Inflammatory and Analgesic Profiles in Intervertebral Disc Herniation: Variability with Respect to Neurological Deficit.

BACKGROUND: Disc herniation is both a mechanical and biochemical process in which contact between intervertebral discs and spinal nerves causes compression, chemical irritation, inflammation, and pain. The inflammatory process is known to vary depending on pain duration, herniation type, and pain severity, but the relationship with neurological deficits remains unknown. AIM: This study aimed to compare individuals with lumbar disc herniation with and without neurological deficits (WND/WOND) and healthy individuals in terms of serum levels of tumor necrosis factor alpha (TNF-α), interleukin 6

Corticotropin-releasing hormone and proopiomelanocortin-derived peptides are present in human myometrium.

CRH and POMC-derived peptides are produced at a number of intrauterine sites in both the nonpregnant and pregnant states. It is hypothesized that CRH and POMC-derived peptides may be produced locally by the uterus to modulate myometrial contractility. This study has examined the distribution of these peptides in human uterine tissue during the ovulatory cycle and pregnancy. The immunoperoxidase staining method was used to localize CRH and POMC-derived peptides: ACTH, beta-endorphin, and alphaMSH. Immunoreactive (IR-) CRH and IR-POMC-derived peptides, beta-endorphin and alphaMSH, were observed

The possible involvement of endogenous opioid peptides and catecholestrogens in provoking menstrual irregularities in women athletes.

It is well known that women athletes engaged in strenuous physical exercise and endurance training may develop "athletic menstrual irregularities" (AMI). Although many studies have appeared dealing with the immediate endocrinological and physiological changes in these women, the underlying mechanisms have remained unknown to date. A number of hypotheses have been put forward, the most well-known among them, for example, defending the existence of a critical percent of body fat necessary to trigger or maintain normal menstruation. All these theories have, however, their own, sometimes numerous,

Endocrine systems of the skin.

Apart from its protective function, the skin is also both hormonally and metabolically active. Many hormones are produced in the skin and the regulation of these changes is controlled, as are other organs, by the pituitary gland and hypothalamus. Under the influence of stress factors such as ultraviolet radiation, many reactions are stimulated. After exposure to light in keratinocytes apart from eumelanin, also proopiomelanocortin, dopamine, and opioid substances - β-endorphins are produced. The skin also has a hypothalamic-pituitary-adrenal axis homolog. A corticotropin-releasing hormone is p…

[Changes in the consciousness state as a symptom of intestinal invagination onset].

Intussusception is the most frequent cause of intestinal occlusion in children aged 3-5 years. Diagnosis is easy to perform in cases with ty … to be performed. Some hypothesis may be considered to explain these clinical pictures: 1. increased endorphins excretion during abdominal pain; 2. neurotoxins of bacterial origin released and absorbed by altered bowel; 3. intestinal hormones abnormally produced during the disease. As recommended by Rachmel, the condition should be suspected in all children p …

Cardiovascular effects of a potent opioid peptide, (D-Met2, Pro5)-enkephalinamide.

The cardiovascular effects of morphine and a potent enkephalin analogue, D-Met2, Pro5-enkephalinamide (D-Met2, Pro5-EA) have been examined i … hese observations confirm the conclusions from the literature that, under physiological conditions, endorphins play no primary role in the regulation of the cardiovascular functions but they might be mobilized in certain cases of pathological elevation of the blood pressure probably as a part of a compensatory process.

[Acupuncture for nausea: how does it work?].

Nausea, the unpleasant feeling that one is going to vomit, results from the stimulation of the Chemoreceptor Trigger Zone (CTZ) and Vomiting Center in the brain. Conventional medical therapy is not always effective, and medications often have severe adverse effects. Acupuncture is a treatment in which thin needles (diameter of 0.20-0.30 mm) are inserted into various points along the skin, according to energy channels (meridians) established thousands of years ago. The anti-emetic effects of acupuncture apparently stem from the resultant increase in hypophyseal secretion of beta-endorphins and …

Alcoholism: scientific basis of a neuropsychogenetic disease.

Until recently alcoholism was regarded as being an incurable psychological problem. During the last decade a chain of research has led to a new insight into the causes and potential alleviation of alcohol craving: Recent discoveries indicate that the brain has receptor sites for naturally occurring opiate-like substances (endorphins and enkephalins) which are produced by the nervous system. Opiates such as morphine or heroin, and some of the metabolic products of alcohol (tetrahydroisoquinolines), can also attach themselves to these receptors. It has been further discovered that the craving fo…

The immune response to resistive breathing.

Resistive breathing is an "immune challenge" for the body, initiating an inflammatory response consisting of an elevation of plasma cytokine … late the hypothalamic-pituitary-adrenal axis, leading to production of adrenocorticotropin and beta-endorphins. The adrenocorticotropin response may represent an attempt of the organism to reduce the injury occurring in the respiratory muscles via the production of glucocorticoids and the induction of the acute phase-response proteins. The beta-endorphin res …

Glycyl-glutamine inhibits the respiratory depression, but not the antinociception, produced by morphine.

Glycyl-glutamine (Gly-Gln; beta-endorphin(30-31)) is an endogenous dipeptide that is synthesized through the posttranslational processing of beta-endorphin in brain stem regions that control respiration and autonomic function. This study tested the hypothesis that Gly-Gln administration to conscious rats will prevent the respiratory depression caused by morphine without affecting morphine antinociception. Rats were administered Gly-Gln (1-100 nmol) or saline (10 microl) intracerebroventricularly followed, 5 min later, by morphine (40 nmol icv). Arterial blood gases and pH were measured immedia

Thymic hormones, cancer and behavioural adaptive responses.

The effects of thymic hormones are not restricted within the immune system but are rather pleiotropic. Through neuropeptides the neuroendocrine system participates in the regulation of homeostasis as well as in the control of stress response and behavioural outputs. Thymic hormones increase spontaneous behaviour, inhibit anxiety-like responses and improve resistance to stress in tumour-bearing mice. In addition, thymic hormones modulate secretion of pituitary adrenocorticotrophin (ACTH) and beta-endorphin in both primates and rodents. In turn, both ACTH and beta-endorphin influence stress resp

Physiological and behavioral consequences associated with short-term prevention of crib-biting in horses.

Crib-biting in the horse is frequently prevented in the short-term by horse-owners using physical means. Because it has been proposed that crib-biting may function to reduce stress, the effect of prevention of crib-biting and/or eating on the behaviour, heart rate, and plasma cortisol and beta-endorphin concentrations was measured in six crib-biting and six normal horses. When crib-biters were unable to crib-bite, they showed an increase in ingestive behaviour. When crib-biters were prevented from crib-biting and eating, a relative stasis in the motility of the foregut occurred, suggesting tha

Postoperative pain relief in children. A comparison between caudal bupivacaine and intramuscular diclofenac sodium.

Two hundred and fifty children undergoing herniotomy or orchidopexy under general anaesthesia were randomly allocated to receive pre-operatively either diclofenac sodium 1 mg.kg-1 given intramuscularly or a caudal injection of bupivacaine 0.25% 1 ml.kg-1 with or without adrenaline or no analgesia. Plasma diclofenac and beta-endorphin concentrations were determined in eight and 21 patients respectively. Postoperative pain was assessed by ward nurses who were blinded to the group allocation. Comparison with the control group showed diclofenac to be an effective analgesic. Caudal bupivacaine prov

Behavioral, neuroendocrine, and biochemical effects of 5-hydroxytryptophan administration in panic disorder.

L-5-Hydroxytryptophan (5HTP) was administered to 20 patients suffering from panic disorder and to 20 healthy controls. Subjects received 60 mg 5HTP in 300 ml saline solution. Before, during, and up to 2 hours after 5HTP administration, symptoms of anxiety and depression were assessed. In addition, plasma 5HTP, 3-methoxy-4-hydroxyethylglycol (MHPG), cortisol, beta-endorphin, and melatonin levels were measured at several time points, and the kinetics of 5-hydroxytryptamine (5HT) in blood platelets were measured. During and after the infusion of 5HTP, none of the patients showed an increase in an

Action of an opiate receptor blocker on ovine cardiopulmonary responses to endotoxin.

The involvement of endorphins in the ovine cardiopulmonary response to endotoxin was evaluated by measuring blood levels of opiate receptor binding substances and administering the opiate receptor blocking agent naloxone prior to and after the administration of lipopolysaccharide. The animals were prepared for chronic study by placement of cardiovascular catheters and cannulation of the lung lymphatic at least 1 wk prior to study. The sheep were given endotoxin (0.75 micrograms/kg) and studied for 6 h. This was accomplished twice in the same animal. Naloxone (2 mg/kg bolus + 2 mg X kg-1 X h-1)…

A novel hypothesis: specific oncogenes and tumor suppression genes are involved in the expression of the proopiomelanocortin gene by small cell lung cancer.

The endogenous opioid beta-endorphin, a derivative of proopiomelanocortin, stimulates the growth of cloned human small cell lung carcinoma. The present hypothesis states that mutations of the retinoblastoma gene (a tumor suppressor gene) associated to the malignant transformation of bronchial cells would trigger a cascade of biomolecular events leading to 'de novo' proopiomelanocortin expression in small cell lung carcinoma.

Effects of opiates during baroreceptor and ergoreceptor induced changes in blood pressure.

Various opioids were used to investigate the role they might play in the cardiovascular responses to fatiguing isometric contractions. Changes in blood pressure were measured in cats anaesthetised with alpha-chloralose. Fatiguing isometric contractions of the hind limb muscles (ergoreceptor activation) were generated using a microprocessor controlled stimulator (50 Hz, 0.2 ms, 200-800 mV). Baroreceptor inactivation was elicited by carotid artery occlusion. Muscle contraction caused an increase in mean arterial pressure of 51 (SEM 12) mm Hg and carotid occlusion an increase of 56(9) mm Hg above
