Decoding the Gut-Liver Crosstalk: Microbial Metabolite- Driven AhR Signalling Networks in MASLD Pathogenesis.

Metabolic-dysfunction-associated steatotic liver disease (MASLD), one of the most common liver diseases worldwide, has become an important public health issue, particularly with the increasing prevalence of obesity and diabetes. The gut microbiota is critical to the overall health of the human body, and accumulating evidence indicates that gut microbial alterations and their derived metabolites not only contribute to the development of MASLD but also represent potentially modifiable therapeutic targets. This review proposes that microbiota-derived metabolic signals regulate the aryl hydrocarbo

The Microbiome in Glioblastoma: Mechanisms, Tumor-Immune Interactions, and Translational Perspectives.

Despite multimodal treatment, glioblastoma (GBM) remains difficult to control because of diffuse invasion, tumor heterogeneity, immune dysfunction, and frequent recurrence. Increasing attention has focused on whether intestinal microbial communities and their products can modify systemic and central nervous system processes relevant to GBM. This narrative review critically evaluates current evidence linking microbial communities, microbial metabolites, tumor-associated microbial signals, and host immune responses with GBM biology and clinical translation. Aryl hydrocarbon receptor alleviates intestinal ischemia/reperfusion-induced epithelial barrier damage by inhibiting the 5-hydroxytryptamine-mediated complement C3 cascade.

Activation of the aryl hydrocarbon receptor (AhR) protects against intestinal epithelial barrier (IEB) dysfunction in multiple diseases, particularly during intestinal ischemia/reperfusion (II/R) injury. Studies have shown that AhR protects the epithelial barrier by regulating key functional molecules, however, the roles and mechanisms of AhR in intestinal epithelial cells (IECs) that maintain the IEB remain unclear. In this study, we found that Indole-3-carbinol (I3C) activated AhR, significantly ameliorating II/R-induced IEB damage and inflammatory responses. However, specific knockout of Ah

Amino acid metabolic reprogramming in immunotherapy for hepatocellular carcinoma: Adaptive resistance, patient stratification, and longitudinal monitoring.

BACKGROUND: Immunotherapy has become an important component of systemic treatment for advanced hepatocellular carcinoma (HCC), yet durable benefit remains limited to a subset of patients because of marked response heterogeneity and acquired resistance. PROBLEM: Current biomarkers are often static, single-time-point indicators that incompletely capture evolving tumor-immune metabolic interactions before and during therapy. In HCC, amino acid availability, nitrogen handling, and suppressive metabolite production are shaped by tumor-intrinsic programs, immune-cell competition, and liver-specific

Effects of gliadin-to-glutenin ratio on the thermal aggregation behavior of reconstituted wheat gluten.

Gluten proteins are important in determining the processing properties and textural characteristics of wheat-based products. Their thermal responsiveness directly influences the quality and stability of the final product. This study investigates the mechanism underlying changes in the rheological behavior of reconstituted gluten proteins during heat treatment. Various gliadin-to-glutenin (Gli:Glu) ratios were examined. Results demonstrated that the 2:8 ratio sample exhibited the highest storage modulus (G') and loss modulus (G″), together with relatively low loss tangent (tan δ) values, indica

Kynurenine promotes porcine intestinal epithelial cell proliferation by activating the AHR-MST1-YAP1 axis.

BACKGROUND: The objective of this study was to investigate the effect of L-tryptophan (L-Trp) and its metabolite kynurenine (Kyn) on the regulation of porcine intestinal epithelial cell proliferation. RESULTS: Dietary supplementation of L-Trp significantly increased villus height and decreased crypt depth in the jejunum and ileum of weaned pigs. mRNA sequencing data and qPCR analysis found that L-Trp activated the expression of cell proliferative genes and the AHR (aryl hydrocarbon receptor)-MST1 (mammalian STE20-like kinase 1)-YAP1 (Yes-associated protein 1) axis in the ileum. Antioxidant collapse and microbial metabolic shift underlie perfluorooctanoic acid toxicity in constructed wetlands.

With the increasing accumulation of emerging pollutants in environment, their associated ecological risks have drawn growing attention. Perfluorooctanoic acid (PFOA), a typical persistent pollutant, is widely distributed in wastewater treatment processes and lacks effective removal methods, posing potential threats to the ecological functions of constructed wetland (CW) systems. This study constructed four vertical subsurface flow CWs, which were continuously operated for 100 days under low (0.1 mg/L), medium (1 mg/L), and high (10 mg/L) PFOA concentrations to systematically investigate their

Income, diet, and cognitive function: observational analyses and candidate metabolomic pathways identified by Mendelian randomization.

BACKGROUND: Household income has been shown to have impact on cognitive function, with dietary patterns and gut microbiota-related metabolic pathways potentially acting as mediating pathways. Our study integrates observational analyses and Mendelian randomization (MR) to explore these associations. METHODS: The observational analysis included 13,457 participants from the UK Biobank who experienced cognitive transitions. A multistate Markov model was applied to assess the effect of income level on cognitive trajectory, while mediation analysis and quantile regression were performed using basel

A lung microbial signature for postoperative recurrence in stage I-II non-small cell lung cancer.

BACKGROUND: Postoperative recurrence remains a prevalent and lethal threat to survival in early-stage non-small cell lung cancer (NSCLC), affecting approximately 20% of patients within five years. While emerging evidence implicates the lung microbiota and microbiota-related metabolites in carcinogenesis, their prognostic value for predicting NSCLC recurrence remains underexplored. METHODS: Using preoperative bronchoalveolar lavage fluid from 72 patients with stage I–II NSCLC (17 recurrence and 55 non-recurrence), we performed microbial 2bRAD-M sequencing to profile bacterial, fungal, and archa

Prebiotic modulation of FMT donor microbiota enhances MASLD-relevant taxa and functions in an in vitro gut model.

AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD, formerly non-alcoholic fatty liver disease) is a prevalent and progressive condition closely linked to gut microbiota composition. Fecal microbiota transplantation (FMT) may help restore a health-associated microbiome, but its efficacy is often limited by inconsistent engraftment of beneficial taxa. Prebiotics may selectively support keystone microbes associated with reduced MASLD risk. This study evaluated two prebiotics, inulin and xylooligosaccharides (XOS), for their ability to modulate the microbiota of healthy FMT don

Engineered Melittin Delivers a Drug-Loaded 'Chemo-Sting' to Overcome Efflux-Mediated Multidrug Resistance in Cancer Cells.

Background: Multidrug efflux proteins, frequently overexpressed in cancer cells, reduce intracellular drug accumulation and limit chemotherapeutic efficacy. Short drug-binding peptides containing the WXXW motif have been shown to non-covalently bind multidrug resistance (MDR)-associated drugs, potentially masking structural features recognized by efflux transporters. We hypothesized that incorporating this motif into a membrane-active peptide would generate a hybrid analogue capable of reversible drug binding and enhanced cellular uptake. Methods: A melittin-derived peptide (M3) was rationally

Systemic toxicity of thiamethoxam in the mud crab (Scylla paramamosain): Oxidative stress, neurotoxicity, and growth retardation.

Thiamethoxam (TMX), a widely used neonicotinoid insecticide, poses a potential threat to non-target aquatic organisms. However, systematic toxicity evaluations on economically important crustaceans remain limited. This study comprehensively investigated the toxic effects and possible mechanisms of TMX on the mud crab Scylla paramamosain. Acute toxicity testing revealed a 96-h median lethal concentration (LC50) of 8.78 mg/L, suggesting moderate toxicity. Following a 21-day exposure to sublethal concentrations (8.8, 88, and 880 μg/L), TMX and its metabolite clothianidin accumulated in mud crabs

Characteristic of different thermal treatment-induced dough protein oxidation and its impact on human gut microbiota.

The thermal processing methods of flour products are diverse. This study aims to explore the effects of these methods (e.g., steaming, high-pressure, frying, baking, air frying, and microwave) on wheat dough protein oxidation and these oxidized proteins on human gut microbiota composition. We found that the different methods resulted in varying degrees of browning, decreased protein digestibility, and altered water distribution, protein noncovalent interactions, and surface hydrophobicity in the dough; the protein structure changed from α-helix and random coil to β-sheet and β-turn, and showed

Ruminal metabolism of serotonin metabolites in cattle.

Metabolism of serotonin (5-HT) by gut microorganisms has been demonstrated in several species. However, information regarding 5-HT metabolism by rumen microbiota remains limited. The objective of this study was to evaluate ruminal metabolism of 5-HT-related metabolites and subsequent changes in circulating concentrations in bovines following oral administration of 5-Hydroxytryptophan (5-HTP). In vitro incubation of rumen fluid with 5-HTP (100 µM), 5-HT (100 µM), or 5-Hydroxyindoleacetic acid (5-HIAA; 100 µM) were assessed to evaluate the degradation and metabolization of these compounds. Genistein suppresses renal fibrosis in chronic kidney disease through regulation of the CCAR2/SIRT1/p53 signaling axis.

Chronic kidney disease (CKD), a major global health burden, is characterized by progressive renal fibrosis, for which effective therapies are lacking. While the natural isoflavone genistein (Gen) has been documented to possess broad anti-fibrotic properties, its specific function and underlying mechanism in renal fibrosis remain largely undefined. In this study, we demonstrate that Gen administration significantly attenuates renal fibrosis in both UUO and adenine-induced murine CKD models. To elucidate the molecular mechanism, we employed a strategy using biotin-conjugated Gen as a bait for pu

Vine Tea (Ampelopsis grossedentata) Extract Mitigates High-Salt-Diet-Induced Hypertension by Remodeling the Gut Microbiota-Metabolite Axis in Mice.

Hypertension is a major global health challenge, with excessive dietary salt intake recognized as a key environmental factor contributing to its pathogenesis. However, safe and effective dietary interventions for salt-sensitive hypertension remain limited. Vine tea (Ampelopsis grossedentata), a traditional herbal tea widely consumed for centuries in southern China, has been reported to exhibit antioxidant, anti-inflammatory, and hepatoprotective activities, yet its antihypertensive efficacy and underlying mechanisms remain unclear. In this study, the chemical profile of vine tea aqueous extrac

Mechanisms of sludge reduction in Nostocoides-driven enhanced biological phosphorus removal process.

Nostocoides, a key phosphate-accumulating organism (PAOs), is abundant in wastewater treatment plants (WWTPs); however, the mechanism of sludge reduction in biological phosphorus removal systems under its dominance remains largely unknown. This study employed a lab-scale, exploratory multiparameter approach to examine extracellular polymeric substance (EPS) profiles, intracellular metabolites, microbial community, membrane integrity, and physiological activity to elucidate how Nostocoides-enriched enhanced biological phosphorus removal (EBPR) configurations accomplished sludge reduction. The metabolic effects of post-sepsis feeding of meals with only essential amino acids.

Nutritional support is a vital component of the rehabilitation process following sepsis. Essential amino acids (EAA) are recognized as a critical component for stimulating anabolism. However, it remains unclear whether EAA supplementation alone is more effective than providing a complete profile of total amino acids (TAA). We studied amino acid (AA) metabolism in 51 catheterized pigs (±27 kg) using the pulse tracer approach over a 7-day recovery period following sepsis. Animals were randomly and blindly assigned to receive post-sepsis nutrition containing either an EAA or a TAA mixture based o
