Impacts of impaired control and pharmacological criteria for lifetime alcohol use disorder on relationships between drinking and problems.

BACKGROUND: The contribution of criteria for lifetime alcohol use disorder (AUD) to greater drinking and problems is infrequently considered in community-based studies of alcohol users. However, theoretical and empirical work indicate that relationships between drinking and problems may be quite different among drinkers reporting impaired control (ic) or pharmacological (p) criteria related to AUD (icpAUD). In this study we measured the extent to which the presence of greater number of these criteria affected dose-response relationships between drinking and problems. METHODS: A survey of 2044

Early evolution of alcohol use and drinking motives after bariatric surgery and associated factors: a prospective cohort study.

BACKGROUND: Research indicates the prevalence of alcohol use disorder (AUD) increases following bariatric surgery. However, the evolution of alcohol drinking motives over time and the predictors of increased alcohol use postsurgery remain unclear. OBJECTIVES: This study examined changes in alcohol consumption and drinking motives during the first 6 months after bariatric surgery, as well as psychological factors associated with increased postoperative alcohol use. SETTING: The research was conducted across four Belgian hospitals. METHODS: A prospective cohort study was carried out. Of the 2…

Assessing Autonomic Regulation Under Stress with the Yale Pain Stress Test in Social Drinking and Alcohol Use Disorder.

High levels of stress and individual differences in acute stress responses are important predictors of chronic illness. This study examined the effects of stress and Alcohol Use Disorder (AUD) on heart rate variability (HRV) using the Yale Pain Stress Test (YPST), adapted from the well-established Cold Pressor Test (CPT). The YPST characterized three key (time periods) phases of the cardiovascular stress response to repeated trials of a pain-stress versus no pain-stress control condition: pre-stress baseline, acute reactivity, and recovery, using HRV as a physiological marker. Participants inc

Selective effects of oxytocin on alcohol drinking in subpopulations of male and female mice following intermittent predator stress.

AIMS: Excessive alcohol use is often triggered by traumatic experiences, when subjects consume alcohol-containing beverages as a passive coping mechanism to relieve negative affect. There are no FDA-approved medications that are specifically recommended for patients with alcohol use disorder who use alcohol to decrease the consequences of trauma. The current study used a mouse model of predator stress-enhanced drinking to test whether administration of oxytocin (OXT) could selectively target subjects with increased sensitivity to psychological trauma. METHODS: Male and female C57BL/6J mice es

Public Awareness of Risks Associated with Alcohol Drinking in the US: A Population-Based Cross-Sectional Survey Study.

INTRODUCTION: Alcohol consumption is a leading preventable cause of morbidity and mortality in the United States, contributing to over 178,000 deaths annually. Despite known links to liver disease and cancer, few adults report receiving counseling on alcohol-related risks from health care professionals. Authors of this cross-sectional, population-based study assessed the prevalence of alcohol-related counseling among United States adults and identified sociodemographic and behavioral factors associated with receiving such advice. METHODS: Data were drawn from the 2022 Health Information Natio

Sex differences in compulsive alcohol drinking phenotypes: implications for decision-making and social behavior in a preclinical model.

RATIONALE: Compulsivity is increasingly recognized as a transdiagnostic trait that amplifies vulnerability to alcohol use disorders. However, its specific role in shaping social behavior and decision-making remains underexplored. OBJECTIVE: This study aimed to identify a vulnerable phenotype characterized by compulsive alcohol drinking and evaluate its behavioral alterations within the social behavior and cognitive processes domains of the Research Domain Criteria (RDoC), considering sex as a modulatory factor. METHODS: Male and female Wistar rats were exposed to Schedule-Induced Polydipsia …

Sex-Specific Roles of Hypocretin Receptor Signaling in CRF Neurons on Alcohol Drinking, Anxiety, and BNST Neuronal Excitability.

Alcohol use disorder (AUD) is characterized by compulsive alcohol consumption and negative emotional states during withdrawal, often perpetu … ve to females. Moreover, the double deletion of both Hcrt receptors from CRF neurons led to reduced alcohol drinking in males and dampened anxiety behaviors and BNST excitability in both sexes during protracted withdrawal. These findings suggest that Hcrt signaling in CRF neurons plays a critical role in the persistence of excessive alcohol consumption and the dev …

Changes in US drinking and alcohol use disorders associated with social, health, and economic impacts of COVID-19.

AIMS: The COVID-19 pandemic increased alcohol consumption in the USA as a result of widespread individual changes in drinking patterns. Few studies have utilized longitudinal data allowing the prediction of increased or decreased drinking from COVID-19 economic, social, and health impacts. METHODS: Data are from 1819 respondents in the 2019-20 National Alcohol Survey and a one-year follow-up in early 2021. Changes in past-year alcohol volume, drinking days, days with 5+ drinks, and Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5) alcohol use disorder (AUD) severity

Sex differences in binge drinking-related higher morning cortisol levels and in prospective association with future alcohol intake.

AIMS: Peripheral cortisol represents one biological measure of the hypothalamic-pituitary-adrenal (HPA) axis, a significant component of the stress system, which is altered by chronic alcohol consumption. However, whether heavy alcohol use affects the HPA axis differentially between the sexes and whether basal cortisol levels are a biomarker of prospective alcohol intake is unknown. METHODS: We recruited light moderate (LM) and binge-heavy (BH) drinkers of alcohol (n = 118). Repeated fasting morning cortisol levels were studied over a 2-hour period to assess basal levels while participants un

Drinking and laboratory biomarkers, and nutritional status characterize the clinical presentation of early-stage alcohol-associated liver disease.

Chronic and heavy alcohol consumption is commonly observed in alcohol use disorder (AUD). AUD often leads to alcohol-associated organ injury, including alcohol-associated liver disease (ALD). Approximately 10-20% of patients with AUD progress to ALD. Progression of ALD from the development phase to more advanced states involve the interplay of several pathways, including nutritional alterations. Multiple pathologic processes have been identified in the progression and severity of ALD. However, there are major gaps in the characterization and understanding of the clinical presentation of early-

Intolerance of Uncertainty, Negative Reinforcement Alcohol Use Motives, and Hazardous Drinking in College Students with Clinically Elevated Worry.

Background: Hazardous drinking has remained high for college students in recent years, and individuals who endorse drinking to cope with emotional distress or to conform socially report higher levels of alcohol use. Intolerance of uncertainty, a core process underlying generalized anxiety disorder, has been linked to negative reinforcement drinking motives; however, no research, to date, has examined the role of intolerance of uncertainty in terms of alcohol use motives and hazardous drinking among individuals with generalized anxiety disorder. Objective: The current study was designed to exam

What Predicts Treatment Adherence and Low-Risk Drinking? An Exploratory Study of Internet Interventions for Alcohol Use Disorders.

INTRODUCTION: Internet interventions for alcohol problems are effective, but not all participants are helped. Further, the importance of adherence has often been neglected in research on internet interventions for alcohol problems. Prediction analysis can help in prospectively assessing participants' probability of success, and ideally, this information could be used to tailor internet interventions to individual needs. METHODS: Data were obtained from a randomized controlled trial on internet interventions for alcohol use disorders. Twenty-nine candidate predictors were run in univariate log

The Relationship Between Drinking Behavior and Conversational Processes During a Brief Alcohol Reduction Intervention for People with HIV.

People with HIV (PWH) frequently engage in unhealthy alcohol use, which can adversely affect antiretroviral adherence and HIV disease progression. Brief interventions based on Motivational Interviewing (MI), including the Brief Negotiated Interview (BNI), can help to reduce drinking. This study examines MI processes observed during a single 15-20 min BNI session delivered by social workers to PWH with unhealthy alcohol use (N = 59) in the context of a stepped care intervention to reduce alcohol consumption. BNI sessions were coded for technical and relational processes encouraged in MI, such a

Does insomnia mediate the link between childhood trauma and impaired control over drinking, alcohol use, and related problems?

INTRODUCTION: Hyperarousal theory states that stressful negative events can result in a physiological response in the body leading to poor sleep quality. Childhood trauma is associated with many negative health consequences persisting into adulthood such as insomnia. Insomnia itself is a driver of poor physical and psychological health including excessive alcohol use. We examined the direct and indirect relationships between trauma (i.e., physical-neglect, physical, emotional, and sexual abuse) as well as emotionally supportive families on insomnia, impaired control over drinking, alcohol use,

Intra-dorsolateral striatal AMPA receptor antagonism reduces binge-like alcohol drinking in male and female C57BL/6J mice.

The dorsolateral striatum (DLS) is involved in addiction, reward, and alcohol related behaviors. The DLS primarily receives excitatory inputs which are gated by post-synaptic AMPA receptors. We antagonized AMPA receptors in the DLS to investigate how such modulation affects binge-like alcohol drinking in male and female C57BL/6J mice and whether an associated alcohol drinking history alters dorsomedial striatum (DMS) and DLS AMPA receptor expression. We also investigated the effect of intra-DLS NBQX on locomotor activity and saccharin drinking in mice. Mice were allowed free access to 20% alco…

mTORC1 pathway is involved in the kappa opioid receptor activation-induced increase in excessive alcohol drinking in mice.

KOP-r agonist U50,488H produces strong aversion and anxiety/depression-like behaviors that enhance alcohol intake and promote alcohol seeking and relapse-like drinking in rodents. Mammalian target of rapamycin complex 1 (mTORC1) pathway in mouse striatum is highly involved in excessive alcohol intake and seeking, and in the U50,488H-induced conditioned place aversion. Therefore, we hypothesized that KOP-r activation increases alcohol consumption through the mTORC1 activation. This study focuses on: (1) how chronic excessive alcohol drinking (4-day drinking-in-the-dark paradigm followed by 3-we…

Effects of alcohol intoxication on self-reported drinking patterns, expectancies, motives and personality: a randomized controlled experimental study.

Alcohol intoxication may affect self-reports of alcohol use and related constructs, such as impulsivity and dependence symptoms. Improved knowledge about potential systematic reporting biases induced by alcohol, e.g. through disinhibition, may be relevant for the assessment of intoxicated individuals both in clinical routine and research. We therefore randomly assigned 54 socially drinking males aged 18 to 19 without lifetime diagnosis of DSM-IV alcohol dependence to one of two experimental arms: either placebo infusion at day 1 and alcohol infusion at day 2, or vice versa. The lab-based intra
