Molecular dynamics analysis of a docking-derived baicalein-PPARγ ligand-binding-domain model suggests a pocket-associated AF-2-anchor-silent ensemble.

Baicalein was examined in a selected docking-derived peroxisome proliferator-activated receptor γ (PPARγ) ligand-binding-domain (LBD) model to test whether pocket association, local helix 12 (H12) organization, and canonical activation function-2 (AF-2) anchoring were coupled under the sampled simulation conditions. Because no baicalein-bound PPARγ cocrystal structure was used, the results are interpreted as a selected-pose molecular dynamics model rather than as a definitive experimental binding mode. Three independent 100-ns replicas were analyzed with bound-state filtering, H12 and AF-2 des

Stabilization of Ligand-Free Small Gold Nanocluster Within a Metal-Organic Framework for Enhanced Hydrogen Evolution and Horseradish Peroxidase-Mimicking Catalysis in Aqueous Media.

Atom-precise metal nanoclusters (NCs) have garnered considerable attention due to their photoluminescence and catalytic properties arising from the quantum confinement effect and metal-ligand interaction. However, the tendency of NCs to aggregate under reaction conditions hinders their catalytic activity. Surface-protecting ligands can enhance the stability, but they often reduce surface exposure and, consequently, catalytic activity. The incorporation of surface-cleaned clusters into the pores of metal-organic framework (MOF) is highly desirable. Here, we constructed a hybrid structure in whi…

Ligand-Dependent and -Independent Functions of Activation Function 1 of Progesterone Receptor in Genome-Wide Gene Regulation and in Cell Proliferation and Apoptosis of Breast Cancer Cells.

Progesterone receptor (PR) regulates gene expression through recruiting coregulators and general transcription factors by activation functions AF1 and AF2. AF1 localizes to the non-conserved and disordered N-terminal domain and is believed to facilitate tissue- and gene-specific activity. Our previous proteomic analysis identified three key residues (K464, K481 and R492) in AF1 that are monomethylated. Methylation mimic mutations KKR → FFF created hypoactive PR, whereas the KKR → QQQ mutation generated hyperactive PR in gene reporter assays. The current study used these mutants to determine th

Dual-Ligand Synergistic Construction of Double-Walled Cage Zn-MOFs: Methyl Modification Regulates Pore Size and Selective C2H2/CO2 Separation Performance.

Due to the extremely similar molecular sizes and melting and boiling points of acetylene (C2H2) and carbon dioxide (CO2), their efficient separation and purification have long been significant challenges in the field of industrial purification. Based on the ligand construction strategy, this study selected 2,4,6-tris(4-carboxyphenyl)-1,3,5-triazine as the main ligand and coordinated it with 3-methyl-1H-1,2,4-triazole and 3,5-dimethyl-1H-1,2,4-triazole, respectively, to assemble metal-organic frameworks (MOFs), successfully preparing two double-walled MOF crystal materials: Zn-TATB-fmtrz and Zn

Ligand-based design and synthesis of novel 3-spiro-2-oxindoline derivatives as potent VEGFR-2 inhibitors with antiproliferative activity and cell cycle arrest.

Rational design, synthesis, and biological evaluation of a novel series of 3-spiro-2-oxindoline derivatives as potential VEGFR-2 inhibitors were performed. Guided by the pharmacophoric requirements of type II VEGFR-2 inhibitors, the compounds were designed to incorporate key structural features required for VEGFR-2 recognition, with particular emphasis on aromatic linker integration to optimize binding interactions within the kinase active site. The synthesized derivatives were fully characterized and evaluated for their antiproliferative activity against HepG2 and HCT-116 cancer cell lines. S

HATP ligand-engineered Co-MOF-74 synthesized by dielectric barrier discharge microplasma and its application in electrochemical sensing of p-benzoquinone in water samples.

p-Benzoquinone (PBQ), as a key toxic derivative of benzene, is widely present in industrial wastewater and the production and manufacturing of daily chemicals. Long-term exposure poses a serious health threat, making the development of rapid and sensitive on-site detection technologies highly significant. Electrochemical methods, due to their simple operation, high sensitivity, and ease of miniaturization, are an ideal approach for detecting PBQ. Co-MOF-74 is extensively used in the electrochemical field because of its structural stability, tunable metal nodes, and large surface area, but its

Ligand-Defined Interfacial Chemistry Enables Instant and Sequence-General DNA Chemisorption on Gold Nanoparticles Toward Label-Free SERS With Single-Base Resolution.

A coherent mechanistic framework for DNA adsorption on gold nanoparticles (AuNPs) remains elusive, hindering the rational construction of spherical nucleic acids (SNAs). Here, we show that ligand-defined interfacial charge regulation, achieved using weakly ionized ascorbic acid (AA) ligands, markedly lowers the kinetic barrier for DNA chemisorption on AuNPs. Under near-neutral conditions, this interface enables rapid, sequence-general functionalization of AuNPs with both thiolated and non-thiolated oligonucleotides through simple vortex mixing, even for DNA containing only a single terminal ad…

Ligand-modulated Fe(III)/Fe(II) cycling enables dual sulfide control via accelerating abiotic oxidation and ROS-mediated microbial suppression.

Sulfide (S(-II)) accumulation in engineered systems poses persistent risks, yet its in situ oxidation by O2 is severely hindered by intrinsically sluggish kinetics and is continuously counteracted by microbial sulfur reduction. Here, we demonstrate the application of Fe(III)-ligand (L) complexes as a novel dual-functional strategy for S(-II) control, which involves (i) accelerating abiotic oxidation and (ii) suppressing microbial S(-II) regeneration via boosting reactive oxygen species (ROS) production. By identifying a "thermodynamic-kinetic framework", we decipher how ligands modulate Fe(III…

C-X-C Motif Chemokine Ligand 5 (CXCL5) Exhibits a U-Shaped Risk Profile for Mortality in Patients with Suspected Coronary Chest Pain.

CXCL5 is a platelet-derived chemokine which promotes inflammatory responses in neutrophils and monocytes through CXCR2. Previous studies on CXCL5 in atherogenesis are to some degree conflicting, with scarce outcome data following acute coronary syndrome. This study aimed to assess the utility and risk profile of CXCL5 as a prognostic marker of all-cause mortality at 5-year follow-up in patients hospitalized for chest pain of suspected coronary origin. We measured CXCL5 levels in platelet-poor plasma in 826 consecutive patients included in the "Risk Markers in the Acute Coronary Syndrome" (RACS

Anti-Receptor Activator of Nuclear Factor-κB Ligand Improves Muscle Dysfunction and Strengthens Bone in Laminin-α2-Deficient dy2J/dy2J Dystrophic Mice.

Congenital muscular dystrophy type 1A (CMD1A) is a severe genetic neuromuscular disorder caused by mutations in the LAMA2 gene, which encodes the laminin-α2 subunit, an essential structural protein for maintaining muscle integrity during contraction. Affected children experience progressive muscle damage, hypotonia, delayed motor development, and respiratory tract insufficiency. CMD1A muscles undergo repeated cycles of degeneration and regeneration accompanied by chronic inflammation. Beyond its established role in bone remodeling, the receptor activator of nuclear factor-κB/receptor activator

A novel ligand for chicken Toll-like receptor 5 from Eimeria maxima: dihydrolipoyl dehydrogenase drives M2-like polarization and Treg-like responses.

Avian coccidiosis is a major parasitic disease that poses a significant threat to the poultry industry. Elucidating the regulatory mechanisms of key molecules involved in the interplay between Eimeria and the host immune system is of great importance for the development of novel control strategies. Toll-like receptor 5 (TLR5) is a critical pattern recognition receptor in host innate immunity; however, research on its ligands has primarily focused on bacterial-derived molecules, and Eimeria-derived ligands along with their regulatory mechanisms remain largely uncharacterized. Using immunoprecip…

Improving ratiometric-based analysis via designing intense bi-ligand metal-organic frameworks: dual-mode for detection of amoxicillin in milk and pharmaceutical samples.

The development of dual-emission metal-organic frameworks (MOFs) is essential for advancing luminescence-based applications. In this study, a highly fluorescent bi-ligand Eu-based MOF (BiL-Eu-MOF) was synthesized exploiting coordination-induced emission (CIE) and dual-antenna effect phenomena. Upon excitation, the BiL-Eu-MOF emits both blue (CIE effect) and intense red (dual-antenna effect) fluorescence, enabling ratiometric detection. Amoxicillin (AMX) selectively quenches the red emission via the inner filter effect, while the blue emission remains stable, serving as an internal reference. T

Unravelling the link between mismatch repair protein deficiency and immune checkpoint markers - programmed death ligand 1 and galectin-9 expression in malignant melanoma.

BACKGROUND: The diagnostics and treatment of multiple myeloma (MM) requires precise analysis of serum immunoglobulins, which might be limited by the sensitivity of standard examination methods. Hevylite method enables quantitative analysis of heavy/light chain pairs (HLC) of normal and tumor IgG and IgA immunoglobulin and their ratio (HLC-r). The aim of the study was to assess the contribution of Hevylite method in the diagnostics of MM in comparison with nephelometry (NEF), standard protein electrophoresis (SPE), immunofixation electrophoresis (IFE) and the examination of serum free light cha

Discovery of Pyrazole-Containing 4‑Arylindolines as Potent Small-Molecule Inhibitors of the Programmed Cell Death-1/Programmed Cell Death-Ligand 1 Interaction.

The development of small-molecule inhibitors targeting the programmed cell death-1-(PD-1)/programmed cell death-ligand 1 (PD-L1) interaction has emerged as a promising strategy in tumor immunotherapy. In this study, a series of 4-arylindoline derivatives containing a pyrazole moiety were designed and synthesized through a medicinal chemistry campaign based on compound 1, a potent immunomodulator previously reported by our group. Among these, compound J29 was identified as the most promising inhibitor, exhibiting an IC50 value of 5.5 nM against the PD-1/PD-L1 interaction in a biochemical assay.

Unexpected bonding ambiguity of the open/closed-shell R2PO ligand family: the case of phosphinoyl vs. phosphoryl if bound to transition metals in their low oxidation state.

DFT pre-exploration of a broad set of heterocyclic compounds, focusing on 1,3,2-dihetero-phospholanes/phospholidines and open-shell R2P-O iron complexes, reveals a square pyramidal coordination geometry of the resulting iron(+I) centre, in stark contrast to the trigonal bipyramidal structure of the precursor iron(0) complexes. The latter clearly indicates an intramolecular redox process, initiated by a homolytic N-O bond cleavage transforming a phosphanoxyl ligand into a phosphoryl ligand, but not into a stable phosphinoyl ligand. The reaction cascade and the influence of co-ligands on the spi…
