Clinical implications of drug interactions with coxibs.

Nonsteroidal antiinflammatory drugs (NSAIDs) often are prescribed to patients who are taking concomitant drugs. Cyclooxygenase (COX)-2 inhibitors (coxibs) rofecoxib and celecoxib are highly selective inhibitors of COX-2, differentiating them from nonselective NSAIDs, which substantially inhibit both COX-1 and COX-2. Like nonselective NSAIDs, coxibs are hepatically metabolized: rofecoxib primarily by reduction by cytosolic enzymes and celecoxib by the cytochrome P450 (CYP) enzyme system. Because rofecoxib is not significantly metabolized by CYP, it has fewer confirmed or potential drug interact

Mitochondrial DNA mutations in Alzheimer's disease.

Several reports have indicated that point mutations of the mitochondrial DNA (mtDNA) contribute to the pathogenesis of Alzheimer's disease (AD). However, other groups have failed to find similar associations between these mutations and AD. A recent report described a set of mutations in the mtDNA encoded cytochrome oxidase genes which may account for 20% of all AD cases. We screened brain tissue from 65 AD patients for each of these previously reported mtDNA mutations but were unable to find an increased incidence of any of them in our AD sample. However, one patient with a mutation in the APP

Near-infrared spectroscopy in experimental pneumococcal meningitis in the rabbit: cerebral hemodynamics and metabolism.

Near-infrared spectroscopy is a noninvasive technique which measures oxidized cytochrome aa3, oxygenated Hb, and deoxygenated Hb and calculates total Hb in tissue. This technique, in conjunction with measurement of cerebral blood flow, was used in rabbits with experimental bacterial meningitis to determine whether there was evidence for cerebral energy depletion and alterations in the cerebral vascular bed with infection. Rabbits with meningitis had a significant reduction in cerebral blood flow, cerebral oxidized cytochrome aa3 and a relative increase in the deoxygenated Hb fraction and a dec

Muscle histopathology in myoclonus epilepsy with ragged-red fibers (MERRF).

Histopathologic findings were examined in skeletal muscle biopsies from 6 patients with myoclonus epilepsy with ragged-red fibers (MERRF) who had an A to G base substitution at mitochondrial DNA (mtDNA) nucleotide pair 8344. In addition to variation in fiber size and ragged-red fibers, all specimens in cross sections showed focal cytochrome c oxidase (CCO) deficiency, suggesting that this finding is crucial in elucidating the role of the mutant mtDNA in the pathogenesis of this disorder. Along the length of single muscle fibers, defects in CCO activity were distributed segmentally with blurred

[Cytochrome oxidase activity of peripheral blood leukocytes in patients with diffuse toxic goiter].

Patients with diffuse toxic goiter showed a distinct reduction of the cytochromoxidase activity in the peripheral blood leucocytes. The degree of reduction of the cytochromoxidase activity in the leucocytes depended on the severity of the clinical course and degree of increase of the thyroid gland. Results of determination of the cytochromoxidase activity of leucocytes in patients with diffuse toxic goiter in association with other clinico-laboratory data reflect the severity of the clinical course of the disease, the hypoxia degree and efficacy of instituted treatment.

The effect of pH on redox titrations of haem a in cyanide-liganded cytochrome-c oxidase: experimental and modelling studies.

Isolated cytochrome-c oxidase ligated with cyanide was titrated by Flash-Induced chemical photoREduction (FIRE) (Moody, A.J. and Rich, P.R. (1988) EBEC Short Rep. 5, 69) using cytochrome c as a redox indicator. Haem a is found to titrate in a complex manner consistent with its interacting anticooperatively with at least two other components. We assign CuB as the major interactant at neutral pH, and CuA as the minor interactant. In the pH range 7.0-8.1 the strength of the interaction with CuB is found to decrease with increasing pH, while the interaction with CuA remains essentially constant. T

Heteroplasmy of mitochondrial genomes in clonal cultures from patients with Kearns-Sayre syndrome.

We have analyzed heteroplasmy of mitochondrial DNA in clonal cultures from two patients with Kearns-Sayre syndrome, and have found that individual muscle or fibroblast clones contained either a mixed (i.e. heteroplasmic) population of normal and deleted mitochondrial DNAs, or only normal mitochondrial DNAs (i.e. homoplasmic at a level of detection of less than 1% deleted genomes). The heteroplasmic clones grew significantly more slowly than did "homoplasmic" clones, probably due to defects of respiratory chain enzymes containing mtDNA-encoded polypeptides.

Human achondroplasia: defective mitochondrial oxidative energy metabolism may produce the pathophysiology.

A summary is presented of previous studies by other investigators of human achondroplasia and dyschondroplastic animal models. In addition, studies previously reported from our laboratories are discussed, and they demonstrate that defective oxidative energy metabolism is present in mitochondrial preparations from achondroplastic human subjects and rabbits (ac/ac) with chondrodystrophy. The results of the studies support the hypothesis discussed fully in the manuscript that a partial defect in mitochondrial oxidative metabolism in achondroplastic subjects is expressed specifically in the growth

Variations of activities in the segments of respiratory chain among tissues in a patient with mitochondrial encephalomyopathy.

Enzymatic activities of NADH cytochrome c reductase and cytochrome c oxidase were determined in the mitochondria from various tissues of a patient with mitochondrial encephalomyopathy and compared with those of controls. NADH cytochrome c reductase in the present patient decreased significantly in the liver and spleen and to a less extent in the kidneys. On the other hand, cytochrome c oxidase of the patient decreased severely in the skeletal muscle and kidneys and partially in the heart. Carbon disulfide effects on the visual system. II. Retinogeniculate degeneration.

We examined the morphological effects of carbon disulfide exposure on neurons and vasculature of the visual system of macaque monkeys. Five monkeys were exposed to 256 ppm carbon disulfide (CS2) by inhalation for 6 hr a day, 5 days a week. One monkey, sacrificed immediately after exposure, had numerous axonal swellings in the distal optic tract. Four other monkeys survived the exposure period for at least 1 year and were found to have suffered marked degeneration of central retinal ganglion cells, with little or no effect on other neurons in the retina. No evidence was found for arteriosclerot

Demarcations of the mechanosensory projection zones in the raccoon thalamus, shown by cytochrome oxidase, acetylcholinesterase, and Nissl stains.

To determine anatomically the boundaries and internal organization of the kinesthetic and cutaneous mechanosensory regions of the ventrobasal thalamus, alternate section series from electrophysiologically mapped tissues from 14 raccoons were stained for cytochrome oxidase, myelinated fibers, acetylcholinesterase, and Nissl substance. Microelectrode tracks, along with electrolytic lesions placed as tissue markers, reveal that the mechanoreceptor projection zones have higher cytochrome oxidase and lower acetylcholinesterase staining than some neighboring regions. Both these enzymatic stains reve

31P nuclear magnetic resonance in vivo spectroscopy of the metabolic changes induced in the awake rat brain during KCN intoxication and its reversal by hydroxocobalamine.

Radiofrequency surface coils were chronically implanted in rats, which were subsequently subjected to 31P nuclear magnetic resonance (NMR) investigations at 4.7 T. The implanted coil allowed study of the animals without need for anesthesia, which is a prerequisite for studies of normal brain metabolism. The animals may be kept in the NMR probe for several hours. During subsequent experiments, they may be placed in the same position, therefore allowing follow-up studies for periods as long as 2 months. This method has been used in the study of sublethal KCN intoxication. KCN, a cytochrome c oxi

Effect of cyanide binding on the copper sites of cytochrome c oxidase: an X-ray absorption spectroscopic study.

Cu x-ray absorption spectroscopy (XAS) has been used to investigate the effect of cyanide treatment on the structures of the copper sites in beef heart cytochrome c oxidase. The Cu K-edge spectrum changes significantly upon cyanide binding to resting state enzyme, as does the Cu extended x-ray absorption fine structure (EXAFS) spectrum. The Cu EXAFS Fourier transfer (FT) exhibits an enhanced peak for the cyanide-treated enzyme in the region containing the Cu...Fe peak in the resting state FT (at R' approximately equal to 2.6-2.7 A). This peak in the cyanide-treated sample is hypothesized to ar

Local tissue oxygen tension-cytochrome a,a3 redox relationships in rat cerebral cortex in vivo.

Simultaneous measurements were made from rat cerebral cortex, in situ, of focal changes in both tissue oxygen tension (ptO2) and the reduction/oxidation ratio of cytochrome c oxidase (cytochrome a,a3) in order to study relationships between oxygen supply and consumption in small regions of tissue. Local ptO2 was measured with polarographic microelectrodes and the redox state of cytochrome a,a3 with a dual wavelength reflectance spectrophotometer. Increased ptO2, produced by respiration of gas mixtures with elevated O2 and/or CO2 content, was accompanied by increased oxidation of cytochrome a,a

A histoenzymological study of the saccus vasculosus of the freshwater teleost, Clarias batrachus (L.).

The saccus vasculosus (SV) of C. batrachus is comparatively small and situated latero-dorsal to the pituitary in between the lobi inferiores. It is in open communication with the third ventricle and is made up of coronet and supporting glial cells with interspersed liquor contacting neurons. These cellular constituents are arranged in one to three layers which are not thrown into folds. The PAS positive nature of the apical part of some coronet cells and their continuation with the PAS and AF positive material present in the lumen strongly suggest their secretory role. The coronet cells exhibi

Changes in activity of the Cu-Zn superoxide dismutase enzyme in tissues of the rat with changes in dietary copper.

The effects of dietary copper level on tissue activities of the copper containing superoxide dismutase (CuSOD) were investigated, and these activities related to those of other copper containing enzymes particularly cytochrome oxidase. Male weaning rats were fed a basal diet (containing 0.8 mg Cu/kg) or this diet supplemented with 4 or 24 mg Cu/kg. After 6 weeks, rats fed the basal diet were then repleted using the high copper diet. In the two copper supplemented groups, no differences were observed in any of the parameters measured. In these groups, tissue activities of CuSOD were in the orde

Comparison of kinetic and end-point microdensitometry for the direct quantitative histochemical assessment of cytochrome oxidase activity.

Cytochrome oxidase activity has been assessed by a method of kinetic microdensitometry which involves applying tissue sections to gel films containing phenylamine substrates and measuring the rate of azine dye production by continuously recording the rate of change in extinction. Optimum conditions for the technique were defined, and the results compared with those obtained by conventional end-point microdensitometry in which sections are incubated in histochemical substrate solutions and azine dye production estimated by a single measurement of extinction at the end of the incubation period.
