Enhanced ITGA3 expression induced by helicobacter pylori infection facilitates gastric cancer progression via NF-κB and Smad4.

Helicobacter pylori (H. pylori) infection is a key risk factor in the onset and progression of gastric cancer (GC). The integrin subunit alpha 3 (ITGA3) is frequently upregulated in various malignancies and may influence cell proliferation and metastasis under specific conditions. However, the mechanisms that govern ITGA3 activation and its role in the inflammation-driven transformation of GC remain poorly understood. This study explores the impact of ITGA3 in response to H. pylori infection and its involvement in gastric tumorigenesis. Elevated ITGA3 expression was observed in gastric tissues

GLUT1/SGLT1 balance associates with high-glucose buffering of metformin-induced lysosomal deacidification and mTORC1 inhibition in colorectal cancer.

Metformin has anticancer potential, but benefit in colorectal cancer (CRC) is heterogeneous and determinants of tumor cell response remain unclear. We tested whether extracellular glucose and the relative expression of glucose transporter 1 (GLUT1/SLC2A1) and sodium-glucose cotransporter 1 (SGLT1/SLC5A1), quantified as a glucose transporter ratio score (GTRS), are associated with buffering of metformin effects. We combined a 1,2-dimethylhydrazine (DMH)-induced CRC model, glucose-controlled cell-line experiments, and analyses of public bulk, single-cell, and spatial transcriptomic datasets. In

Molecular Profiling Identifies CD49d and CD79b as Predictive Markers for Acquired Acalabrutinib Resistance in Patients With Chronic Lymphocytic Leukemia.

Contemporary studies of Bruton tyrosine kinase inhibitor (BTKi) resistance focus on mutations in the B-cell receptor (BCR) pathway, but alternative mechanisms of resistance remain undefined. Here, we sought to identify novel predictive markers of acquired resistance to acalabrutinib, a second-generation BTKi, in patients with chronic lymphocytic leukemia (CLL). Clinical samples from 41 patients with relapsed/refractory or treatment-naive CLL receiving acalabrutinib as part of a clinical trial (NCT02029443) were divided into two groups: those who continued to respond to treatment (NP, n = 23) a

NADPH oxidase 2 inhibitor GSK2795039 exerts antiplatelet and antithrombotic activity.

Reactive oxygen species (ROS) production by NADPH oxidase (NOX) is a key promoter of platelet activation, making NOX inhibition an attractive antiplatelet strategy. This study evaluated the effects of the novel NOX2 inhibitor, GSK2795039, on human platelet functional responses and ROS-mediated signaling pathways. GSK2795039 effectively suppressed both extracellular and intracellular ROS production in collagen-stimulated platelets. Consequently, GSK2795039 treatment significantly inhibited collagen-induced platelet aggregation, dense-granule ATP release, and α-granule P-selectin exposure. Furth

Substrate stiffness modifies gene expression and transcriptional response of equine endometrial fibroblasts to TGF-β1.

Equine endometrial fibrosis is a leading cause of subfertility in aging mares. Fibrosis is a reparative response involving excess deposition of extracellular matrix (ECM) and increasing tissue stiffness. Augmented rigidity itself can drive fibrosis, by stimulating transition from fibroblasts to myofibroblasts. Myofibroblasts release latent transforming growth factor beta 1 (TGF-β1) from the ECM, thereby activating this profibrotic cytokine. Tissue culture polystyrene (TCP) is commonly used for in vitro experiments. The endometrium, however, is considerably softer than TCP. This study criticall

Stratification of brain-derived extracellular vesicles of Alzheimer's disease patients indicates a unique proteomic content and a higher seeding capacity of small extracellular vesicles.

BACKGROUND: Alzheimer's disease (AD) is the most prominent form of dementia worldwide. It is characterized by tau lesions that spread throughout the brain in a spatio-temporal manner. This has led to the prion-like propagation hypothesis implicating a transfer of pathological tau seeds from cell to cell. Human brain-derived extracellular vesicles (BD-EVs) isolated from the brain-derived fluid of AD patients contain seeds that contribute to this tau pathology spreading. Knowing the rich diversity of EVs, isolation of functional EV sub-populations is required to unravel their implication in the

Targeting OxLDL-mediated CD36 + CAF reprogramming potentiates PD-1 immunotherapy in osteosarcoma.

BACKGROUND: Osteosarcoma demonstrates limited responsiveness to PD-1 blockade, largely due to its immunosuppressive tumor microenvironment (TME). The specific mechanisms by which cancer-associated fibroblasts (CAFs) contribute to immunosuppression in osteosarcoma are not fully understood. METHODS: We performed single-cell RNA sequencing (scRNA-seq) on osteosarcoma tissues from patients treated with neoadjuvant chemotherapy and anti-PD-1 therapy to investigate the tumor microenvironment. Cellular composition, gene expression programs, and signaling pathways were analyzed. Functional assays, pu

Tregs With High CD29 Expression Promote Cell Adhesion and Contribute to the Malignant Transformation of MASLD.

BACKGROUND AND AIM: Regulatory T cells (Tregs) are highly enriched in the metabolic dysfunction-associated steatotic liver disease (MASLD) microenvironment, but their role in driving metabolic dysfunction-associated steatohepatitis (MASH) progression to hepatocellular carcinoma (HCC) remains unclear. Here, it is demonstrated that integrin β1 (ITGβ1, CD29) expression is upregulated by Tregs, enhancing cell adhesion and driving the malignant transformation of MASLD. METHODS: A MASLD mouse model was established via high-fat diet (HFD) and 2-acetylamino fluorene (2-AAF). Single-cell RNA sequencin

Mechanism of retinal angiogenesis induced by HIF-1α and HIF-2α under hyperoxic conditions.

In retinopathy of prematurity (ROP), preventing avascular dysplasia may be more critical than inhibiting abnormal neovascularization. While hypoxia-inducible factors (HIFs) are implicated in angiogenesis, their role in preventing ROP remains unclear. Oxygen-induced retinopathy (OIR) model and hyperoxic cell model were used in this study. Immunofluorescence, western blot, ELISA, cell counting kit-8 (CCK-8), and flow cytometry were applied to assess the effects of hyperoxia on the astrocytes. Co-culture of astrocytes with retinal microvascular endothelial cells (RMECs) was used to observe the ef

Tartrate-Resistant Acid Phosphatase 5 (TRAP5) Promotes Eosinophil Migration During Allergic Asthma.

Allergic asthma is characterized by Type 2 inflammation and eosinophilia. Tartrate-resistant acid phosphatase 5 (TRAP5; also referred to as acid phosphatase 5 [ACP5]) is a metallophosphatase expressed by alveolar macrophages that dephosphorylates osteopontin, a phosphoglycoprotein with increased expression in asthma. To investigate the role of TRAP5 during asthma, we used a murine model of ovalbumin (OVA)-induced allergic airway inflammation as well as IL-33-induced airway inflammation, including Trap5-/- and wild-type (WT) mice. Histological analyses of murine lung revealed that OVA-induced i

CTC cluster in breast cancer: synergetic metastasis promotion mechanism and transformation path from platelet cloaking to leukocyte escort.

Circulating tumor cell (CTC) clusters are potent drivers of metastasis in breast cancer, and their survival and dissemination within the circulatory system are critically governed by dynamic interactions with the host microenvironment. Two key mechanisms-"platelet cloaking" and "leukocyte escort"-have emerged as central axes that amplify the metastatic potential of these clusters. This review synthesizes these parallel yet intersecting pathways, elucidates the biological basis for their synergistic interplay, and proposes a translational roadmap from mechanistic insight to clinical application

SIRT1 Mediates the Effects of Sera from Athletes Who Engage in Aerobic Exercise Training in Activating Cells for Wound Healing.

Background/Objectives: Exercise training (ET) can improve wound healing and prevent the recurrence of skin lesions. Aerobic ET stimulates the NAD+-dependent deacetylase sirtuin 1 (SIRT1). The beneficial effects of ET and SIRT1 activation in wound healing have been characterized when considered separately. This study aimed to investigate the potential role of SIRT1 as a mediator of the effects of sera isolated from athletes who regularly participate in aerobic ET (middle-distance running, MDR) on cells primarily involved in wound healing. Methods: Human keratinocytes, fibroblasts and endothelia

Quantitative serum proteomic analysis for biomarker discovery in post-COVID-19 postural orthostatic tachycardia syndrome (PC-POTS) patients.

Postural orthostatic tachycardia syndrome (POTS) is a chronic, debilitating condition that is characterized by an excessive increase in heart rate upon orthostatic challenge. Before the COVID-19 pandemic, POTS affected 0.5 % to 1 % of the U.S. population. Since the pandemic, the incidence has risen sharply, adding an estimated 6-7 million new cases in the U.S. Despite its importance, there is currently no reliable biomarker for POTS, leading to significant diagnostic delays. A major hurdle in identifying biomarkers is the heterogeneous nature of the syndrome. To address this, we focused on a h

Angiopoietin-2 regulates the phenotypic switch of vascular smooth muscle cells.

During uterine spiral artery remodeling, vascular smooth muscle cells (VSMCs) become disorganized and undergo phenotypic switching from a contractile to a more synthetic phenotype. We have previously reported that uterine natural killer cells induce this VSMC phenotypic switching by secreting angiopoietin-2 (Ang-2). Here, we identified the specific mechanisms by which Ang-2 plays a role in this phenomenon. VSMCs isolated from human umbilical arteries were used as an in vitro model to investigate the role of Ang-2 in phenotypic switching. Human decidua tissue from preeclamptic and control pregn

Multifunctional nanoparticle-mediated targeting of metabolic reprogramming and DNA damage response pathways to treat drug-resistant triple-negative breast cancer.

Multi-drug resistance and immunosuppressive triple-negative breast cancer (TNBC) is triggered by the Warburg effect, which promotes homologous recombination repair (HRR) and upregulates expression of P-glycoprotein (P-gp), in turn preventing DNA damage from chemotherapy and creating an immunosuppressive microenvironment. It is therefore of clinical relevance to develop an effective delivery system that targets metabolic reprogramming and DNA damage response pathways for the treatment of drug-resistant TNBC. Herein, a P-gp-inhibiting and GSH-responsive multifunctional drug carrier targeting int
