Despite numerous therapeutic strategies targeting specific programmed cell death pathways, effectively eliminating malignant colorectal cancer (CRC) remains a significant challenge. Disulfidptosis, a newly identified form of cell death, is characterized by rapid NADPH depletion and abnormal disulfide bond formation in cytoskeletal proteins in cells with high SLC7A11 expression under glucose starvation. Given the aberrant expression of SLC7A11 and the hypermetabolic phenotype of CRC cells, targeting the disulfidptosis pathway offers a promising therapeutic approach for CRC treatment. Here, we d

The overuse of acetaminophen (APAP) poses significant health risks to human health, which necessitates the development of an efficient and rapid detection method for APAP. Hence, an electrochemical sensor using molybdenum disulfide-grafted carbon nanotubes (MoS2/CNTs) was developed for the ultra-sensitive detection of acetaminophen (APAP). The linear detection range of the sensor was 0.07 to 150 µM, and the limit of detection for APAP was 0.43 µM. The MoS2/CNTs composite demonstrated significantly enhanced catalytic activity toward APAP, attributed to its enormous specific surface area and eff

Redox homeostasis is frequently disrupted in cancer and contributes to tumor progression, metastasis, and therapy resistance. This review fo … ced on emerging redox-responsive approaches such as cyclic oligochalcogenides, cell-penetrating polydisulfides, and redox-sensitive antibodies. Overall, this review highlights extracellular redox regulation as a tumor-associated feature that can serve both as a biomarker and as a basis for next-generation cancer theranostics, offering complementary opportuni …

Human epidermal growth factor receptor 2 (HER2) is a critical therapeutic target in various cancers, but its accurate noninvasive quantification remains challenging. Existing positron emission tomography (PET) tracers, such as antibodies and peptides, are limited by suboptimal clearance, insufficient tumor uptake and retention, and poor proteolytic stability. Here, we introduce a new class of disulfide-directed multicyclic peptides (DDMPs) that combine a conformationally rigid and stable scaffold with excellent HER2-binding properties. Using yeast display and directed evolution, we identified

Given the significant advantages the eye presents as a target organ for gene therapy, it has remained at the forefront of translational research in this field. However, gene delivery to the desired tissue, particularly the retina, via a noninvasive route of administration poses a substantial physical challenge due to the presence of multiple ocular barriers. Here, we develop disulfide-appended small interfering RNA (DS-siRNA) Nanoeyedrops that target pathological retinal angiogenesis. These Nanoeyedrops enable noninvasive and effective gene delivery into the retina through the thiol-mediated u

The structural dynamics of biopolymers, such as proteins and DNA, feature the unique combination of simultaneous conformational adaptivity and configurational reversibility. However, it remains challenging to design a synthetic system featuring dynamic covalent bonds with secondary structural folding and full recycling capabilities. Here we present a synthetic covalent polymer, constructed from biologically relevant units (amino acids and disulfides), that can reversibly adapt between disordered and helical conformations and can recycle itself to the original small molecules. This dual dynamic…
