Development of an iron overload HepG2 cell model using ferrous ammonium citrate.

Cell-based iron overload models provide tremendous utility for the investigations into the pathogenesis of different diseases as well as assessing efficacy of various therapeutic strategies. In the literature, establishing such models vary widely with regards to cell lines, iron source, iron treatment conditions and duration. Due to this diversity, researchers reported significant differences in the measured outcomes, either in cellular function or response to a stimulus. Herein, we report the process required to establish an iron overload HepG2 cell model to achieve a consistent and reproduci

Oral iron supplementation ameliorated alterations in iron uptake and utilization in copper-toxic female Wistar rats.

OBJECTIVE: Women are more susceptible to both iron deficiency and copper toxicity due to monthly flow and estrogen action, respectively. Oral iron is beneficial for menstruating women and enhances erythropoiesis, but both deficiency and excess of copper impact iron absorption and mobilization. The aim of this study was to investigate the possibility of mitigating copper toxicity in female Wistar rats while supplementing with iron. METHODS: 20 female rats (160-180g) were grouped into four: Groups 1 (Control) received 0.3mls normal saline, 2- copper-toxic (100m mg/kg Copper sulphate), 3- Copper

Yeast Mitochondria Import Aqueous FeII and, When Activated for Iron-Sulfur Cluster Assembly, Export or Release Low-Molecular-Mass Iron and Also Export Iron That Incorporates into Cytosolic Proteins.

Iron-sulfur cluster (ISC) assembly occurs in both mitochondria and cytosol. Mitochondria are thought to export a low-molecular-mass (LMM) iron and/or sulfur species which is used as a substrate for cytosolic ISC assembly. This species, called X-S or (Fe-S)int, has not been directly detected. Here, an assay was developed in which mitochondria were isolated from 57Fe-enriched cells and incubated in various buffers. Thereafter, mitochondria were separated from the supernatant, and both fractions were investigated by ICP-MS-detected size exclusion liquid chromatography. Aqueous 54FeII in the buffe

Efficient removal of chloroform from groundwater using activated percarbonate by cellulose nanofiber-supported Fe/Cu nanocomposites.

Chloroform (CF) is a recalcitrant halogenated methane (HM) that has received widespread attention due to its frequent detection in groundwater and its potential carcinogenic risk. In this study, TEMPO-oxidized cellulose nanofiber-supported iron/copper bimetallic nanoparticles (TOCNF-Fe/Cu), a novel composite catalyst, was synthesized to activate sodium percarbonate (SPC) for the removal of CF from groundwater. The results showed that over 96.3% of CF could be removed in a neutral reaction medium (pH 6.5-9) within 180 min using 0.66 g L-1 of TOCNF (0.32)-Fe/Cu (1) and 1 mM of SPC, which outperf

NIR-II-driven intracellular photocatalytic oxygen-generation on Z-Scheme iron sulfide/cobalt sulfide nanosheets for hypoxic tumor therapy.

Tumor hypoxia not only promotes the proliferation, invasion and metastasis of cancer cells but also seriously hinders photodynamic therapy (PDT). Here, second near-infrared (NIR-II) photocatalytic O2 generation is introduced to relieve hypoxia. FeS2/CoS2@PEG (FCs@PEG) nanosheets (∼80 nm) are prepared with Fe-Co layered double hydroxides (LDHs) as precursor. As-synthesized samples have great NIR-II harvest and photothermal conversion efficiency (50.5 %, 1064 nm). In addition, photothermal effect can elevate the thermal energy of nanocomposite to supply extra energy and to excite FeS2 (1.16 eV)

Thermodynamics of iron, tetrahydrobiopterin, and phenylalanine binding to phenylalanine hydroxylase from Chromobacterium violaceum.

Phenylalanine hydroxylase (PheH) is a pterin-dependent, mononuclear nonheme iron(II) oxygenase that uses the oxidative power of O2 to hydroxylate phenylalanine to form tyrosine. PheH is a member of a superfamily of O2-activating enzymes that utilizes a common metal binding motif: the 2-His-1-carboxylate facial triad. Like most members of this superfamily, binding of substrates to PheH results in a reorganization of its active site to allow O2 activation. Exploring the energetics of each step before O2 activation can provide mechanistic insight into the initial steps that support the highly spe

Carboxymethyl cellulose stabilized ferrous sulfide@extracellular polymeric substance for Cr(VI) removal: Characterization, performance, and mechanism.

Iron-based materials, especially ferrous sulfide (FeS), effectively remediate chromium pollution. However, the agglomeration of FeS reduces its reactivity to chromium. Herein, carboxymethyl cellulose stabilized ferrous sulfide@extracellular polymeric substance (CMC-FeS@EPS) was developed to remove hexavalent chromium (Cr(VI)) from water. CMC-FeS@EPS (98.00%) exhibited excellent removal efficiency of 40 mg/L Cr(VI) than those of FeS (57.35%) and CMC-FeS (68.60%). CMC-FeS@EPS showed good removal efficiency of Cr(VI) in wide pH range (from 4 to 9) and the co-existence of ions. FTIR and XPS result

Fe(II)-dosed ceramic membrane bioreactor for wastewater treatment: Nutrient removal, microbial community and membrane fouling analysis.

Ferrous dosing is used to reduce phosphorus concentration and alleviate polymeric membrane fouling in membrane bioreactor (MBR). However, limited studies have been conducted to investigate the impacts of ferrous dosing on ceramic membrane fouling, nutrient removal efficiency and microbial community. Accordingly, the aim of this study was to investigate the effect of intermittent ferrous dosing with Fe/P molar ratios of 2 and 1 (with a dosing frequency of every two days) on the overall nutrient removal, functional microbial changes and membrane fouling in ceramic membrane bioreactors (CMBR) in

A novel micro-ferrous dosing strategy for enhancing biological phosphorus removal from municipal wastewater.

Ferrous salts have been widely used to enhance phosphorus removal in full-scale wastewater treatment plants, with an average dosage of 0.24-0.35 mM. However, such high dosage inevitably caused serious concerns on operation, potential biological toxicity and excessive sludge production. Thus, this study investigated the effect of micro-dosing of ferrous salt at the level of 0.02 mM on enhanced biological phosphorus removal (EBPR) in sequencing batch reactors. Results showed that micro-dosing of ferrous salt enhanced the overall performance, with average COD, TN and TP removal of more than 4.2%,

Iron Redox Chemistry Promotes Antiparallel Oligomerization of α-Synuclein.

Brain metal dyshomeostasis and altered structural dynamics of the presynaptic protein α-synuclein (αS) are both implicated in the pathology of Parkinson's disease (PD), yet a mechanistic understanding of disease progression in the context of αS structure and metal interactions remains elusive. In this Communication, we detail the influence of iron, a prevalent redox-active brain biometal, on the aggregation propensity and secondary structure of N-terminally acetylated αS (NAcαS), the physiologically relevant form in humans. We demonstrate that under aerobic conditions, Fe(II) commits NAcαS to

Complexes with Tunable Intramolecular Ferrocene to Ti(IV) Electronic Transitions: Models for Solid State Fe(II) to Ti(IV) Charge Transfer.

Iron(II)-to-titanium(IV) metal-to-metal-charge transfer (MMCT) is important in the photosensitization of TiO2 by ferrocyanide, charge transfer in solid-state metal-oxide photocatalysts, and has been invoked to explain the blue color of sapphire, blue kyanite, and some lunar material. Herein, a series of complexes with alkynyl linkages between ferrocene (Fc) and Ti(IV) has been prepared and characterized by UV-vis spectroscopy and electrochemistry. Complexes with two ferrocene substituents include Cp2Ti(C2Fc)2, Cp*2Ti(C2Fc)2, and Cp2Ti(C4Fc)2. Complexes with a single ferrocene utilize a titanoc

Redox interactions between Cr(VI) and Fe(II) in bioreduced biotite and chlorite.

Contamination of the environment with Cr as chromate (Cr(VI)) from industrial activities is of significant concern as Cr(VI) is a known carcinogen, and is mobile in the subsurface. The capacity of Fe(II)-containing phyllosilicates including biotite and chlorite to alter the speciation, and thus the mobility, of redox-sensitive contaminants including Cr(VI) is of great interest since these minerals are common in soils and sediments. Here, the capacity of bacteria, ubiquitous in the surface and near-surface environment, to reduce Fe(III) in phyllosilicate minerals and, thus, alter their redox re

Comparison of chemical interactions with Li⁺ and catalytic reactivity of electrochemically generated [FeICl(L)]²⁻ and [CoI(L)]⁻ complexes (L = salen or salophen).

The cyclic voltammetric behavior of [FeIIICl(salen)] complexes has been investigated in CH3CN and compared to that obtained with [CoII(salen)] analogues. Details of the mechanism associated with iron- and cobalt-salen complex reduction in the presence of the lithium cation have been elucidated by comparison of simulated and experimental voltammograms. Electrogenerated [FeICl(salen)]²⁻ and [FeICl(salophen)]²⁻ complexes catalyze the dehalogenation of halo-alkyl compounds as is the case with [CoI(salen)]⁻ complexes, even in the presence of the lithium cation which allows the reduction to occur at

Biotite dissolution in brine at varied temperatures and CO2 pressures: its activation energy and potential CO2 intercalation.

For sustainable geologic CO(2) sequestration (GCS), it is important to understand the effects of temperature and CO(2) pressure on mica's dissolution and surface morphological changes under saline hydrothermal conditions. Batch experiments were conducted with biotite (Fe-end member mica) under conditions relevant to GCS sites (35-95 °C and 75-120 atm CO(2)), and 1 M NaCl solution was used to mimic the brine. With increasing temperature, a transition from incongruent to congruent dissolution of biotite was observed. The dissolution activation energy based on Si release was calculated to be 52 ±

Cr(II) reactivity of taurine/alpha-ketoglutarate dioxygenase.

The interaction of CrII with taurine/alpha-ketoglutarate (alphaKG) dioxygenase (TauD) was examined. CrII replaces FeII and binds stoichiometrically with alphaKG to the FeII/alphaKG binding site of the protein, with additional CrII used to generate a chromophore attributed to a CrIII-semiquinone in a small percentage of the sample. Formation of the latter oxygen-sensitive species requires the dihydroxyphenylalanine (DOPA) quinone form of Tyr-73. This preformed side chain is generated by intracellular self-hydroxylation of Tyr-73 to form DOPA, which is subsequently oxidized to the quinone. No ch
