Co-Expression of CD34, CD90, OV-6 and Cell-Surface Vimentin Defines Cancer Stem Cells of Hepatoblastoma, Which Are Affected by Hsp90 Inhibitor 17-AAG.

Cancer stem cells (CSCs) are nowadays one of the major focuses in tumor research since this subpopulation was revealed to be a great obstacle for successful treatment. The identification of CSCs in pediatric solid tumors harbors major challenges because of the immature character of these tumors. Here, we present CD34, CD90, OV-6 and cell-surface vimentin (csVimentin) as reliable markers to identify CSCs in hepatoblastoma cell lines. We were able to identify CSC characteristics for the subset of CD34+CD90+OV-6+csVimentin+-co-expressing cells, such as pluripotency, self-renewal, increased expres

Phase II trial of co-administration of CD19- and CD20-targeted chimeric antigen receptor T cells for relapsed and refractory diffuse large B cell lymphoma.

PurposeAnti-CD19 chimeric antigen receptor T (CAR-T) cell therapy has demonstrated remarkable efficacy for refractory and relapsed diffuse large B cell lymphoma (R/R DLBCL). However, this therapy failed in nearly 25% patients mainly due to antigen loss. The authors performed a phase Ⅱ trial by coadministration of anti-CD19 and anti-CD20 CAR-T cells treatment for R/R DLBCL and evaluated its efficacy and toxicity.MethodsTotally 21 patients with DLBCL were enrolled in this study. The patients were conditioned with fludarabine and cyclophosphamide before the infusion of anti-CD19 and anti-CD20 CAR

Co-infusion of haplo-identical CD19-chimeric antigen receptor T cells and stem cells achieved full donor engraftment in refractory acute lymphoblastic leukemia.

BACKGROUND: Elderly patients with relapsed and refractory acute lymphoblastic leukemia (ALL) have poor prognosis. Autologous CD19 chimeric antigen receptor-modified T (CAR-T) cells have potentials to cure patients with B cell ALL; however, safety and efficacy of allogeneic CD19 CAR-T cells are still undetermined. CASE PRESENTATION: We treated a 71-year-old female with relapsed and refractory ALL who received co-infusion of haplo-identical donor-derived CD19-directed CAR-T cells and mobilized peripheral blood stem cells (PBSC) following induction chemotherapy. Undetectable minimal residual dis

Time dependent and cell-specific action of polychlorinated biphenyls (PCB 153 and PCB 126) on steroid secretion by porcine theca and granulosa cells in mono- and co-culture.

To characterize PCB action on follicular cell steroidogenesis two PCB congeners were selected as model substances. PCB 126 because of its dioxin-like configuration and high toxicity and PCB 153 because it is one of the most commonly detected PCB congeners in breast milk. The direct effect of PCBs was investigated using a culture system of porcine theca and granulosa cells collected from porcine preovulatory follicles. Granulosa and theca cells were cultured in M199 medium supplemented with 1, 10 or 100 pg/ml of PCB 126 or 1, 10 and 100 ng/ml of PCB 153. The media were changed after 48, 96 and

Two modes of homologous C3 deposition on Ramos Burkitt's lymphoma cell substrains co-expressing DAF (CD55), CD59, and CR2 (CD21), and on cells lacking them.

We established decay-accelerating factor (DAF)/CD59-positive and -negative substrains of a human B cell line, Ramos, R(DAF+/CD59+) and R(DAF-/CD59-) respectively. Unexpectedly, treatment of R(DAF+/CD59+) cells with Mg2(+)-EGTA-serum resulted in efficient C3 deposition, while treatment of R(DAF-/CD59-) cells did not. All six substrains of R(DAF-/CD59-) cells were CR2-negative, and treatment of the cells with M177 [a membrane cofactor protein (MCP) cofactor-blocking antibody] and/or acidic buffer only minimally affected the extent of C3 deposition. However, when R(DAF-/CD59-) cells were pretreat

The causal relationship between immune cells, white matter microstructure and dementia: a Mendelian randomization study and co-localization analysis.

With the advancement of global aging, the prevalence of dementia is on the rise. Peripheral immune dysregulation and white matter microstructural changes are potential inducing factors for dementia. However, it is not yet clear which specific types of immune cells and parameters of white matter microstructure have a causal relationship with dementia. This study used Mendelian randomization (MR) to assess causal relationships and mediating pathways among immune cell-related phenotypes, white matter microstructure-related phenotypes, and dementia-related phenotypes, and used co-localization anal

Co-delivery of a STING agonist and indoleamine 2,3-dioxygenase 1 blockade activates type I dendritic cells in cancer.

The stimulator of interferon genes (STING) has emerged as a promising target for cancer immunotherapy. However, STING agonists alone have demonstrated limited clinical success. Indoleamine 2,3-dioxygenase 1 (IDO1) has been implicated as a key resistance mechanism that undermines the antitumor effects of STING activation, yet the cell-type-specific roles and the combined impact of STING and IDO1 on tumor immune orchestration remain poorly understood. In this study, we report that a co-delivery nanosystem of STING agonists and IDO1 inhibitors, rather than their simple combination, yields synergi

Mannosylated graphene oxide nanotherapeutics co-delivering docetaxel and a STING agonist reprogram myeloid cells and potentiate antitumor immunity.

In triple-negative breast cancer (TNBC), tumor-associated macrophages (TAMs) and tumor-infiltrating dendritic cells (TIDCs) often exhibit an immunosuppressive phenotype. This myeloid-dominant milieu reinforces an immune-cold state, promotes metastasis and contributes to immunotherapy resistance. In this study, we developed a mannose-functionalized graphene oxide nanocarrier (GO-EDM) with favorable biocompatibility and high NIR photothermal conversion efficiency. Docetaxel and vadimezan (DMXAA), a prototypical STING agonist, were co-loaded into GO-EDM to generate GO-EDM-DTX-Vad. In vitro, the n

Astragaloside IV Co-Loaded Osimertinib Liposomes Alleviate EGFR-TKI Resistance in Non-Small Cell Lung Cancer.

INTRODUCTION: Advances in tumor biology have established EGFR-tyrosine kinase inhibitors (EGFR-TKIs) as a key therapy for non-small cell lung cancer (NSCLC). Osimertinib (OSI), a third-generation EGFR-TKI, however, often leads to acquired resistance within about a year. Astragaloside IV (AS-IV), a bioactive saponin from Astragalus membranaceus, exhibits anti-tumor potential and may help reverse resistance-related epithelial-mesenchymal transition (EMT), a process mediated by TGF-β. While combining OSI with AS-IV is a promising strategy, its efficacy is limited by poor solubility and toxicity-c

Activation of the unfolded protein response via PERK-ATF4 signalling in deoxynivalenol- and PRRSV-co-exposed MARC-145 cells.

IMPORTANCE: Deoxynivalenol (DON) is a prevalent mycotoxin that is highly toxic to animals, particularly pigs. Porcine reproductive and respiratory syndrome virus (PRRSV) is a major swine pathogen with high infectivity and severe clinical outcomes, but the mechanisms underlying the host cell responses to co-exposure of DON and PRRSV remain unclear. OBJECTIVE: This study examined the effects of the co-exposure of Marc-145 cells with DON and PRRSV. METHODS: A DON- and PRRSV-co-exposed MARC-145 cells model was established, and transcriptomic/proteomic sequencing combined with Western blot and im

Epithelial cancer cells co-opt the death ligand TRAIL to avert extrinsic apoptosis while acquiring killing capacity.

Cancer cells avert apoptosis to sustain growth, evade immunity, and resist therapy. However, how they escape specific extrinsic death signals remains unclear. We show that certain epithelial carcinomas co-opt the death-receptor ligand Apo2L/TRAIL to suppress apoptosis while acquiring killing capacity. Analysis of CRISPR knockout screens from the Cancer Dependency Map uniquely identified TRAIL as an essential tumor necrosis factor superfamily member in epithelial malignancies. Renal cell carcinomas (RCC) and squamous cell carcinomas (SCC) expressed TRAIL within both malignant and nonmalignant c

Multiscale design of core-shell GelMA-alginate composite microspheres enabling spatially compartmentalized co-culture of skin cells.

Biomaterial-based hydrogels are widely used in skin regeneration as wound dressings and cell delivery vehicles owing to their biocompatibility and ability to mimic native skin microenvironments. However, most strategies rely on encapsulating a single cell type within homogeneous hydrogel matrices, which fail to recapitulate the fibroblast-keratinocyte crosstalk essential for wound healing. This limitation arises from the absence of spatial compartmentalization required to balance the disparate proliferation kinetics and functional roles of dermal fibroblasts and epidermal keratinocytes. Theref

FUS-pen: focused ultrasound-mediated micro-sculpting of label-free cells to create functional co-culture microenvironments.

Precise spatial organization of cells and microparticles is essential for engineering physiologically relevant tissues and in vitro disease models, yet current bioink-based bioprinting approaches still face limitations regarding cell density dilution and crosslinking-induced cellular stress. Here we introduce the FUS-pen system, a template-free, focused ultrasound platform that enables non-contact, flexible and cross-scale manipulation of microparticles and cells with demonstrated biocompatibility. Mechanistic investigations reveal a cooperative interaction where acoustic streaming drives long

Tuning HDAC Inhibition, Cell Uptake, and Antiproliferative Activity of Vorinostat-Derived Half-Sandwich Complexes with Phosphine Co-Ligands.

HDAC inhibitors are used as anticancer drugs, and the Zn2+-coordinating hydroxamic acid moiety in the drug vorinostat (SAHA) plays a key role in the mode of action. The introduction of metal centers in the structures of HDAC inhibitors (HDACis) allows for additional interactions between the inhibitor and the protein. To alter the interactions between HDACs and half-sandwich organometallic HDACis, we introduced amphiphilic 1,3,5-triaza-7-phosphaadamantane (PTA) or lipophilic triphenylphosphine (PPh3) as coligands. The half-sandwich compounds were found to be stable in aqueous solution, and the

Genotype-to-Phenotype Mapping of Somatic Clonal Mosaicism via Single-Cell Co-Capture of DNA Mutations and mRNA Transcripts.

UNLABELLED: Somatic mosaicism is pervasively observed in human aging, with clonal expansions of cells harboring mutations in recurrently mutated driver genes. Bulk sequencing of tissues captures mutation frequencies but cannot reconstruct clonal architectures nor delineate how driver mutations affect cellular phenotypes. We developed single-cell genotype-to-phenotype sequencing (scG2P) for high-throughput, highly multiplexed, joint capture of genotyping of mutation hotspots and mRNA markers. We applied scG2P to aged esophagus samples from six individuals and observed large numbers of clones wi

A Scalable Lentiviral Workflow for Laboratory-Scale Generation of BCMA/GPRC5D Co-Transduced CAR-T Cells in Multiple Myeloma.

Efficient and reproducible lentiviral vector production and T-cell transduction remain important technical challenges in CAR-T (Chimeric Antigen Receptor T-cell) cell manufacturing. In this study, we optimized HEK293T transfection and primary T-cell transduction parameters for lentiviral CAR constructs targeting BCMA (B-cell maturation antigen) and GPRC5D (G-protein coupled receptor family C group 5 member D). Lipofectamine 3000 and TurboFectin 8.0 were compared across different seeding densities and reagent-to-DNA ratios, with vector yields quantified by qPCR (Quantitative Polymerase Chain Re

Distinct molecular features of ovarian adult granulosa cell tumors and co-occurring primary malignancies-an exploratory case series.

BACKGROUND: Ovarian adult granulosa cell tumor (aGCT) is a rare gynecological malignancy and has been associated with increased incidence of other primary malignancies, particularly endometrial. However, potential shared molecular features in these tumors remain poorly understood. This study aimed to use Next Generation Sequencing to explore molecular alterations between aGCT and co-occurring breast, colon, and endometrial cancers within the same patients. METHODS: Female individuals diagnosed with aGCT and a synchronous endometrial cancer, or with a history of breast, or colorectal cancer fr
