Immune response effects of diverse vaccine antigen attachment ways based on the self-made nanoemulsion adjuvant in systemic MRSA infection.

Nanoemulsion adjuvants-based vaccines have potent induced immune responses against methicillin-resistant Staphylococcus aureus (MRSA) infection. However, the efficacies and immune responses of different antigen-attaching ways on self-made nanoemulsion adjuvants remain unknown. In this study, we designed three formulations of nanoemulsion adjuvants (encapsulation, mixture, and combination) to explore their immune response-enhancing effects and their underlying mechanism in a systemic infection model of MRSA. Our results showed that the three nanoemulsion-attachment ways formulated with a fusion

Hydrogen-rich water improves neurological functional recovery in experimental autoimmune encephalomyelitis mice.

Multiple sclerosis (MS) is a chronic autoimmune demyelinating disease of the central nervous system (CNS). The high costs, inconvenient administration, and side effects of current Food and Drug Administration (FDA)-approved drugs often lead to poor adherence to the long-term treatment of MS. Molecular hydrogen (H2) has been reported to exhibit anti-oxidant, anti-apoptotic, anti-inflammatory, anti-allergy, and anti-cancer effects. In the present study, we explored the prophylactic and therapeutic effects of hydrogen-rich water (HRW) on the progress of experimental autoimmune encephalomyelitis (

Toxoplasma gondii infection modulate systemic allergic immune response in BALB/c mice.

The increased prevalence of allergies in developed countries has been attributed to a reduced exposure to some microbes. In agreement with epidemiological studies, we previously showed that Toxoplasma gondii infection prevents allergic airway inflammation. The mechanisms would be related to the strong Th1 response induced by the parasite and to regulatory cell induction. Herein we further characterized whether T. gondii allergy modulation extents to a systemic level or if it is limited to the lung. Parasite infection before allergic sensitization resulted in a diminished Th2 cytokine response

Oral tolerance is inefficient in neonatal mice due to a physiological vitamin A deficiency.

Increased risk of allergy during early life indicates deficient immune regulation in this period of life. To date, the cause for inefficient neonatal immune regulation has never been elucidated. We aimed to define the ontogeny of oral tolerance and to identify necessary conditions specific for this stage of life. Ovalbumin (OVA) was administered orally to mice through breast milk and efficiency of systemic tolerance to OVA was assessed in adulthood using a model of allergic airway inflammation. Oral tolerance induction was fully efficient starting third week of life. Inefficiency in neonates w

Immune restoration diseases reflect diverse immunopathological mechanisms.

Up to one in four patients infected with human immunodeficiency virus type 1 and given antiretroviral therapy (ART) experiences inflammatory or cellular proliferative disease associated with a preexisting opportunistic infection, which may be subclinical. These immune restoration diseases (IRD) appear to result from the restoration of immunocompetence. IRD associated with intracellular pathogens are characterized by cellular immune responses and/or granulomatous inflammation. Mycobacterial and cryptococcal IRD are attributed to a pathological overproduction of Th1 cytokines. Clinicopathologica

Chronic brucellosis patients retain low frequency of CD4+ T-lymphocytes expressing CD25 and CD28 after Escherichia coli LPS stimulation of PHA-cultured PBMCs.

Chronic brucellosis patients display a defective Th1 response to PHA. We have previously shown that heat-killed B. abortus (HKBA) can downregulate the PHA-induced increase of CD4+/CD25+ and CD14+/CD80+ cells of brucellosis patients. In the present study, we investigate the effect of E. coli LPS, as a potent stimulant of monocytes and autologous T-lymphocytes, on the PHA-cultured PBMCs of the same groups of patients. Thirteen acute brucellosis (AB) patients, 22 chronic brucellosis (CB) patients, 11 "cured" subjects, and 15 healthy volunteers were studied. The percentage of CD4+/CD25+ and CD4+/C

Environmental influences on the development of the immune system: consequences for disease outcome.

Early T cell responses to external antigens and autoantigens are subject to a variety of regulatory mechanisms. A unifying link between the increase in both Th1-dependent autoimmune disease and Th2-linked atopic allergy would be a disturbed immune regulation involving T regulatory cells. There is a strong global correlation between childhood wheezing and diabetes. It is increasingly recognized that microbial colonization of the gastrointestinal tract, linked with lifestyle and/or geographic factors, may be important determinants of the heterogeneity in disease prevalence throughout the world.

A DNA vaccine encoding lumazine synthase from Brucella abortus induces protective immunity in BALB/c mice.

This study was conducted to evaluate the immunogenicity of the Brucella abortus lumazine synthase (BLS) gene cloned into the pcDNA3 plasmid, which is driven by the cytomegalovirus promoter. Injection of plasmid DNA carrying the BLS gene (pcDNA-BLS) into BALB/c mice elicited both humoral and cellular immune responses. Antibodies to the encoded BLS included immunoglobulin G1 (IgG1) IgG2a, IgG2b, IgG3, and IgM isotypes. Animals injected with pcDNA-BLS exhibited a dominance of IgG2a over IgG1. In addition, spleen cells from vaccinated animals produced interleukin-2 and gamma interferon but not IL-

Cytokine responses in Trypanosoma cruzi-infected children in Paraguay.

Both parasite and host immune factors may contribute to the development and progression of chronic chagasic cardiomyopathy during Trypanosoma cruzi infections. The present study targeted infected children (5-14 years of age) from an endemic area of Paraguay in an analysis of T. cruzi-specific cytokine profiles. This age group is characteristically the most affected by the early phases of infection. Trypanosoma cruzi-induced cytokine gene expression (interleukin-2 [IL-2], and interferon-gamma [IFN-gamma], IL-4, and IL-10) was studied in 25 seropositive children categorized as being either acute

Water-Solubilized Curcuminoids Suppress Influenza A Virus Replication and Ameliorate Virus-Induced T-Cell Immune Dysfunction and Inflammatory Responses.

Influenza A virus (IAV) remains a major global health threat despite available vaccines and antiviral agents, while current therapies are limited by drug resistance and safety concerns. Curcuminoids exhibit antiviral and anti-inflammatory activities but are constrained by poor water solubility and low bioavailability. To address these limitations, we investigated the antiviral and immunomodulatory properties of a water-solubilized curcuminoid nanoparticle formulation (C-S/M) in both in vitro and in vivo models of IAV infection. To evaluate the potential antiviral and anti-inflammatory effects

Induction of Autoimmune Myocarditis in Diversity Outbred Mice.

BACKGROUND: Inbred mouse models of autoimmune myocarditis are routinely used to investigate the immune mechanisms underlying dilated cardiomyopathy. However, their translational relevance is limited because observations made in a single inbred strain may not reflect those of outbred human populations. This limitation can be overcome by using Diversity Outbred (DO) mice, whose genetic variability is comparable to that of humans. METHODS: To investigate the utility of DO mice, we characterized their immune cell distributions and induced myocarditis by immunization with porcine cardiac myosin (P

Fucosyltransferase 4-derived peptide bioconjugates on carbon nanotubes enhance antitumor immunity in an ovarian cancer mouse model.

Ovarian cancer is characterized by a profoundly immunosuppressive tumor microenvironment, which has limited the clinical success of immunotherapy approaches. Antigen-based nanovaccines represent a promising strategy to enhance antitumor immunity by improving antigen delivery and immune activation. In this study, we evaluated carbon nanotubes (CNTs) bioconjugated with a peptide derived from tumor-associated antigen fucosyltransferase 4 (FUT4) as an immunomodulating platform in an immunocompetent ovarian cancer mouse model. FUT4 expression was confirmed in the ID8-Defb29/Vegf-a (ID8DVLuc) ovaria

Refined single-cell profiling captures a CCR5high CD4+ cytotoxic T-cell precursor in multiple sclerosis.

BACKGROUND: Decades of immunological and genetic studies highlight CD4+ T cells as initiators of central nervous system (CNS) pathology in individuals who develop multiple sclerosis (MS). A low-frequency CD4+ T-cell population termed T helper 17.1 (Th17.1) has been argued to play a central role during MS onset due to differences in abundance, yet little is known about their functional heterogeneity and distinctive mode of action in MS. METHODS: Features that distinguished Th17.1 from Th17 and Th1 cells in the blood were explored using spectral flow cytometry. We focused on the brain-homing pr…

24-Nor-ursodeoxycholic acid improves intestinal inflammation by targeting TH17 pathogenicity and transdifferentiation.

BACKGROUND: 24-Nor-ursodeoxycholic acid (NorUDCA) is a novel therapeutic bile acid for treating immune-mediated cholestatic liver diseases, such as primary sclerosing cholangitis (PSC). OBJECTIVE: Since PSC strongly associates with T helper-type-like 17 (TH17)-mediated intestinal inflammation, we explored NorUDCA's immunomodulatory potential on TH17 cells. DESIGN: NorUDCA's impact on TH17 differentiation was assessed using a CD4+TNaive adoptive transfer mouse model, and on intraepithelial TH17 pathogenicity and transdifferentiation using an αCD3 stimulation model combined with interleukin-17

Biomimetic nanovaccines with self-adjuvant effects induced broad-spectrum neutralizing antibodies against SARS-CoV-2 infection in rodents.

The emergence of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) variants seriously threatens the efficacy of current coronavirus disease 2019 (COVID-19) vaccines. Therefore, there is an urgent need to develop next-generation vaccine platforms capable of counteracting current and prospective viral variants. In this study, we employed synthetic biology technology to develop self-adjuvant-effect biomimetic nanovaccines with broad-spectrum capabilities for the prevention of SARS-CoV-2 infection. The biomimetic nanovaccines prepared by loading the SARS-CoV-2 RBD protein into dendritic

When the victim becomes the villain: Platelets as drivers of immune dysregulation in ITP.

Immune thrombocytopenia (ITP) is a heterogeneous autoimmune disorder characterized by immune-mediated destruction of platelets and impaired platelet production. Although autoantibodies have historically been central to the understanding of ITP, current evidence demonstrates that its pathogenesis extends well beyond humoral mechanisms to involve complex dysregulation of both innate and adaptive immune responses. Multiple immune pathways-including autoreactive B and T cells, dendritic cell activation, and regulatory T cell deficiency-contribute to disease onset, progression, and chronicity. More

The altered T cell landscape in Systemic Sclerosis patients is characterized by dysfunctional type 1 immunity.

BACKGROUND: T cells in patients with chronic autoimmunity show features of dysfunction and exhaustion. However, the functionally-defined T cell subsets affected by these changes remain poorly characterized. Here, we sought to reveal aberrations in the composition, phenotype and function of canonical T cell subsets in the blood of Systemic Sclerosis (SSc) patients, compared to healthy subjects and Systemic Lupus Erythematosus (SLE) patients. METHODS: We developed a novel multidimensional flow-cytometry panel to simultaneously detect lineage-defining transcription factors, co-inhibitory recepto
