquitin-proteasome system for cancer treatment: discovering novel inhibitors from nature and drug repurposing.

In the past 15 years, the proteasome has been validated as an anti-cancer drug target and 20S proteasome inhibitors (such as bortezomib and carfilzomib) have been approved by the FDA for the treatment of multiple myeloma and some other liquid tumors. However, there are shortcomings of clinical proteasome inhibitors, including severe toxicity, drug resistance, and no effect in solid tumors. At the same time, extensive research has been conducted in the areas of natural compounds and old drug repositioning towards the goal of discovering effective, economical, low toxicity proteasome-inhibitory

Genetics-directed drug discovery for combating Mycobacterium tuberculosis infection.

Mycobacterium tuberculosis (Mtb), the pathogen of tuberculosis (TB), is one of the most infectious bacteria in the world. The traditional strategy to combat TB involves targeting the pathogen directly; however, the rapid evolution of drug resistance lessens the efficiency of this anti-TB method. Therefore, in recent years, some researchers have turned to an alternative anti-TB strategy, which hinders Mtb infection through targeting host genes. In this work, using a theoretical genetic analysis, we identified 170 Mtb infection-associated genes from human genetic variations related to Mtb infect

In vitro praziquantel test capable of detecting reduced in vivo efficacy in Schistosoma mansoni human infections.

Although great reductions in human schistosomiasis have been observed after praziquantel (PZQ) mass drug administration (MDA), some individuals remain infected after multiple treatments. Many MDA programs now require monitoring for drug efficacy as a key component. No molecular tools for PZQ resistance currently exist and investigations into the dose of PZQ required to kill 50% of adult worms in vivo (ED(50)) present ethical, logistical, and temporal restraints. We, therefore, assessed the feasibility and accuracy of a rapid, inexpensive in vitro PZQ test in the laboratory and directly in the

An effectiveness and cost analysis of presumptive treatment for Mycobacterium tuberculosis.

BACKGROUND: Delay in treatment of tuberculosis has contributed to both the spread of tuberculosis and its case fatality rate. METHODS: Decision analysis was used to examine the effectiveness and cost of presumptive treatment in patients evaluated for tuberculosis. RESULTS: Over a range of assumptions, empiric antituberculous therapy for acid-fast bacillus smear-positive persons lowers mortality and cost per person evaluated when available rapid diagnostic laboratory methods for tuberculosis are used. In contrast, the average cost per life saved by giving presumptive treatment to all acid-fas

Antimicrobial-resistant Neisseria gonorrhoeae can be targeted using inhibitors against evolutionary conservedl-asparaginase.

The emergence of multiple drug-resistant "super gonorrhoea" complicates the management and treatment of Neisseria gonorrhoeae infections due to the progressive accumulation of mutations in the biological targets of frontline antimicrobials. Continuous evaluation and reporting of newer molecular targets and their inhibitors are necessary. Here, we present l-asparaginase of N. gonorrhoeae (NgA) as a new molecular target based on structure-based high-throughput screening, molecular dynamics(MD) simulations, and validation by biophysical, biochemical, and cell viability assays. We observed that th

Metabolomic Profiling of the Malaria Box Reveals Antimalarial Target Pathways.

The threat of widespread drug resistance to frontline antimalarials has renewed the urgency for identifying inexpensive chemotherapeutic compounds that are effective against Plasmodium falciparum, the parasite species responsible for the greatest number of malaria-related deaths worldwide. To aid in the fight against malaria, a recent extensive screening campaign has generated thousands of lead compounds with low micromolar activity against blood stage parasites. A subset of these leads has been compiled by the Medicines for Malaria Venture (MMV) into a collection of structurally diverse compo

Advancements in polymer-based drug delivery systems targeting the tumor microenvironment for immunotherapy of triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype characterized by high invasiveness, frequent recurrence, and metastasis, along with limited targeted therapeutic options. Despite clinical advances achieved with immune checkpoint inhibitors, poly(ADP-ribose) polymerase (PARP) inhibitors, and antibody-drug conjugates, the complex immunosuppressive tumor microenvironment (TME) of TNBC hinders drug delivery and compromises the efficacy of immunotherapy. Multiple adverse features of the TME, including impaired tumor immunogenicity, dysfunctional antigen presentation, dens

Antimicrobial Resistance: A Growing Serious Threat for Global Public Health.

Antibiotics are among the most important discoveries of the 20th century, having saved millions of lives from infectious diseases. Microbes have developed acquired antimicrobial resistance (AMR) to many drugs due to high selection pressure from increasing use and misuse of antibiotics over the years. The transmission and acquisition of AMR occur primarily via a human-human interface both within and outside of healthcare facilities. A huge number of interdependent factors related to healthcare and agriculture govern the development of AMR through various drug-resistance mechanisms. The emergenc

Limited contribution of compensatory mutations to MDR/RR-TB clustering in Hunan Province, China: a population-based whole-genome sequencing study.

Transmission of rifampicin-resistant and multidrug-resistant tuberculosis (MDR/RR-TB) is a major driver of the global drug-resistant TB epidemic, yet the contribution of compensatory mutations to its spread remains uncertain. We analyzed 206 non-duplicate MDR/RR-TB isolates collected from five surveillance sites in Hunan Province, China, between 2013 and 2020 using whole-genome sequencing. Transmission clusters were defined by ≤12 single-nucleotide polymorphism (SNP) differences, and compensatory mutations in rpoA, rpoB, and rpoC were identified through a curated mutation catalog and phylogene
