Heterorhabditis noenieputensis n. sp. (Rhabditida: Heterorhabditidae), a new entomopathogenic nematode from South Africa.

A new entomopathogenic nematode in the genus Heterorhabditis is described from South Africa, from two singular isolates found 1000 km from each other, from beneath a fig tree and in a citrus orchard, respectively. Morphological and molecular studies indicate both isolates to be the same and a new undescribed Heterorhabditis species. Comparison of sequences of the internal transcribed spacer (ITS) rDNA and the D2D3 region of the 28S rDNA gene with available sequences of other described species within the genus, indicate the two isolates as a new species. Phylogenetic analysis of the sequence da

Evaluation of a novel Dot-ELISA assay utilizing a recombinant protein for the effective diagnosis of Taenia pisiformis larval infections.

Cysticercosis, caused by the larvae of Taenia pisiformis, is a common disease in domestic breeds of the rabbit Oryctolagus cuniculus that results in economic losses. At present, there is no convenient and effective method for the rapid detection of T. pisiformis larvae. Here, we developed and tested the efficacy of a Dot-ELISA assay for the diagnosis of T. pisiformis larval infections in rabbits, based on the expression of the recombinant fusion protein (rTp1) from the Tp1 gene. Rapid amplification of cDNA ends (RACE) was used to amplify the 3' ends of the Tp1 gene, based on the unigene simila

Annexin B1 from Taenia solium metacestodes is a newly characterized member of the annexin family.

We previously reported cloning of the Taenia solium annexin B1 gene from a metacestode cDNA expression library and demonstrated that it acts as a protective antigen for effective vaccine development against cysticercosis. In the present study we produced recombinant annexin B1 and antiserum against the protein to investigate its structural and functional properties. Western blotting of metacestode fractions indicated that T. solium annexin B1, similar to vertebrate annexins, associates with acid phospholipids in the presence of Ca(2+). This property was confirmed by the recognition of apoptoti

A novel cDNA clone encoding a specific excretory/secretory antigen of larval Trichinella pseudospiralis.

A cDNA library for Trichinella pseudospiralis was constructed to study the expression of specific antigens. Four positive clones were identified using antibodies against the excretory/secretory (ES) products of the nematode as probe. Sequence analysis showed that they contained identical cDNA inserts of 606 bp, including a 5' non-translated region of 96 bp, a core translated segment of 408 bp and a poly(A)+ 3' terminus. It encoded a novel 136-amino-acid polypeptide. Southern blot analysis indicated that the cDNA did not cross-hybridize to the genomic digests of T. spiralis, mouse, or rat. A si

[DNA prime followed by protein boost enhances the protective efficacy against Schistosoma japonicum infection in mice].

Schistosomiasis japonica is an endemic, zoonotic disease of major public health importance in China. Vaccination is needed as a complementary approach to the ongoing control programs. In the present study, we determined if the efficacies of DNA vaccine encoding the SjGST and Sj32 asparaginyl endopeptidase protein could be enhanced by boosting with SjGST-32 protein vaccines. Mice were inoculated with a VR1012-SjGST-32 DNA vaccine followed by boosting with rSjGST-32 at 0, 14 and 28 d. Two weeks after the final boost, mice were challenged percutaneously with cercariae. On day 45 following the cha

Loop-mediated isothermal amplification method for a differential identification of human Taenia tapeworms.

Loop-mediated isothermal amplification (LAMP), which employs a Bst DNA polymerase with strand-displacement activity and four primers (two inner primers and two outer primers) recognizing six distinct regions on the target DNA, is a highly sensitive, specific, simple, and rapid nucleotide amplification method. Moreover, because the Bst DNA polymerase resists much DNA polymerase inhibitors present in biological specimens, the LAMP method is suitable for the detection of infectious agents from clinical material such as fecal samples. Here, we describe the LAMP method which can differentially dete

A combination of two Brugia malayi filarial vaccine candidate antigens (BmALT-2 and BmVAH) enhances immune responses and protection in jirds.

In this study filarial recombinant protein or DNA vaccine constructs encoding BmALT-2 and BmVAH as single or as cocktail antigens were evaluated. Male jirds were immunized intramuscularly with DNA vaccine constructs or were immunized intraperitoneally with protein vaccine. The single and bicistronic DNA constructs induced substantial interferon-γ responses in spleen cells; antigen-specific responses were higher following immunization with the bicistronic cocktail construct and evoked a significant protective response of 57% in jirds challenged with Brugia malayi that was similar in the antibod

Alternative splicing and sequence diversity of transcripts from the oncosphere stage of Taenia solium with homology to the 45W antigen of Taenia ovis.

Genes and transcripts which show homology to the host-protective 45W antigen of Taenia ovis have been cloned from the human parasite Taenia solium. The T. solium genes cloned in this study (TSO45) show conserved genomic structural features which are also features of the T. ovis 45W gene family. The TSO45 genes consist of a four exon and three intron structure. Eight TSO45 transcripts, encoded by at least five genes, were cloned from T. solium oncospheres and comparison of their DNA sequence indicates that some transcripts have arisen by alternative splicing, the first demonstration of exon inc

Trichinella spiralis (T1) and Trichinella T5: a comparison using animal infectivity and molecular biology techniques.

We compared Trichinella T5 of bobcat (Lynx rufus) origin with Trichinella spiralis (T1) by using animal infectivity and molecular biology techniques. Swine, SD rats, and CF1 mice were highly resistant to infection with Trichinella T5 but sensitive to T. spiralis, whereas deer mice (peromyscus maniculatus) had similar sensitivity to both parasites. The fecundity of Trichinella T5 in deer mice was 10-35-fold higher in comparison to the fecundity in laboratory rodents (SD rats and CF1 mice). Fecundity of T. spiralis was approximately the same in both groups. A western blot, using excretory-secret

Another host, another mystery: Infection of polychaete Pista palmata (Annelida: Terebellidae) with a new blood fluke (Digenea: Aporocotylidae) in the coastal Northwest Atlantic Ocean.

Terebelliform annelids can serve as intermediate hosts for blood flukes of marine fishes and sea turtles. The scarcity of global reports on the life cycles of these parasites reflects the need for further exploration. Our objective was to further our knowledge of the diversity and life cycles of blood flukes in coastal South Carolina (SC), United States. Four species of terebellids (Enoplobranchus sanguineus, Amphitrite ornata, Pista palmata, and Streblosoma hartmanae) were collected (total N = 201) at low tide from September 2025 to February 2026 at two localities in SC. Population structure of Clinostomum complanatum (Trematoda: Digenea) with new data on haplotype diversity of flukes from Slovakia and Italy.

The fluke Clinostomum complanatum, a parasite of piscivorous birds, but also reptiles and rarely mammals, has established several foci in the western Palaearctic regions. Previous studies pointed out the complicated taxonomy of the genus, but broader population genetic analysis of C. complanatum has not yet been carried out. The aim of this study was to determine the structure, intraspecific variability, and diversity of mitochondrial cox1 haplotypes of C. complanatum from different localities in Slovakia (Danube floodplain forests) and Italy (Emilia-Romagna and Tuscany), as well as to evaluat

Description of three new species of Gyrodactylus von Nordmann, 1832 (Monogenea: Gyrodactylidae) on bitterling fishes (Acheilognathinae) from China.

Three new species of Gyrodactylus are described from three species of bitterling in Donghu Lake, China: Gyrodactylus ocellorhodei n. sp. from Rhodeus ocellatus; G. sinenorhodei n. sp. from Rhodeus sinensis; and G. acheilorhodei n. sp. from Acheilognathus macropterus. All the three new species showed similar opisthaptor morphology, especially the marginal hooks: all had a slender and perpendicular sickle shaft, and flat sickle base with distinct heel and inner arch which was different from the G. rhodei-group species parasitic on bitterling. Multivariate analyses based on hamulus and marginal h

Expanding on expansus: a new species of Scaphanocephalus from North America and the Caribbean based on molecular and morphological data.

Members of the genus Scaphanocephalus mature in accipitrids, particularly osprey, Pandion haliaetus, with metacercaria causing Black Spot Syndrome in reef fishes. In most of the world, only the type species, Scaphanocephalus expansus (Creplin, 1842) has been reported. Recent molecular studies in the Western Atlantic, Mediterranean and Persian Gulf reveal multiple species of Scaphanocephalus, but have relied on 28S rDNA, mainly from metacercariae, which limits both morphological identification and resolution of closely related species. Here we combine nuclear rDNA with mitochondrial sequences f

Methodological assessment for efficient collection of Schistosoma mansoni environmental DNA and improved schistosomiasis surveillance in tropical wetlands.

Schistosomiasis, caused by trematodes of genus Schistosoma, is among the most seriously neglected tropical diseases. Although rapid surveillance of risk areas for Schistosoma transmission is vital to control schistosomiasis, the habitat and infection status of this parasite are difficult to assess. Environmental DNA (eDNA) analysis, involving the detection of extra-organismal DNA in water samples, facilitates cost-efficient and sensitive biomonitoring of aquatic environments and is a promising tool to identify Schistosoma habitat and infection risk areas. However, in tropical wetlands, highly

Molecular and morphological evidence of a new species of Crassicutis Manter 1936 (Digenea), a parasite of cichlids in South America.

A new species of Crassicutis Manter, 1936 (Digenea: Megaperidae) is described from the intestine of Satanoperca jurupari (Cichlidae) in the Amazon River basin, Brazil. The genus Crassicutis currently contains eight species. Crassicutis manteri n. sp. is morphologically very similar to Crassicutis cichlasomae Manter, 1936, a parasite of cichlids reported from Mexico, the Antilles, and Central and South America. Molecular data revealed that C. cichlasomae represents a species complex in Middle American cichlids. The new species can be readily distinguished from C. cichlasomae sensu lato, and the
