Antioxidant lipid nanoparticles enhance mRNA stability for regeneration therapy and gene editing.

The success of mRNA vaccines has motivated the development of mRNA therapeutics to treat a wide range of diseases, as well as applications in regenerative medicine. However, the inherent instability of mRNA under conditions of oxidative stress renders these applications challenging. Here we report a general approach to shield mRNA from oxidative degradation by incorporating antioxidant lipid within lipid nanoparticles (LNPs). Specifically, we design a library of antioxidant ionizable lipids and identify a lead LNP formulation containing a 4-hydroxyphenyl-modified antioxidant lipid. Mechanistic…

Lipid-based systems for colorectal cancer: advances in early diagnosis and therapeutic efficacy.

Cancer is a leading cause of death worldwide. Particularly, colorectal cancer (CRC) is one of the most prevalent and deadly cancers in both men and women. The symptoms of this malignancy appear only at advanced stages; when detected, CRC has already spread to distant organs. Although several therapies are available, treatment is often not effective at advanced stages, where the estimated 5-year survival rate is about 10 %. For this reason, the investigation of new and innovative therapies for CRC management is of utmost importance. The association of chemotherapeutic drugs with delivery system

Extrahepatic Gene Editing In Vivo Using Organic Solvent-Free Lipid Nanoparticles.

Targeted therapy, which modifies genes and their expression, holds great promise for treating a variety of diseases, including cancer, inborn errors of metabolism, and acute and chronic inflammatory and infectious conditions. However, it also presents challenges related to RNA delivery, immune responses, side effects of delivery vectors, and the need for individualized formulations. To overcome these limitations, the choice of lipids and formulation processes might be re-evaluated, with a focus on eliminating critical components, such as poly(ethylene glycol) (PEG) and ethanol. Thus, a purely …

In Vitro and In Vivo Assessment of Darolutamide Encapsulated Lipid-Extruded PEGylated Liposomal Formulation.

Darolutamide (DA), a non-steroidal androgen receptor inhibitor, has emerged as a promising therapeutic agent for non-metastatic castration-resistant prostate cancer (nmCRPC); however, its clinical utility is limited by poor aqueous solubility and the need for high therapeutic doses. To address these challenges, PEGylated liposomes encapsulating darolutamide (DALP) were developed to improve its pharmacokinetic profile and enhance anticancer efficacy through sustained drug release. The optimized DALP exhibited a mean particle size of 107.4 ± 5.2 nm, a polydispersity index of 0.1505 ± 0.02, and a

Loss of cardiomyocyte AKT signaling causes deterioration of lipid metabolism and cellular atrophy.

The mammalian heart critically depends on oxidative metabolism of fatty acids, glucose, ketones, and amino acids to meet its extensive ATP demands. AKT/protein kinase B plays a central role in regulating cell growth and metabolism by coordinating an anabolic metabolism in response to insulin or IGF1, particularly by elevating glucose uptake and mTOR activity. Here, we investigated the effect of simultaneous deletion of the two major cardiac isoforms AKT1 and AKT2 on the function and metabolism of the adult mouse heart. Inducible cardiomyocyte specific AKT1 AKT2 double knockout mice developed a

Novel lipid-lowering therapies versus statins in primary prevention of cardiovascular disease.

BACKGROUND: Scarce evidence exists comparing clinical outcomes of novel lipid-lowering monotherapy with standard statin therapy for primary prevention. We compared clinical outcomes among primary prevention patients treated with novel lipid-lowering monotherapy versus statin monotherapy. METHODS: We performed a retrospective matched cohort study using the TriNetX network comprising 107 predominantly U.S. healthcare organizations. Adults with LDL-C > 100 mg/dL, between 2015 and 2024, and no prior ischemic heart disease, heart failure, cerebral infarction, or peripheral vascular disease were in

Endogenous targeting lipid nanoparticles for systemic mRNA delivery to lung cancer tumors.

Systemic delivery of mRNA therapeutics to lung cancer tumors remains challenging despite recent advances in organ-targeting lipid nanoparticles (LNPs). This study introduces novel LNPs, comprising commercially available DC-cholesterol (a cationic derivative of cholesterol) and 113-O12B ionizable lipid (DC_113), which provide selective mRNA translation in healthy lungs while achieving preferential protein expression in pulmonary tumors compared to adjacent lung tissue in an orthotopic cancer model. Analysis of LNPs with diverse ionizable lipids revealed that DC_113 has an apparent pKa above 7.9…

Aberrant protein palmitoylation promotes hepatic lipid accumulation and injury in dairy cows.

Elevated circulating nonesterified fatty acids (NEFA) represent a key pathological feature in dairy cows with fatty liver. Palmitic acid (PA), a major component of NEFA, can be enzymatically attached to proteins via a reversible post-translational modification known as palmitoylation, which potently modulates protein activity and function. Studies have revealed that aberrant hepatic palmitoylation is a crucial mechanism promoting lipid accumulation in non-ruminants. Nevertheless, the extent and pathological relevance of hepatic protein palmitoylation in dairy cows with fatty liver have largely

Targeting lipid metabolism enzymes in breast cancer: Mechanistic basis and therapeutic vulnerabilities.

Breast cancer constitutes a metabolically heterogeneous disorder wherein enzymes associated with lipid metabolism are instrumental in tumor progression, adaptation to the microenvironment, and therapeutic response. Beyond their catalytic roles in fatty acid, cholesterol, and phospholipid metabolism, these enzymes may function as regulatory nodes linking lipid flux to membrane remodeling, lipid peroxide detoxification, autophagy-dependent recycling, epithelial-mesenchymal plasticity, tumor-macrophage crosstalk, epigenetic stabilization, and multidrug resistance (MDR). This review adopts a subty

Combination Lipid-Lowering Therapy After ACS: Should this be the New Standard of Care?

PURPOSE OF REVIEW: To examine whether early combination lipid-lowering therapy after acute coronary syndrome (ACS) should be considered as a standard approach. The review assesses current international guideline recommendations, supporting clinical evidence, and real-world implementation gaps. RECENT FINDINGS: Recent European and U.S. guidelines emphasise lower low-density lipoprotein-cholesterol (LDL-C) targets and suggest earlier use of combination therapy. Data from randomised trials, registry-based emulations, and observational cohorts demonstrate that early initiation of high-intensity re

Update on familial hypercholesterolemia: An expert clinical consensus from the National Lipid Association.

Familial hypercholesterolemia (FH) is a common genetic disorder characterized by lifelong elevated low-density lipoprotein cholesterol (LDL-C), leading to a high risk of early onset atherosclerotic cardiovascular disease (ASCVD). This document provides an update to the National Lipid Association's 2011 clinical guidance, summarizing the remarkable progress in the field. With a global prevalence of approximately 1 in 311, FH remains severely underdiagnosed. This guidance reviews current diagnostic criteria, including the expanding role of genetic testing to complement diagnosis and to facilitat

Lipid metabolism of hepatocyte-like cells supports intestinal tumor growth in Drosophila.

Tumors reprogram lipid metabolism in distant tissues to support their growth. In adult Drosophila, gut tumors secrete the PDGF/VEGF-like factor Pvf1, which activates the TORC1-Hnf4 pathway in hepatocyte-like oenocytes. This drives production of very long-chain fatty acids and wax esters essential for tracheal growth around the tumor. Blocking Hnf4 or the elongase mElo in oenocytes strongly suppresses tracheogenesis, tumor progression, and cachexia-like organ wasting, while extending host lifespan. The same pathway also controls tracheal development in healthy flies. Lipoprotein receptor LpR2 d

Lipid nanoparticle-based multi-scale systemic immune programming for cancer therapy.

The clinical reach of cancer immunotherapy is currently limited by off-target toxicity, physical barriers in solid tumors, and the complex manufacturing of personalized cell therapies. This review presents multi-scale systemic immune programming as a transformative approach, in which lipid nanoparticles (LNPs) function not merely as delivery vehicles but as programmable platforms that direct immune responses across biological scales. At the molecular and cellular levels, LNP design and intracellular delivery regulate mRNA stability, translation, and antigen presentation. At the tissue level, i

A Multifunctional Polymeric Modulator Targeting Autophagic Lipid Efflux for Efficient Atherosclerosis Therapy.

Impaired lipid metabolism and persistent accumulation of inflammatory macrophages represent major obstacles to effective anti-atherosclerotic (AS) therapy. To overcome the inherent limitations of conventional oral agents and nanocarrier-based systems, we developed a macrophage-targeted modulator by integrating curcumin (Cur) with gold nanoparticles (AuNPs). This construct was designed to concurrently correct dysregulated lipid homeostasis and suppress the expansion of pro-inflammatory macrophage populations. Specifically, protoporphyrin (Por) as a π-conjugated moiety was conjugated to polycapr

Lipid Droplet-Targeted Biomimetic Liposomes Potentiate Chemo-Ferroptosis Therapy in Leukemia.

Chemotherapy remains the primary treatment modality for leukemia, yet relapse frequently occurs due to the persistence of chemoresistant leukemia cells (LCs) within the bone marrow (BM). Ferroptosis-based therapies provide a promising strategy for eliminating these resistant cells. Here, we demonstrate that cytarabine chemotherapy promotes lipid droplet (LD) accumulation in BM-resident LCs, thereby conferring resistance to ferroptosis. Based on these findings, we developed a biomimetic liposome (REM@HLipo) co-encapsulating RSL3 (a ferroptosis inducer), elacytarabine (a cytarabine prodrug), and

Metabolic regulation of gut barrier integrity in IBD: protein, lipid, and carbohydrate pathways.

Dietary macronutrients-proteins, lipids, and carbohydrates-influence the pathogenesis of inflammatory bowel disease (IBD) through distinct but interconnected metabolic pathways that extend beyond their roles as energy substrates. This narrative review synthesizes current evidence on how each macronutrient class modulates gut microbiota composition, intestinal barrier integrity, and mucosal immune responses. In the protein domain, the study examines how amino acid metabolism, particularly the tryptophan-kynurenine-indole axis and the arginine-inducible nitric oxide synthase-arginase balance, se…

Optimized Protocols to Extract Total Transcripts and Proteins from Lipid-Rich Tissues.

BACKGROUND: Highly lipidic tissues (e.g., adipose tissue, brain, and liver) are challenging for transcript and protein extraction and for next-generation sequencing. Lipids can clog filters, columns, and pipettes; cause autofluorescence and quenching in imaging; and interfere with centrifugation-based separation. AIM: To identify the most suitable method for extracting total RNA for RT-qPCR and an alternative method for extracting total protein for quantification in mice fed a regular or high-fat diet.
