Resilient nanostructured bioanalytic microneedle longitudinally monitors preclinical renal and hepatic drug clearance and dysfunction.

Wearable microneedle biosensors promise real-time molecular monitoring for precision medicine but are limited by low sensitivity and tissue abrasion. Overcoming these challenges, we recast electrode functionality not merely as a sensing substrate but as a mechanism for resilient, high signal-to-noise ratio (SNR) measurements in tissue. Our microneedle-based resilient nanostructured bioelectrode (RNB) is fabricated using a bilayer process that strengthens the electrode with a micrometer-thick gold adhesion layer and reduces fabrication-induced stress through controlled dealloying. The resulting

MGMT promoter methylation in glioblastoma: Molecular mechanisms, tumor microenvironment interactions, and therapeutic implications.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults and is associated with poor prognosis despite advances in combination therapy. Among the molecular biomarkers guiding therapeutic decision-making, O⁶-methylguanine-DNA methyltransferase (MGMT) promoter methylation has emerged as clinically robust predictors of treatment response and patient survival. MGMT encodes a DNA repair enzyme that removes cytotoxic O⁶-alkylguanine lesions generated by alkylating agents such as temozolomide (TMZ). Epigenetic silencing of MGMT through promoter CpG islands methylation decreases DNA rep

RNA therapeutics 2.0: Expanding the landscape from mRNA vaccines to splicing modulators and beyond.

RNA therapeutics have progressed into a disruptive drug class quickly, replacing a variety of primary experimental agents which included vaccines, antisense oligonucleotides (ASOs), small interfering RNAs (siRNAs), aptamers and RNA editing systems. First-generation modalities, demonstrated by fomivirsen and pegaptanib were limited by vulnerability to nuclease attack, inefficient delivery and immune stimulation were treated with clinical feasibility. Recent clinical achievements, including mRNA vaccinations against COVID-19, have been based on developments in backbone chemistry, nucleoside modi

Thrombosis at the crossroads of inflammation, coagulation, and vascular biology: Recent advances and emerging therapeutic strategies.

Thrombosis is a multifaceted pathological process involving intravascular clot formation that drives a range of cardiovascular and cerebrovascular diseases, including myocardial infarction, ischemic stroke, venous thromboembolism, and cancer-associated thrombosis. Despite major advances in anticoagulation and thrombolytic therapies, clinical outcomes remain limited by bleeding risk, incomplete clot dissolution, and variable patient response. Emerging evidence increasingly portrays thrombosis as a dynamic immunoinflammatory disorder, integrating coagulation, inflammation, and immune signaling i

Redefining Liver Transplantation Indications for Hepatic Malignancies in the Era of Precision Transplant Oncology: An Up-to-Date Narrative Review.

Background: Hepatic malignancies are a major global health burden, with rising incidence, high mortality, and frequent diagnosis at advanced or unresectable stages. Although surgical resection, locoregional therapies, and systemic treatments have improved outcomes, many patients remain ineligible for curative strategies because of tumor burden, anatomical constraints, or liver dysfunction. Liver transplantation (LT) has therefore evolved from a treatment limited to selected hepatocellular carcinoma (HCC) cases within strict morphological criteria to a broader oncologic option guided by tumor b

Cross-omics interpretable neural network for discovery of molecular markers in prostate cancer.

Determining molecular markers that mediate clinically aggressive phenotypes in prostate cancer is a significant challenge. While traditional linear models offer some interpretability, they often lack the precision needed for complex multi-omics data. Conversely, conventional deep learning methods provide robust predictions but typically remain opaque, hindering the identification of impactful molecular markers and biological mechanisms. To address this, we propose the Cross-omics Interpretable Neural Network (CINN), a biomimetic framework designed to predict prostate cancer states and identify

An intelligent DNAzyme theranostic probe for in situ FTO bioimaging and real-time therapeutic efficacy evaluation in living cells.

Real-time bioimaging of epigenetic dynamics in living cells is essential for precision oncology. However, it remains technically challenging due to the lack of theranostic probes capable of both sensitive detection and dynamic efficacy feedback. Here, we report an intelligent DNAzyme-based theranostic probe (GF@ZD) for in situ bioimaging of endogenous active FTO and therapeutic efficacy evaluation in living cells. GF@ZD is constructed by encapsulating glucose oxidase (GOx) and Fe0 within a pH-responsive zeolitic imidazolate framework-8 (ZIF-8), followed by surface conjugation of an m6A-locked

Specific PET Imaging for Precision Management of Lung Cancer: Advances, Clinical Translation and Future Directions.

Lung cancer continues to be the leading cause of cancer-related deaths worldwide, primarily due to persistent challenges in early detection and the limited effectiveness of precision medicine. Although low-dose computed tomography (CT) has been widely implemented for lung cancer screening and has contributed to a measurable reduction in disease-specific mortality, its diagnostic accuracy is limited by its inability to reliably distinguish benign from malignant pulmonary nodules. Furthermore, the clinical standard for metabolic imaging18F-fluorodeoxyglucose (18F-FDG) positron emission tomograp

Intelligent Diagnosis and Treatment of Psoriasis Research: A Bibliometric Analysis from 2005-01-01 to 2025-12-31.

BACKGROUND: Psoriasis is a chronic inflammatory disease induced by genetic, immune and environmental interactions. The central mechanism is the aberrant activation of the Th17/IL-23 pathway, which leads to uncontrolled proliferation of keratinocytes and systemic inflammation. Psoriasis affects about 1-3% of the global population, with 70-90% of patients suffering from moderate to severe itching of the skin, 50% of patients suffering from thickening or loss of nails, and even 30% of patients suffering from a combination of arthropathy, and 59.1% of patients suffering from a psychological burden

The skeletal connectome: From molecular circuitry to systemic network medicine.

Although classically viewed as a mechanical scaffold, the skeleton is increasingly recognized as a dynamic endocrine and sensory hub that integrates systemic biophysical and biochemical cues. In this review, we rethink skeletal physiology by proposing the 'skeletal connectome,' a network medicine framework that links circuit-level remodeling programs to multiorgan homeostasis. Structurally, we first distill the cellular logic of bone remodeling, emphasizing how osteoblasts, osteoclasts, and osteocytes act as plastic network nodes whose state transitions are constrained by topology, compartment
