An Update on the Role of Androgens and Androgen Receptor in Triple-Negative Breast Cancer.

Androgen receptor (AR) signaling has emerged as a potential molecular target in triple-negative breast cancer (TNBC), a clinically aggressive and biologically heterogeneous subtype of breast cancer with limited targeted treatment options. Androgens, the main ligands of AR, have been reported to exert antiproliferative and anti-estrogenic effects in normal mammary epithelium; however, the role of AR signaling in TNBC remains controversial and appears to depend strongly on tumor molecular context. In certain experimental settings, elevated androgen levels have been associated with reduced tumor

In Silico Design and Evaluation of Diosmin Analogs for Targeting Peroxisome Proliferator-activated Receptor γ (PPAR-γ) Against Alzheimer's Disease.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressively declining cognitive abilities and memory impairment. This disease increasingly challenges the quality of life and health of the elderly population, underscoring the need for effective therapeutic strategies. The existing anti-AD medications are designed to improve symptoms but not to cure the disease. Novel drugs are urgently needed to target the specific mechanisms that mediate disease progression. Peroxisome proliferator-activated receptor gamma (PPAR-γ) is a potential target for the development of anti-A

A relapsed/refractory acute promyelocytic leukemia achieving complete response after chemotherapy with venetoclax.

RATIONALE: The vast majority of patients newly diagnosed with acute promyelocytic leukemia (APL) can achieve complete response (CR) through induction therapy that includes all-trans retinoic acid (ATRA), and subsequently achieve molecular response after consolidation therapy. Relapses are quite rare after achieving molecular response. Nonetheless, 5% to 10% of APL patients who receive ATRA plus anthracycline chemotherapy may experience relapse. Compared to newly diagnosed APL patients, the treatment of relapsed/refractory APL remains a challenge due to its rarity. This article reports a case o

Breast cancer molecular subtypes: from TNBC to QNBC.

Treatment protocols for breast cancer depend predominantly on receptor status with respect to estrogen (estrogen receptor alpha), progesterone (progesterone receptor) and human epidermal growth factor [human epidermal growth factor receptor 2 (HER2)]. The presence of one or more of these receptors suggests that a treatment targeting these pathways might be effective, while the absence of, or in the case of HER2, lack of overexpression of, all of these receptors, termed triple negative breast cancer (TNBC), indicates a need for the more toxic chemotherapy. In an effort to develop targeted thera

Targeting The Tumor Microenvironment in Lymphomas: Emerging Biological Insights and Therapeutic Strategies.

PURPOSE OF REVIEW: This review aims to discuss recent advances in elucidating the tumor microenvironment (TME) in B lymphomas and resultant novel therapeutic development. RECENT FINDINGS: While tumor morphology, immunophenotype, and molecular profile are established factors that predict outcome and guide therapy, the prognostic impact of infiltrating, non-tumor cells is now emerging. This is simultaneously facilitating the development of new therapies that target non-tumor cells. The tumor microenvironment (TME) is a complex ecosystem composed of infiltrating cells and byproducts, extracellul

LXRα activation and Raf inhibition trigger lethal lipotoxicity in liver cancer.

The success of molecular therapies targeting specific metabolic pathways in cancer is often limited by the plasticity and adaptability of metabolic networks. Here we show that pharmacologically induced lipotoxicity represents a promising therapeutic strategy for the treatment of hepatocellular carcinoma (HCC). LXRα-induced liponeogenesis and Raf-1 inhibition are synthetic lethal in HCC owing to a toxic accumulation of saturated fatty acids. Raf-1 was found to bind and activate SCD1, and conformation-changing DFG-out Raf inhibitors could disrupt this interaction, thereby blocking fatty acid des

New developments in the medical treatment of Cushing's syndrome.

Cushing's syndrome (CS) is a severe endocrine disorder characterized by chronic cortisol excess due to an ACTH-secreting pituitary adenoma, ectopic ACTH production, or a cortisol-producing adrenal neoplasia. Regardless of the underlying cause, untreated CS is associated with considerable morbidity and mortality. Surgery is the primary therapy for all causes of CS, but surgical failure and ineligibility of the patient to undergo surgery necessitate alternative treatment modalities. The role of medical therapy in CS has been limited because of lack of efficacy or intolerability. In recent years,

Amphicrine Carcinomas of the Pancreas Showing Neuroendocrine and Acinar Cell Differentiation: A Case Report and Literature Review.

This study clarifies the distinct clinicopathological and molecular features of amphicrine pancreatic carcinoma (AmpCA) through a case analysis and literature review. We present a case of AmpCA in a 73-year-old female, characterized by co-expression of acinar cell markers and neuroendocrine markers within individual tumor cells. Molecular profiling revealed high-risk mutations in KRAS G12V, TP53, and RB1, along with a high Ki-67 index (85%). Literature comparison confirms that AmpCA exhibits a unique bidirectional, single-cell differentiation pattern, fundamentally distinguishing it from the b

Optimized Retinal Cell Dissociation for Downstream Analysis of Cone Photoreceptors.

The isolation of viable and homogenous retinal cells is essential for downstream molecular analysis, such as single-cell sequencing and transcriptomics. However, the dissociation of retinal cells can be challenging due to the intricate nature of retinal tissue and the fragility of its cells. Furthermore, cone photoreceptors are highly susceptible to degeneration. We developed an optimized protocol for retinal cell dissociation that improves the quality of cones for downstream molecular analysis. Our protocol employs a papain enzymatic digestion, mechanical dissociation, and filtration to obtai

Molecular pathophysiology of priapism: emerging targets.

Priapism is an erectile disorder involving uncontrolled, prolonged penile erection without sexual purpose, which can lead to erectile dysfunction. Ischemic priapism, the most common of the variants, occurs with high prevalence in patients with sickle cell disease. Despite the potentially devastating complications of this condition, management of recurrent priapism episodes historically has commonly involved reactive treatments rather than preventative strategies. Recently, increasing elucidation of the complex molecular mechanisms underlying this disorder, principally involving dysregulation o

Integrated Network Pharmacology and Molecular Modeling Reveal Therapeutic Potential of Silybum marianum Compounds against NAFLD.

INTRODUCTION: Non-alcoholic fatty liver disease (NAFLD) is a global health burden lacking effective pharmacological treatments. Silybum marianum, a hepatoprotective herb, contains flavonolignans with potential therapeutic effects against NAFLD, yet its active components and mechanisms remain unclear. METHODS: Active compounds were screened using TCMSP and literature data based on oral bioavailability (OB ≥ 30%) and drug-likeness (DL ≥ 0.18). Putative targets were predicted via SwissTargetPrediction and intersected with NAFLD-related genes from GeneCards, OMIM, and DisGeNET. Key targets were i

Neuroprotective efficacy of Curcumae Rhizoma against ischemic stroke via attenuation of COX-2-mediated neuroinflammation: a systematic study predicting thrombin inhibition and highlighting alexandrin and hederagenin.

Ischemic stroke is a major cause of mortality globally, but single-target therapies often fail due to the complex "ischemic cascade". This study investigated the neuroprotective effects and multitarget mechanisms of Curcumae Rhizoma (CR) using an integrated network pharmacology approach and in vivo validation. The active compounds of CR were screened through ADME filtering, and their potential targets were identified using the TCMSP, TTD, and UniProt databases. Molecular docking was used to validate the structural binding stability between CR ligands and core targets. In vivo efficacy was eval

Single-cell transcriptome deciphers key targets of thrombopoietin receptor agonists and immune microenvironment characteristics of immune thrombocytopenia.

Thrombopoietin receptor agonists (TPO-RAs) represent a cornerstone in immune thrombocytopenia (ITP) management, yet their molecular mechanisms remain incompletely elucidated. This study systematically deciphered the key targets and signaling networks of four TPO-RAs (romiplostim, eltrombopag, avatrombopag, hetrombopag) in ITP pathogenesis. Network pharmacology was integrated with single-cell high-dimensional weighted gene co-expression network analysis (hdWGCNA) using bone marrow scRNA-seq data from ITP patients and healthy controls. Metacell-based co-expression modules to hematopoietic bone m
