Results of using a new enzyme-linked immunosorbent assay (ELISA) kit for detecting rubella IgM antibodies were compared with those obtained by fractionating samples of serum on sucrose density gradients followed by rubella haemagglutination inhibition tests on the fractions. There was close agreement between the two methods. A major advantage of the ELISA method was the ability to complete tests within one day.

We have investigated the response of rat liver nuclear, nuclear envelope, and microsomal cytochrome P-450 (or P-448) to various treatments. Responses of these subcellular fractions to 3-methylcholanthrene pretreatment were generally similar. In endoplasmic reticulum preparations, we observed an increase in cytochrome P-450 content following phenobarbital pretreatment, which was reduced by subsequent thioacetamide treatment. Cell structure abnormalties are found in acute leukaemia and preleukaemic states. Studies on bone marrow cells and peripheral leucocytes of 4 patients with idiopathic acquired sideroblastic anaemia showed patterns in cell culture similar to those reported in acute leukaemia: 2 of these patients later developed leukaemia. The transport of [(14)C]biotin into cells of a biotin prototroph, Escherichia coli K-12 strain Y10-1, was investigated. The vitamin taken up by the cells in this strain existed primarily in the free form. Addition of glucose enhanced the rate of uptake six- to eightfold and the steady level was reached in 2 to 3 min resulting in accumulation of biotin against a concentration gradient. The biochemical and physiological basis of density heterogeneity in Renografin of Bacillus subtilis W23 spores was determined by analysis of metals, macromolecules, and dipicolinic acid in the two density classes of the population. Germination rate and heat resistance were measured in both density classes. The glycoprotein, but no other virion protein, of vesicular stomatitis virus was solubilized by the nonionic detergent Triton X-100 in low ionic strength buffer. The solubilized viral glycoprotein induced the synthesis of antibody that formed a single precipitin line with the glycoprotein and that neutralized the infectivity of the virus. The neutralizing activity of the antibody was efficiently blocked by purified glycoprotein.

OBJECTIVE: The Kato-Katz technique misses a proportion of low-intensity schistosomiasis cases. We set out to estimate the rate of false negative results after Kato-Katz.
METHOD: Comparison with the more sensitive Percoll technique and polymerase chain reaction (PCR).

The outer membrane of the classical Vibrio cholerae strain 569B was isolated by sucrose density centrifugation. Secretory granules from rat antral tissue were isolated by differential centrifugation in sucrose and were confirmed as intact by electron microscopy.
