Pancreatic acinar cells: ionic dependence of acetylcholine-induced membrane potential and resistance change.

1. Intracellular recordings of membrane potential, input resistance and time constant have been made in vitro from the exocrine acinar cells of the mouse pancreas using glass micro-electrodes. The acinar cells were stimulated by acetylcholine (ACh). In some cases ACh was simply directly added to the tissue superfusion bath, in other experiments ACh was applied locally to pancreatic acini by micro-iontophoresis. 2. Current-voltage relations were investigated by injecting rectangular de- or hyperpolarizing current pulses through the recording micro-electrode. Within a relatively wide range (-20 …

Sex-Dependent Alterations in Behavioral Development of Mice Lacking α5-Nicotinic Acetylcholine Receptors.

Complex neurodevelopmental disorders such as autism spectrum disorders (ASD) and schizophrenia are characterized by impaired social interactions, cognitive deficits, and repetitive behaviors. Genome-wide association studies (GWAS) have identified the α5 nicotinic acetylcholine receptor (α5-nAChR) subunit as a significant risk factor for these neuropsychiatric conditions. The α5 subunit is a critical modulator of neural circuit dynamics, specifically through its functional involvement within vasoactive intestinal peptide (VIP)-positive interneurons; however, its role in the sex-dependent matura…

Pharmaceutical stability of compounded acetylcholine chloride intraocular solution for intracoronary provocative vasospasm testing.

INTRODUCTION: Provocative coronary vasospasm testing requires intracoronary acetylcholine (ACh) administration, however an ACh drug formulation approved for intracoronary use is not commercially available. Invasive coronary vasomotor testing programs have thus resorted to reconstituting and diluting ophthalmic ACh solutions for off-label and off-target intracoronary use, despite a lack of comprehensive ACh stability testing data required by pharmacy compounding regulations USP <797 > . This study aims to characterize the stability of compounded ACh intraocular solutions according to current ph…

Two populations of heteromeric acetylcholine receptors are present at Caenorhabditis elegans neuromuscular junctions.

Ligand-gated acetylcholine receptors (AChRs) are pentameric transmembrane proteins that display extensive molecular diversity through combinatorial assembly of subunits. In the nematode Caenorhabditis elegans, this diversity is exceptionally expanded, with nearly 60 AChR subunit genes, yet the composition and function of many receptor subtypes remain poorly defined. At neuromuscular junctions (NMJs), L-AChRs are heteromeric AChRs sensitive to levamisole, a nematode-specific L-AChR agonist, that play a crucial role in locomotion. Forward genetic screens for mutants resistant to levamisole have …

Acetylcholine-protective activity of chemo-enzymatically synthesized aryl vicinal diols and hydroxy ketones.

Acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are the main enzymes responsible for the development and progression of Alzheimer's disease (AD). AChE and BChE hydrolyze acetylcholine (ACh), disrupting its management in neurotransmitters. In order to prevent and alleviate the symptoms of AD, the use of new compounds, chemo-enzymatically synthesized in environmentally friendly method from propenylbenzene derivatives as AChE and BChE inhibitors has been proposed. Molecular modeling and isothermal titration calorimetry were used to determine the AChE and BChE inhibition parameters. I…

Secoisolariciresinol diglucoside ameliorates muscarinic acetylcholine receptor mediated activation of NLRP3 inflammasome in cardiomyocytes.

BACKGROUND: Secoisolariciresinol diglucoside (SDG), the main lignan found in flaxseed, contributes to cardioprotection. However, its molecular mechanism of action remains unknown. NOD-like receptor protein containing domain 3 (NLRP3), a critical mediator in inflammatory pathways, is one of the key players in cardiovascular disease progression and is a promising target for cardioprotective interventions. This study investigated the immunomodulatory effect of SDG in regulating acetylcholine receptor-mediated activation of the NLRP3 inflammasome. METHODS AND RESULTS: HL-1 mouse cardiomyocytes we…

Sulfonium Moieties as Ammonium Bioisosteres: Novel Ligands for the Alpha7 Nicotinic Acetylcholine Receptor.

In the pressing quest of novel treatments for chronic pain, α7 nAChR silent agonists show efficacy as anti-inflammatory modulators and represent a promising strategy. Recent findings reveal that a sulfonium ion can replace the quaternary ammonium nitrogen as an alternative pharmacophore for nAChR silent activation. This study reports the design, synthesis, and electrophysiological evaluation of a new series of sulfonium-based derivatives inspired by the archetypal silent agonist NS6740. Our findings identify NSS-9 as a novel sulfonium α7 silent agonist that effectively alleviates inflammatory

Adiphenine Alleviates Impulsive-Like Behaviors in Aged Mice by Inhibiting Nicotinic Acetylcholine Receptors.

Postoperative neurocognitive disorders (PND) are prevalent complications in the elderly following surgical interventions and anesthesia, with the underlying protective and damaging mechanisms remaining largely elusive. Impulsive-like behaviors and a decline in cognitive function were observed in the mouse model of PND established through tibial fracture surgery, accompanied by increased activity of astrocytes in the hippocampal CA1 region. Additionally, transcriptome sequencing revealed increased expression of nicotinic acetylcholine receptors (nAChRs) in PND. In this study, we aimed to invest…

Rapid, objective intraoperative mapping of Hirschsprung disease with a portable electrochemical acetylcholine sensor.

PURPOSE: Surgical procedures for bowel aganglionosis require accurate identification of enteric ganglion cells and cholinergic nerve hypertrophy. Current intraoperative mapping through frozen section histopathology-histochemistry is time-consuming and demands skilled interpretation. This study explores an electrochemical sensor for objective, rapid intraoperative mapping of the aganglionic bowel segment via tissue acetylcholine (ACh) detection. METHODS: An electrochemical biosensor was developed using nanostructured electrodes functionalized with acetylcholinesterase(AChE) and choline oxidase…

[Acetylcholine Receptor Antibody Neutralization and Selective B-Cell Suppression Using Fc Fusion Protein].

Myasthenia gravis (MG) is an autoimmune disease characterized by formation of autoantibodies against the nicotinic acetylcholine receptor (AChR). Some patients do not show sufficient improvement and develop adverse effects following administration of conventional immune therapy; therefore, the development of new treatments is important. Based on the concept of "selective removal of pathogenic antibodies and cells without suppression of normal immunity," we are developing a fusion protein referred to as AChR-Fc (composed of the AChR alpha subunit and Fc region of human immunoglobulin G1), which…

Ocular versus generalized myasthenia gravis: a continuum associated with acetylcholine receptor antibody titers.

The aim of this study was to evaluate clinical and serological differences between the ocular myasthenia gravis (oMG) and generalized MG (gMG). This study is a retrospective chart review, in which data was collected from patients fulfilling 2 of 3 diagnostic MG criteria (positive antibodies, evidence of neuromuscular transmission defect on neurophysiological examination, positive effect of pyridostigmine treatment). 350 patients were included and data concerning demographics and MG medical history were collected. Patients with oMG accounted for 15.7 % of the included patients. The two subgroup

Ocular Myasthenia: Clinical Course and the Diagnostic Utility of Assaying Acetylcholine Receptor Antibodies.

Myasthenia gravis (MG) is an autoimmune disease that causes neuromuscular junction transmission defect and has a predilection for the with neuromuscular junction transmission defect and predilection for extra-ocular and eyelid muscles. Most cases of ocular MG (OMG) convert later to generalised MG (GMG). Assaying acetylcholine receptor antibodies (AchRA) has been used to diagnose MG, but the reported sensitivity in OMG is lower (50%) than in GMG. We report the clinical course and the diagnostic yield of assaying AchRA in a Kuwaiti cohort of patients with OMG. We carried out a retrospective revi…

Mice lacking α4 nicotinic acetylcholine receptors are protected against alcohol-associated liver injury.

BACKGROUND: Chronic heavy alcohol consumption is a major risk factor for the development of liver steatosis, fibrosis, and cirrhosis, but the mechanisms by which alcohol causes liver damage remain incompletely elucidated. This group has reported that α4 nicotinic acetylcholine receptors (α4 nAChRs) act as sensors for alcohol in lung cells. This study tested the hypothesis that α4 nAChRs mediate the effects of alcohol in the liver. METHODS: Expression of acetylcholine receptor subunits in mouse liver was determined by RNA sequencing (RNA-seq). α4 nAChR knockout (α4 KO) mice were generated in C…
