An impaired mitochondrial electron transport chain increases retention of the hypoxia imaging agent diacetylbis(4-methylthiosemicarbazonato)copperII.

Radiolabeled diacetylbis(4-methylthiosemicarbazonato)copper(II) [Cu(II)(atsm)] is an effective positron-emission tomography imaging agent for myocardial ischemia, hypoxic tumors, and brain disorders with regionalized oxidative stress, such as mitochondrial myopathy, encephalopathy, and lactic acidosis with stroke-like episodes (MELAS) and Parkinson's disease. An excessively elevated reductive state is common to these conditions and has been proposed as an important mechanism affecting cellular retention of Cu from Cu(II)(atsm). However, data from whole-cell models to demonstrate this mechanism

Malnutrition at the cellular level in uremia: a new frontier for research.

Previously reported studies showed that protein-calorie malnutrition (PCM) in children is associated with significant alterations in muscle cell composition, characterized by increased cell water and Na, with decreased concentrations of K, Mg, and some glycolytic and citric acid cycle metabolites. In PCM, the i.c. H2O and levels of Na and K were significantly related, by multiple regression analysis, to the combined set of simultaneously measured levels of cell metabolites, phosphoenolpyruvate, pyruvate, alpha-ketoglutarate, and oxaloacetate. Cell bioactivities also were affected by PCM. Activ…

Characterization of the metabolome and proteome of stem cell-derived human primordial germ cell-like cells: a multi-omics approach.

Primordial germ cells (PGCs) are the population of cells that, in the human embryo, are initially specified at day 12 post-fertilization and … articularly evident at the proteome level. This included a shift between canonical and noncanonical citric acid cycles in hPGCLCs, downregulation of late-stage glycolysis and reduction of nucleotide de novo synthesis. By providing an integrative map of these metabolic networks, we aim to provide insight into the metabolism of hPGCLC development that could help impr …

Microfluidic contact lens for continuous monitoring of ocular oxidative stress.

Oxidative stress-induced ocular dysfunctions are a significant global health concern. Glutathione (GSH), an abundant antioxidant in tears, can serve as an index for oxidative stress (OS). A coumarin-based fluorescent probe named CCAE was synthesized for GSH monitoring, functioning through structural changes via Michael's addition with GSH and elimination by H2O2, which restores the conjugation structure to assess the severity of OS-induced ocular diseases. CCAE demonstrated a high sensitivity with a detection limit (LOD) of 0.12 mM and reversibility for 15 cycles. A wearable contact lens senso

Biophysical and biochemical studies support TP0094 as a phosphotransacetylase in an acetogenic energy-conservation pathway in Treponema pallidum.

The mechanisms of energy generation and carbon-source utilization in the syphilis spirochete Treponema pallidum have remained enigmatic despite complete genomic sequence information. Whereas the bacterium harbors enzymes for glycolysis, the apparatus for more efficient use of glucose catabolites, namely the citric-acid cycle, is apparently not present. Yet, the organism's energy needs likely exceed the modest output from glycolysis alone. Recently, building on our structure-function studies of T. pallidum lipoproteins, we proposed a "flavin-centric" metabolic lifestyle for the organism that pa

Mathematical modeling reveals spontaneous emergence of self-replication in chemical reaction systems.

Explaining the origin of life requires us to elucidate how self-replication arises. To be specific, how can a self-replicating entity develo … whereby reactants are transformed into end products with the assistance of intermediates (as in the citric acid cycle), whereas some others are self-replicating, that is, different parts replicate each other and the system self-replicates as a whole (as in the formose reaction, in which sugar is replicated from formaldehyde); 2) side reactions do not always inhibit …

Metabolomic profiling reveals new insights into human adenovirus type 7 infection.

Human adenovirus type 7 (HAdV-7) is a prominent pathogen that causes severe pneumonia in children in China. However, the interaction between HAdV-7 infection and host metabolism is still poorly understood. To gain a comprehensive understanding of the metabolic interplay between host cells and the virus, we analysed the energy and lipid metabolism profiles of the HAdV-7-infected lung cancer cell line A549 by ultrahigh-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (ESI-QTRAP-MS/MS). Our study revealed significant alterations in various metabolic processes, includi

Genetic Dissociation of Glycolysis and the TCA Cycle Affects Neither Normal nor Neoplastic Proliferation.

Rapidly proliferating cells increase glycolysis at the expense of oxidative phosphorylation (oxphos) to generate sufficient levels of glycolytic intermediates for use as anabolic substrates. The pyruvate dehydrogenase complex (PDC) is a critical mitochondrial enzyme that catalyzes pyruvate's conversion to acetyl coenzyme A (AcCoA), thereby connecting these two pathways in response to complex energetic, enzymatic, and metabolic cues. Here we utilized a mouse model of hepatocyte-specific PDC inactivation to determine the need for this metabolic link during normal hepatocyte regeneration and mali

Biophysical and biochemical evidence for the role of acetate kinases (AckAs) in an acetogenic pathway in pathogenic spirochetes.

Unraveling the metabolism of Treponema pallidum is a key component to understanding the pathogenesis of the human disease that it causes, syphilis. For decades, it was assumed that glucose was the sole carbon/energy source for this parasitic spirochete. But the lack of citric-acid-cycle enzymes suggested that alternative sources could be utilized, especially in microaerophilic host environments where glycolysis should not be robust. Recent bioinformatic, biophysical, and biochemical evidence supports the existence of an acetogenic energy-conservation pathway in T. pallidum and related treponem

Magnetic Mesoporous γ‑Fe2O3 Supports from Meso-MIL-88A via Controlled Oxidation-Reduction Pyrolysis: Enabling Recyclable Lipase Catalysis in Phospholipid Synthesis.

Enzyme immobilization on solid supports enhances stability and reusability, yet nanoscale carriers such as metal-organic frameworks (MOFs) still face challenges in efficient recovery. While pyrolysis can magnetize Fe-MOFs, conventional methods often compromise either enzyme activity or structural integrity. This study presents a rational two-step oxidation-reduction (O-R) pyrolysis strategy to convert Meso-MIL-88A into a magnetically recyclable, mesoporous biocatalyst support (O-R500). Unlike one-step carbonization, which generates enzyme-incompatible Fe3O4, or carbonization-oxidation routes t
