Disruption of GABAergic tone in the dorsomedial hypothalamus attenuates responses in a subset of serotonergic neurons in the dorsal raphe nucleus following lactate-induced panic.

Panic patients are vulnerable to induction of panic attacks by sub-threshold interoceptive stimuli such as intravenous (i.v.) sodium lactate infusions. Facilitation of serotonergic signaling with selective serotonin reuptake inhibitors can suppress anxiety and panic-like responses, but the mechanisms involved are not clearly defined. We investigated the effects of i.v. 0.5 M sodium lactate or saline, in control and panic-prone rats on c-Fos expression in serotonergic neurons within subdivisions of the midbrain/pontine raphe nuclei. Rats were chronically infused with either the GABA synthesis i

Accuracy of cricothyroidotomy performed in canine and human cadaver models during surgical skills training.

BACKGROUND: Surgical skills training is an integral component of the Advanced Trauma Life Support (ATLS) Course. Teaching techniques are continuously reevaluated and updated. Recognition of recurring technical errors in the performance of cricothyroidotomy in canine models prompted this comparison to the performance of the procedure in human cadavers. STUDY DESIGN: Thirty-three ATLS physician students performed cricothyroidotomy in canine models. Ten flight nurses performed a bimonthly surgical skills practicum on similarly prepared animals. Neck specimens were excised, fixed, and later mappe

Klotho-deficient mice are resistant to bone loss induced by unloading due to sciatic neurectomy.

Unloading induces bone loss as seen in experimental animals as well as in space flight or in bed-ridden conditions; however, the mechanisms involved in this phenomenon are not fully understood. Klotho mutant mice exhibit osteopetrosis in the metaphyseal regions indicating that the klotho gene product is involved in the regulation of bone metabolism. To examine whether the klotho gene product is involved in the unloading-induced bone loss, the response of the osteopetrotic cancellous bones in these mice was investigated. Sciatic nerve resection was conducted using klotho mutant (kl/kl) and cont

Time-of-flight MR angiography with Gd-DTPA hexamethylene diamine co-polymer blood pool contrast agent: comparison of enhanced MRA and conventional angiography for arterial stenosis induced in rabbits.

Vascular stenoses were induced in the external iliac arteries of New Zealand white rabbits by a combination of hypercholesterolemic diet and repeat balloon injury. Two-dimensional (2D) and three-dimensional (3D) time-of-flight (TOF) magnetic resonance angiography (MRA) was performed with a specifically designed phased array coil in a 1.5 T system. Enhancement with gadolinium-diethylene triamine pentaacetic acid (Gd-DTPA) hexamethylene diamine co-polymer (Nycomed: NC 22181), a blood pool MR contrast agent, was measured after contrast administration and compared with pre-contrast images at the s

[Mechanism of vitexin in treating acute hyperlipidemia by combination of metabolomics and network pharmacology].

The study aims to investigate the mechanism of vitexin in treating acute hyperlipidemia through a combination of network pharmacology, molecular docking, and metabolomics approaches. An acute hyperlipidemia mouse model was established in mice by intramuscular injection of tyloxapol(Triton WR-1339). By using network pharmacology, the core target of vitexin in hyperlipidemia treatment was identified, and its binding affinity with the core target and stability were assessed through molecular docking. The screened core targets were validated through animal experiments to confirm their lipid-loweri

Echolocation call intensity and directionality in flying short-tailed fruit bats, Carollia perspicillata (Phyllostomidae).

The directionality of bat echolocation calls defines the width of bats' sonar "view," while call intensity directly influences detection range since adequate sound energy must impinge upon objects to return audible echoes. Both are thus crucial parameters for understanding biosonar signal design. Phyllostomid bats have been classified as low intensity or "whispering bats," but recent data indicate that this designation may be inaccurate. Echolocation beam directionality in phyllostomids has only been measured through electrode brain-stimulation of restrained bats, presumably excluding active b

The effects of spaceflight and fracture healing on distant skeletal sites.

Spaceflight results in reduced mechanical loading of the skeleton, which leads to dramatic bone loss. Low bone mass is associated with increased fracture risk, and this combination may compromise future, long-term, spaceflight missions. Here, we examined the systemic effects of spaceflight and fracture surgery/healing on several non-injured bones within the axial and appendicular skeleton. Forty C57BL/6, male mice were randomized into the following groups: (1) Sham surgery mice housed on the earth (Ground + Sham); (2) Femoral segmental bone defect surgery mice housed on the earth (Ground + Sur

Sonar detection of jittering real targets in a free-flying bat.

The auditory system measures time with exceptional precision. Echolocating bats evaluate the time delay between call and echo to measure object range. An extreme and disputed result on ranging acuity was found in the virtual delay jitter experiments. In these studies, echoes with alternating delays were played back to bats, which detected a jitter down to 10 ns, corresponding to a ranging acuity of 1.7 microm. The current study was designed to measure the ranging acuity of the nectarivorous bat Glossophaga soricina under semi-natural conditions. Three free-flying bats were trained to discrimin

Developing protocols for recombinant adeno-associated virus-mediated gene therapy in space.

With the advent of the era of International Space Station (ISS) and Mars exploration, it is important more than ever to develop means to cure genetic and acquired diseases, which include cancer and AIDS, for these diseases hamper human activities. Thus, our ultimate goal is to develop protocols for gene therapy, which are suitable to humans on the earth as well as in space. Specifically, we are trying to cure the hemoglobinopathies, beta-thalassemia (Cooley's anemia) and sickle cell anemia, by gene therapy. These well-characterized molecular diseases serve as models for developing ex vivo gene

[Activation of respiratory sinus arrhythmia in monkeys during space flight].

During the first day in space the rhesus monkey, called Vernyi, flown on Cosmos-1667 showed higher values of the daytime, night, and daily respiratory sinus arrhythmia (RSA) when compared to those observed before launch and before or the first day of a simulation study. During orbital flight, the mean daily value of RSA was in the range 13.08-16.00 beats/min. In the simulation study, this parameter increased from 4.68 beats/min during the first day to 15.16 beats/min on the 6th day. The mean RSA values during days 1 through 3 were significantly higher in real than in simulation flight. Analysi

Food waste composition, hotspots and sustainability impacts in an airline catering kitchen.

Aviation generates substantial food waste worldwide, yet the focus has mainly been on post-flight waste, leaving upstream production processes in airline catering understudied. This mixed-methods single case study investigated food waste composition, hotspots and drivers in a Swedish airline catering kitchen, and assessed associated environmental (carbon), economic (procurement cost) and social (nutritional) impacts. Over 21 weekdays, waste audits recorded 1.39 tonnes of food waste, largely edible (89%), corresponding to 3.1 tonnes of carbon dioxide equivalents, €9250 in procurement costs, and

Lévy statistics define anxiety and depression in mice subjected to chronic stress.

INTRODUCTION: Current rodent models for anxiety and depression assessment face methodological challenges compromising both scientific rigor and animal welfare. METHODS: This study introduced a novel approach using the Lévy flight (LF) statistical method to analyze spontaneous movement in open spaces. We employed three models: Chronic unpredictable mild stress (CUMS), Electric shock stress (ES), and Chronic Restraint Stress (CRS)-utilizing a total of 540 mice for LF fitting. A support vector machine algorithm was applied to distinguish each model group based on the two-dimensional distribution

Glossina from the Republic of the Congo: species identification by MALDI-TOF MS and research of associated micro-organisms.

Human African trypanosomiasis (HAT) and Animal African trypanosomiasis (AAT), transmitted by Glossina species, remain major health and economic burdens in Africa. Accurate vector identification is essential for effective control strategies. However, current identification methods of Glossina species based on morphological and/or molecular techniques have several limitations that often hinder reliable species-level classification. This study assessed matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) as an alternative or complementary approach to morphol

Flavonol glycosides accumulation in faba bean grown under combined selenium and sulfur application.

INTRODUCTION: Faba bean (Vicia faba L.) leaves are edible; hence, they are primarily used as animal feed in agriculture. Additionally, seed pods and other plant tissues are considered edible and are used as green vegetables in many parts of the world. OBJECTIVES: Flavonol glycosides are well-known in faba bean leaves; accordingly, in this study, we followed a targeted metabolomic approach to explore glycosylated flavonols and their concentrations in response to contrasting levels of selenium (Se) and sulfur (S) enrichment under faba bean-Rhizobium symbiosis. METHODS: Faba bean plants were cu

Clinical serum metabolomics study on fluoxetine hydrochloride for depression.

BACKGROUND: Fluoxetine hydrochloride is one of the familiar antidepressants of the second generation and has the effect of inhibiting the reuptake of 5-hydroxytryptamine by central nervous system. Both clinical trials and animal experiments show that it has good antidepressant effect, but there are few reports on its clinical efficacy in treating depression patients from the perspective of metabolomics. This study aimed at evaluating the antidepressant effect of fluoxetine hydrochloride by metabolomics, so that to find out its specific biomarkers and related metabolic characteristics of depres

Ablation of the cardiac ryanodine receptor phospho-site Ser2808 does not alter the adrenergic response or the progression to heart failure in mice. Elimination of the genetic background as critical variable.

BACKGROUND: Phosphorylation of the cardiac ryanodine receptor (RyR2) phospho-site S2808 has been touted by the Marks group as a hallmark of heart failure (HF) and a critical mediator of the physiological fight-or-flight response of the heart. In support of this hypothesis, mice unable to undergo phosphorylation at RyR2-S2808 (S2808A) were significantly protected against HF and displayed a blunted response to adrenergic stimulation. However, the issue remains highly controversial because several groups have been unable to reproduce these findings. An important variable not considered before is

Contact pressures in the patellofemoral joint during impact loading on the human flexed knee.

Currently, a bone fracture criterion is used by the automotive industry to assess the potential for a lower extremity injury from impact directed on the flexed knee. However, recent studies with animal models indicate irreversible damage to articular cartilage due to overpressures generated within the patellofemoral (P-F) joint without bone fracture, and suggest this injury may lead to a progressive, degenerative disease of the joint. The purpose of this investigation was to measure contact pressure in the human P-F joint during impact loading on the isolated, flexed knee. Loads were delivered
