Synthesis and Inhibitory Activity of Machaeridiol-Based Novel Anti-MRSA and Anti-VRE Compounds and Their Profiling for Cancer-Related Signaling Pathways.

Three unique 5,6-seco-hexahydrodibenzopyrans (seco-HHDBP) machaeridiols A−C, reported previously from Machaerium Pers., have displayed potent activities against methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Enterococcus faecium, and E. faecalis (VRE). In order to enrich the pipeline of natural product-derived antimicrobial compounds, a series of novel machaeridiol-based analogs (1−17) were prepared by coupling stemofuran, pinosylvin, and resveratrol legends with monoterpene units R-(−)-α-phellandrene, (−)-p-mentha-2,8-diene-1-ol, and geraniol, and their inhibitory ac

Far-UVC light (222 nm) efficiently inactivates clinically significant antibiotic-resistant bacteria on diverse material surfaces.

UNLABELLED: In recent years, there has been a gradual increase in the prevalence of drug-resistant bacteria, primarily attributed to the widespread use of antibiotics. This has resulted in heightened mortality rates, morbidity, and exorbitant healthcare costs associated with antibiotic-resistant bacterial infections. In order to mitigate the spread of antibiotic-resistant bacteria, environmental disinfection plays a crucial role. Ultraviolet radiation C (UVC) light disinfection has emerged as a potent technique to limit the transmission of nosocomial pathogens and prevent healthcare-associated

Evaluation of Oxford Nanopore Technologies workflows for genomic epidemiology of outbreak-associated bacterial isolates in the clinical setting.

Accurate and efficient whole-genome sequencing (WGS) is crucial for clinical diagnostics and surveillance of bacterial infections. Here, we investigate the potential of a new Oxford Nanopore Technologies (ONT) workflow for WGS of clinically relevant bacterial isolates. Specifically, we assess the performance of R10.4.1 flow cells in combination with the V14 version of the transposase-based (RBK) library preparation kit to provide rapid and accurate genomic epidemiological comparisons of bacterial species of clinical importance. We focused on retrospective collections of outbreak-associated Cor

MRSA new treatments on the horizon: current status.

There is a choice of anti-MRSA antibiotic available with proven efficacy in the treatment of complicated skin and skin structure infection (cSSSI). Additional anti-MRSA antibiotics are in development, which have the potential to influence how such infections are managed. The emergence of resistance to current anti-MRSA agents, toxicity, and general lack of oral agents with proven efficacy for deep seated infection justify the development of new agents. However, there is a relative dearth of information specific to patients with orthopaedic-related infection. Combination therapy is often used i

Discovery of antibacterial and synergistic acylhydrazone functionalized α-mangostin derivatives against drug-resistant pathogens.

By strategically combining the acylhydrazone scaffold with α-mangostin (α-MG)'s membrane-targeting properties, we designed 48 novel conjugates to enhance antibacterial potential and biofilm penetration while reducing toxicity. The candidate compound 7-8 demonstrated exceptional anti-Methicillin-Resistant Staphylococcus aureus (MRSA) activity (MIC = 1 μg/mL) with in vivo efficacy (5 mg/kg, s.c.) comparable to vancomycin. 7-8 exhibited superior safety profiles versus α-MG, including minimal cytotoxicity (IC50 > 100 μg/mL in A549/LO-2 cells), negligible hemolysis (HC50 > 400 μg/mL), excellent in

Impact of growth conditions on the abundance and diversity of cultivable bacteria recovered from Pheronema carpenteri and investigation of their antimicrobial potential.

The deep sea is a largely unexplored extreme environment supporting a diverse biological community adapted to low temperatures and high pressures. Such environments support microbial life that may be a source of novel antibiotics and other drugs. Whilst this is often the case, many species with bioactive capabilities may be missed with traditional culturing methods. In this study, a total of 16 different concentrations and types of media were employed, to culture 389 bacterial isolates using Dilution to Extinction methods and Actinobacteria Directed Cultivation techniques. This generated 72 (1

Homotypic Membrane Vesicle-Formulated VAN@ΔagrMVs for Methicillin-Resistant Staphylococcus aureus Biofilm Clearance.

Multidrug-resistant (MDR) pathogens such as methicillin-resistant Staphylococcus aureus (MRSA) pose a substantial challenge to global public health, particularly because of chronic and persistent infections associated with bacterial biofilms, which call for safe and innovative therapeutic strategies. Here, we present a novel antibiofilm system inspired by the preferential uptake properties of isogenous bacterial membrane vesicles (MVs). This system employs vancomycin (VAN) for bacterial killing, while MVs act as delivery vehicles to increase VAN penetration into biofilms. VAN@ΔagrMVs demonstra

Euglycemic Diabetic Ketoacidosis Triggered by Sepsis in a Patient on Sodium-Glucose Co-transporter 2 (SGLT2) Inhibitor Therapy: A Case of Diagnostic and Therapeutic Challenges.

Euglycemic diabetic ketoacidosis (EDKA) is an uncommon life-threatening condition increasingly recognized in patients treated with sodium-glucose co-transporter 2 (SGLT2) inhibitors. Its diagnosis is often delayed due to the absence of significant hyperglycemia. Sepsis can further complicate the clinical scenario, enhancing insulin resistance and promoting ketoacidosis. We report the case of a 69-year-old male with a history of diabetes mellitus managed with insulin, semaglutide, and empagliflozin, who presented with syncope, bradycardia, and gastrointestinal symptoms following antibiotic trea

Novel Dry Hyaluronic Acid-Vancomycin Complex Powder for Inhalation, Useful in Pulmonary Infections Associated with Cystic Fibrosis.

Polyelectrolyte-drug complexes are interesting alternatives to improve unfavorable drug properties. Vancomycin (VAN) is an antimicrobial used in the treatment of methicillin-resistant Staphylococcus aureus pulmonary infections in patients with cystic fibrosis. It is generally administered intravenously with a high incidence of adverse side effects, which could be reduced by intrapulmonary administration. Currently, there are no commercially available inhalable formulations containing VAN. Thus, the present work focuses on the preparation and characterization of an ionic complex between hyaluro

Bifidobacteria metabolize lactulose to optimize gut metabolites and prevent systemic infection in patients with liver disease.

Progression of chronic liver disease is precipitated by hepatocyte loss, inflammation and fibrosis. This process results in the loss of critical hepatic functions, increasing morbidity and the risk of infection. Medical interventions that treat complications of hepatic failure, including antibiotic administration for systemic infections and lactulose treatment for hepatic encephalopathy, can impact gut microbiome composition and metabolite production. Here, using shotgun metagenomic sequencing and targeted metabolomic analyses on 847 faecal samples from 262 patients with acute or chronic liver
