Resilience and vulnerability in the midbrain dopamine system during aging.

The midbrain dopamine system is integral to movement, reward, and motivation, and its dysfunction underlies a range of brain disorders. Age is a primary risk factor for many of these disorders, yet systematic analysis of how the dopamine system changes during healthy aging remains relatively understudied. In this review, we first examine the impact of healthy aging on the dopamine system across sexes and species, including flies, mice, rats, and humans. We next consider the neuroadaptive mechanisms that sustain dopamine neuron function with age. Finally, we contrast the molecular mechanisms of…

Effects of dopamine and serotonin on persistence for delayed rewards in humans.

People often must decide how long to continue waiting for rewards that will arrive at an uncertain time in the future. We propose that these persistence decisions involve weighing the benefits of continued waiting against opportunity costs of waiting, a balance that may shift over time. This framework suggests that persistence decisions share neural mechanisms with foraging decisions, which require ongoing comparisons between a current resource and possible alternatives. Dopamine and serotonin have been proposed to play opposing roles in foraging, with dopamine promoting exploration and seroto…

On the mechanisms of dopamine receptor agonists in restless legs syndrome.

Several dopaminergic compounds, including the clinically used pramipexole, are labeled as preferential dopamine D3 receptor (D3R) agonists based on their moderately higher affinity for the D3R versus other D2-like receptor subtypes. It is therefore generally believed that D3R is the main target for their initial therapeutic response in restless legs syndrome (RLS). Here, we review the results of recent comprehensive pharmacological studies that demonstrate that in addition to the binding affinities of D3R agonists, their functional responses at the different D2-like receptors depend on other p…

Impact of NPAS2 on mPFC dopamine synthesis and nap behavior.

The biological basis of the afternoon nap, a widespread yet poorly understood phenomenon, has remained elusive. Here we identify NPAS2, among core circadian regulators, as a sex-independent determinant of the nap behavior in mice. Specifically, medial prefrontal cortex (mPFC)-expressed NPAS2 orchestrates nap regulation through circadian modulation of local dopaminergic activity. We demonstrate that tyrosine hydroxylase-positive (TH+) neurons in mPFC exhibit time-of-day dependent wake-promoting activity, showing minimal excitation precisely during nap hours. Mechanistically, NPAS2 achieves this…

Dopamine depletion in Parkinson's increases directed but not random exploration.

We investigated how patients with Parkinson's disease (PD) manage the explore-exploit trade-off in a structured reward-learning task. Patients were tested either on (n = 34) or off (n = 34) dopaminergic medication (levodopa), with age-matched polyneuropathy patients serving as controls (n = 35). Behaviorally, patients off medication showed marked learning and decision-making deficits, characterized by overexploration (excessive sampling of novel options) and insufficient exploitation (re-selection of previously rewarding options). To clarify the underlying mechanisms, we applied a Gaussian pro…

Longitudinal Trajectory of Dopamine and Serotonin Transporters in Parkinson Disease.

Parkinson disease (PD) is a multisystem disorder marked by progressive dopaminergic neuronal degeneration in the substantia nigra, as well as nondopaminergic systems. Our aim was to investigate longitudinal changes in N-(3-[18F]fluoropropyl-2β-carbomethoxy-3β-(4-iodophenyl)nortropane (18F-FP-CIT) binding at the putamen, substantia nigra, and raphe nuclei in PD. Methods: This retrospective cohort study enrolled 127 patients with PD, who underwent 18F-FP-CIT PET scans twice or more, and 71 age- and sex-matched healthy controls. A temporal trajectory model was created to estimate the longitudinal…

Parthenolide acts as a cocaine antagonist in the human dopamine transporter.

In this work, I describe the effects of parthenolide, a naturally occurring sesquiterpene lactone, on radiolabeled cocaine binding and on radiolabeled dopamine uptake in the human dopamine transporter expressed on HEK-293 cells (HEK/DAT cells). Parthenolide displaces radiolabeled cocaine in a concentration-dependent manner and does not display any significant inhibition of radiolabeled dopamine uptake. Further, parthenolide seems to alleviate the inhibition of dopamine uptake by 5 μM unlabeled cocaine at sub-toxic parthenolide concentrations.

Dopamine Partial Agonists in Pregnancy and Lactation: A Systematic Review.

Background/Objectives: Dopamine partial agonists are drugs initially developed to treat schizophrenia, seeking a double effect of increased dopaminergic transmission in the prefrontal cortex and decrease in the accumbens/striatum. Of these drugs, aripiprazole, brexpiprazole, and cariprazine are currently marketed and used in schizophrenia spectrum and mood disorders. It is debated whether patients with psychiatric disorders becoming pregnant should discontinue or continue their antipsychotic treatment despite some risks for the fetus, i.e., whether it is worse to have an untreated disorder or …

Dopamine dynamics are dispensable for movement but promote reward responses.

Dopamine signalling modes differ in kinetics and spatial patterns of receptor activation1,2. How these modes contribute to motor function, motivation and learning has long been debated3-21. Here we show that action-potential-induced dopamine release is dispensable for movement initiation but supports reward-oriented behaviour. We generated mice with dopamine-neuron-specific knockout of the release site organizer protein RIM to disrupt action-potential-induced dopamine release. In these mice, rapid in vivo dopamine dynamics were strongly impaired, but baseline dopamine persisted and fully suppo…

Interactions of Oxytocin and Dopamine-Effects on Behavior in Health and Disease.

The hypothalamic neuropeptide and hormone oxytocin are of fundamental importance for maternal, social, and sexual behavior. Deviations in oxytocin levels have also been associated with anxiety, autism spectrum disorders (ASD), depression, ADHD (attention deficit hyperactivity disorder), and schizophrenia. Both oxytocin and dopamine are often considered reward- and feel-good hormones, and dopamine is associated with the above-mentioned behaviors and, and dopamine is also associated with the above-mentioned behaviors and disorders. Although being structurally totally different, oxytocin, a pepti…

Dopamine in the regulation of glucose and lipid metabolism: a narrative review.

OBJECTIVE: Owing to the global obesity epidemic, understanding the regulatory mechanisms of glucose and lipid metabolism has become increasingly important. The dopaminergic system, including dopamine, dopamine receptors, dopamine transporters, and other components, is involved in numerous physiological and pathological processes. However, the mechanism of action of the dopaminergic system in glucose and lipid metabolism is poorly understood. In this review, we examine the role of the dopaminergic system in glucose and lipid metabolism. RESULTS: The dopaminergic system regulates glucose and li…

Is prolactin receptor signaling a target in dopamine-resistant prolactinomas?

The hypothalamic neuroendocrine catecholamine dopamine regulates the lactotroph function, including prolactin (PRL) secretion, proliferation, and apoptosis. The treatment of PRL-secreting tumors, formerly known as prolactinomas, has relied mainly on this physiological characteristic, making dopamine agonists the first therapeutic alternative. Nevertheless, the group of patients that do not respond to this treatment has few therapeutical options. Prolactin is another physiological regulator of lactotroph function, acting as an autocrine/paracrine factor that controls PRL secretion and cellular …

How to manage intolerance to dopamine agonist in patients with prolactinoma.

PURPOSE: Dopamine agonists (DA) are the gold-standard for prolactinoma and hyperprolactinemia treatment. Intolerance to DA leading to drug drop out occurs in 3 to 12% of cases. We provide here a review of published data about DA intolerance and present a case report concerning the use of intravaginal cabergoline. METHODS: We review the literature on the definition, the pathogenesis, frequency and management of DA intolerance. In addition, the review provides strategies to enhance tolerability and avoid precocious clinical treatment withdrawal. RESULTS: Cabergoline is often cited as the most …

Reappraising the role of dopamine in adolescent risk-taking behavior.

Adolescence is characterized by increased risk-taking, which is often ascribed to developmental changes in dopaminergic signaling. Popular models propose that these behaviors are caused by dopamine-induced hypersensitivity to rewards, which overrides adolescents' immature self-control mechanisms. However, these models are often based on oversimplified notions about the workings and functions of dopamine. Here we discuss the relationship between changes in the dopaminergic system and adolescent behavior in light of current theories/models about the functions of dopamine. We show that dopamine i…

Endothelium-Derived Dopamine and 6-Nitrodopamine in the Cardiovascular System.

The review deals with the release of endothelium-derived dopamine and 6-nitrodopamine (6-ND) and its effects on isolated vascular tissues and isolated hearts. Basal release of both dopamine and 6-ND is present in human isolated umbilical cord vessels, human popliteal vessels, nonhuman primate vessels, and reptilia aortas. The 6-ND basal release was significantly reduced when the tissues were treated with Nω-nitro-l-arginine methyl ester and virtually abolished when the endothelium was mechanically removed. 6-Nitrodopamine is a potent vasodilator, and the mechanism of action responsible for thi…

Metabolic effects of prolactin and the role of dopamine agonists: A review.

Prolactin is a polypeptide hormone that is well known for its role in reproductive physiology. Recent studies highlight its role in neurohormonal appetite regulation and metabolism. Elevated prolactin levels are widely associated with worsening metabolic disease, but it appears that low prolactin levels could also be metabolically unfavorable. This review discusses the pathophysiology of prolactin related metabolic changes, and the less commonly recognized effects of prolactin on adipose tissue, pancreas, liver, and small bowel. Furthermore, the effect of dopamine agonists on the metabolic pro…

Neuroimaging of Dopamine Transporter Density in the Striatum of Disordered Gamblers.

The aim of the present research was to add to the growing literature on dopamine and gambling disorder (GD) by assessing whether GD is associated with dopamine transporter (DAT) density in the ventral striatum compared to healthy controls and whether DAT density was associated with key characteristics of GD (e.g., abstinence, craving). In a cross-sectional investigation using single-photon emission computed tomography with a technetium-99m-labeled tropane derivative as a radiotracer with SPECT imaging, fifteen participants with GD and 15 controls (non-gambling individuals, matched for age, gen…

Dopamine receptor 3: A mystery at the heart of cardiac fibrosis.

Dopamine receptors have been extensively studied in the mammalian brain and spinal cord, as dopamine is a vital determinant of bodily movement, cognition, and overall behavior. Thus, dopamine receptor antagonist antipsychotic drugs are commonly used to treat multiple psychiatric disorders. Although less discussed, these receptors are also expressed in other peripheral organ systems, such as the kidneys, eyes, gastrointestinal tract, and cardiac tissue. Consequently, therapies for certain psychiatric disorders which target dopamine receptors could have unidentified consequences on certain funct…
