Beneficial effects of melatonin in protecting against cyclosporine A-induced cardiotoxicity are receptor mediated.

Melatonin, the chief product secreted by pineal gland, is capable of reducing free radical damage by acting directly as a free radical scavenger, and indirectly, by stimulating of antioxidant enzymes. Cyclosporine A (CsA) is the most widely used immunosuppressive drug, but its therapeutic use has several side effects including, i.e. nephrotoxicity and cardiotoxicity. This study was designed to examine the beneficial effects of melatonin in preventing CsA-induced cardiotoxicity. Additionally, we investigated the ability of melatonin to protect the rat heart via melatonin receptor. In one group …

The effect of acute hypothermia and serum potassium concentration on potassium cardiotoxicity in anesthetized rats.

We examined the effects of hypothermia on serum K+ concentration and the interaction of body temperature and K+ load on cardiac toxicity in anesthetized rats. Serum K+ concentration significantly decreased to 2.61 +/- 0.13, 2.59 +/- 0.19 and 2.39 +/- 0.14 mmol/l at 31.0 degrees C, 28.0 degrees C and 25.0 degrees C, respectively, from the control value of 2.80 +/- 0.15 mmol/l at 37.0 degrees C. We used a 300% increase in baseline QRS duration as evidence of cardiac toxicity. Serum K+ concentrations of 4.95 +/- 0.12, 4.71 +/- 0.10, 4.45 +/- 0.14 and 4.07 +/- 0.14 mmol/l resulted in cardiac toxic

Morin mitigates doxorubicin-induced cardiotoxicity via suppression of oxidative stress, inflammation, and structural remodeling.

The clinical use of the anticancer drug doxorubicin is restricted by its ability to cause cardiotoxicity, often progressing to heart failure. Current cardiovascular treatments fail to restore cardiac function or improve long-term survival rates, demanding alternative therapeutic approaches to minimize the cardiotoxicity of conventional cancer treatments. Thus, this study investigated the alleviating potential of morin, a flavonoid with known cardioprotective, antioxidant, and anti-inflammatory effects, against doxorubicin-induced cardiotoxicity at two different time points. Sixty male Wistar r…

Inhibition of Calpain-1 ameliorates doxorubicin-induced cardiotoxicity by suppression of p53-mediated ferroptosis.

Doxorubicin (Dox), a widely applied anthracycline antitumor agent, exhibits severe dose-dependent cardiotoxicity that greatly restricts its clinical utilization, yet the molecular mechanism linking Calpain-1 to cardiomyocyte ferroptosis in doxorubicin-induced cardiotoxicity (DIC) remains poorly clarified. This study aimed to explore how Calpain-1 regulates cardiomyocyte ferroptosis in DIC. Differentially expressed proteins from Dox-treated H9c2 cells were screened by proteomics, stable Calpain-1 knockdown cell lines were generated to detect cell viability and ferroptosis indicators through CCK…

Network pharmacology-based therapeutic intervention of Mentha arvensis targeting cancer and doxorubicin-induced cardiotoxicity.

BACKGROUND: Doxorubicin (Dox) is a widely used chemotherapeutic agent effective against hematological malignancies and solid tumors, including leukemia, breast and bladder cancers, lymphomas, and Kaposi's sarcoma. Despite its clinical efficacy, the therapeutic application of Dox is severely limited by dose-dependent cardiotoxicity, manifesting as both acute and chronic Doxorubicin-induced cardiotoxicity (DIC). Dexrazoxane (Dexa) remains the only FDA-approved cardioprotective agent available; however, it compromises the anti-cancer activity of Dox and poses risks such as secondary malignancies,…

Chimeric antigen receptor-T cell therapy-induced cardiotoxicity: Pathophysiological mechanisms and pharmacological intervention strategies.

Chimeric antigen receptor (CAR)-T cell therapy has emerged as a revolutionary treatment for hematologic malignancies and solid tumors. Multiple approved products and ongoing clinical trials have demonstrated its remarkable antitumor efficacy. However, its clinical application is severely limited by prominent cardiotoxicity, which is closely associated with high morbidity and mortality, posing an urgent clinical challenge. This review systematically elucidates the pathophysiological mechanisms underlying CAR-T cell therapy-induced cardiotoxicity, which can be primarily categorized into acute an…

Cardiac substructure-guided radiotherapy planning for esophageal cancer: a novel strategy for cardiotoxicity reduction.

BACKGROUND: A novel method was proposed to protect the sensitive cardiac substructures in esophageal cancer patients and reduce the risk of cardiotoxicity. METHODS: A retrospective study was conducted on 20 patients with esophageal cancer. For each patient, a conventional clinical plan (c-IMRT) and a plan to protect sensitive cardiac substructures (p-IMRT) were designed. Dosimetric parameters of the two plans were calculated. The tumor control probability (TCP) of the target and the normal tissue complication probability (NTCP) of normal tissue were compared between two plans. Paired sample t…

Randomized Comparison of Cardiotoxicity With 60 Versus 90 mg Daunorubicin in AML Induction Therapy.

Anthracyclines are an essential component of induction therapy for acute myeloid leukemia (AML), but their optimal dosing and the associated risk for cardiotoxicity remain under debate. The DaunoDouble trial compared daunorubicin at 60 (Dauno60) versus 90 mg/m2 (Dauno90) in combination with cytarabine (100 mg/m2 for 7 days) in newly diagnosed AML patients aged 18-65 years. Cardiac function was assessed by left ventricular ejection fraction (LVEF) and cardiac biomarkers (high-sensitivity troponin T [hsTnT], NT-proBNP) before treatment and on Day 15 in 317 randomized patients. Median LVEF declin…

Lipoic acid-modified epirubicin liposomal system for tumor-targeted drug delivery and cardiotoxicity reduction.

This study aimed to address the bottlenecks of low delivery efficiency and high cardiotoxicity of anthracyclines by constructing liposomes of epirubicin functionalized with lipoic acid (Epi Lip@LA). This system was endowed with dual functions of dynamic targeting and microenvironmental response through lipoic acid modification: the lipoic acid group in DSPE-PEG2000-LA enhanced the uptake efficiency of tumor cells through dynamic covalent bond-mediated targeted delivery, and the high concentration of glutathione (GSH) in the tumor microenvironment triggered the specific cleavage of disulfide bo…

FAERS based disproportionality analysis and network pharmacology investigation of doxorubicin associated drug induced cardiotoxicity.

Doxorubicin (DOX) is crucial for cancer treatment but its use is limited by cardiotoxicity. Liposomal doxorubicin (Lip-DOX) was designed to reduce this toxicity compared to conventional doxorubicin (Con-DOX). This study utilized the U.S. Food and Drug Administration Adverse Event Reporting System (FAERS) database to compare the cardiotoxicity profiles of the two formulations through disproportionality analysis and to explore potential mechanisms using network pharmacology. We analyzed 10,695 adverse event reports for Con-DOX and 13,164 for Lip-DOX. Disproportionality analysis, measured by the …

Phyllanthin mitigates doxorubicin-induced acute cardiotoxicity: role of Bax/Bcl2 and Caspase-3 regulation.

Doxorubicin (DOX)-induced toxicity impairs cardiovascular functions. Phyllanthus amarus exhibits anti-inflammatory, antiviral, anticancer, antioxidant, nephroprotective, and hepatoprotective effects. However, its potential against DOX-induced acute cardiotoxicity is lacking. This study aimed to assess the underlying mechanism of action of P. amarus against DOX-induced acute cardiotoxicity in Sprague-Dawley rats. Phyllanthin was isolated from the methanolic extract of the aerial parts of P. amarus and characterized using HPLC-ESI-MS/MS. Its cardioprotective efficacy was evaluated in DOX-intoxic…
