Postmenopausal osteoporosis: treatment with low-dose sodium fluoride and estrogen.

Eleven menopausal patients were treated for 12 to 18 months with low-dose sodium fluoride and calcium. Six patients also received estrogen replacement. A significant increase in spine or hip bone mineral density measured by dual photon absorptiometry was observed in all patients. The estrogen-treated group had the greatest increase in bone density. Addition of estrogen seems to supplement bone gain and allow sodium fluoride to be administered in lower doses, which are easily tolerated and yet effective.

Supraacetabular and femoral head stress fracture during fluoride treatment.

A woman treated with fluoride for corticosteroid-induced osteoporosis presented 1 year later with an unusual localized supraacetabular followed by a same-sided femoral head fracture. Fluoride was increased in serum and urine. Transiliac bone biopsy revealed typical bone fluorosis with elevated trabecular bone fluoride.

[Fluorosis following the long-term treatment of osteoporosis with sodium fluoride].

The fluorosis in fluor-exposed workers has been known already since nearly 50 years. It is characterized by a massive regeneration of bones. The fluorosis is only rarely observed as a sequel of the sodium fluoride therapy of osteoporosis. Out of the number of our own patients a medicamentously induced fluorosis after a 12-year sodium fluoride therapy of a steroid osteoporosis is described. The pathological value of this iatrogenic fluorosis is relatively insignificant. Nevertheless, due to the picture of complaints resulting from this a sodium fluoride therapy should be controlled. In this cas

Effects of fluoride ingestion on white-tailed deer (Odocoileus virginianus).

The effects of the addition of 25 or 50 ppm fluoride (F), as sodium fluoride (NaF), to the rations of 5-mo-old male white-tailed deer were similar to those observed in domestic cattle fed similar amounts of fluoride. The ingestion of 50 ppm F for 2 yr resulted in the accumulation of over 7,000 ppm F in bone ash. Accumulation of fluoride in antlers was extensive and occurred more rapidly than in skeletal tissue. Fluoride ingestion resulted in lesions on the developing incisors that were similar, but not identical to those seen in other species. Increased molar wear in the deer fed 50 ppm F was

Mortality and cancer morbidity after heavy occupational fluoride exposure.

A cohort of 431 male cryolite workers employed for at least six months between 1924 and 1961 was identified from personnel records at the Copenhagen cryolite factory. During this period, heavy fluoride exposure resulted in at least 74 cases of skeletal fluorosis. All workmen in the cohort were followed up in Denmark until July 1, 1981. During 1941-1981, 206 men died, while only 149.3 deaths were expected from national mortality statistics. Significant excesses were seen in the following causes of death: violent death and all cancers, in particular cancer of the respiratory system. When compare

Aluminum: a requirement for activation of the regulatory component of adenylate cyclase by fluoride.

Activation of the purified guanine nucleotide-binding regulatory component (G/F) of adenylate cyclase by F- requires the presence of Mg2+ and another factor. This factor, which contaminates commercial preparations of various nucleotides and disposable glass test tubes, has been identified as Al3+. In the presence of 10 mM Mg2+ and 5 mM F-, AlCl3 causes activation of G/F with an apparent activation constant of approximately 1-5 muM. The requirement for Al3+ is highly specific; of 28 other metals tested, only Be2+ promoted activation of G/F by F-.

Glycogen accumulation in the parathyroid gland of the rat after fluoride ingestion.

The parathyroid glands of young male rats given 150 ppm fluoride in their drinking water for 10 weeks were examined by transmission electron microscopy. As a result of fluoride ingestion, the parathyroid chief cells of the experimental animals accumulated glycogen in excess of that seen in control animals given distilled drinking water for the same time period. In the majority of active chief cells, glycogen granules were diffusely spread throughout the cytoplasm as single granules or in small deposits. Large aggregations of glycogen granules were also seen within intercellular spaces. Accompa

An investigation of methods proposed for the analysis of the various forms of fluoride in toothpaste.

Analytical methods for the determination of soluble fluoride, soluble fluorine and total fluorine derivatives in toothpaste were investigated. Potentiometric determinations of soluble fluoride in TISAB-toothpaste slurries were shown to be inaccurate. Based on recovery measurements, analyses for soluble fluoride and soluble fluorine derivatives in the supernatant liquid were found to be reliable provided the slurrying time was standardized. A gas chromatography method for the analysis of total fluorine, using the silane derivative fluoride, was compared with methods involving ashing and distill

Fluoride concentrations in fissure and cervical enamel of unerupted human teeth.

Fluoride concentration of fissure enamel from unerupted third molars was higher than that of the cervical-lingual but not cervical-buccal surfaces at a similar depth of the removed enamel. The highest fluoride concentration was found in the fissure enamel due probably to its earlier formation, higher permeability and embedding tissue before eruption.

Antimicrobial effect of fluorides on Bacteroides melaninogenicus subspecies and Bacteroides asaccharolyticus.

This study was undertaken to determine whether the suspected periodontopathogens Bacteroides melaninogenicus ss melaninogenicus and intermedius and Bacteroides asaccharolyticus are killed by clinically used topical fluorides. The viability of each organism was examined after exposure to three fluoride compounds, stannous (SnF2), acidulated phosphate (APF), and neutral sodium (NaF), for 1 and 8 h. Stannous fluoride was found to be effective at lowest fluoride concentrations with APF next in effectiveness. NaF required the highest concentrations of fluoride to kill test organisms. Longer exposur

An investigation of fluoride protection against dietary induced osteoporosis in the rat.

The purpose of this study was to determine whether the accumulation of substantial levels of fluoride in the skeleton during bone formation would significantly reduce, or prevent, the occurrence of osteoporosis experimentally induced at a later time with a low mineral-containing diet. Male weanling rats were pretreated for 15 weeks with either 0 or 50 ppm F in their drinking water, followed by treatment for four to twelve weeks with diets either optimal or deficient in both Ca and P. Intake of the Ca and P deficient diet resulted in a progressive loss of bone mass (per unit volume of femur), w

Inhibitory effect of fluoride on glucose tests with glucose oxidase strips.

Sodium fluoride was inadvertently added as a preservative to the urine of an eight-year-old boy with diabetes mellitus before urinary glucose was measured. On preliminary screening of the urine, the test by glucose oxidase paper reagent strip gave a negative reading for glucose, whereas quantitative urinary glucose assay by the coupled enzyme reaction (hexokinase-glucose 6-phosphate dehydrogenase) gave a glucose concentration of 81.5 g/liter. Inadvertent use of sodium fluoride as a urine preservative may cause a falsely negative result with the glucose tests involving oxidase.

Comparative Evaluation of Remineralization Efficacy of Nanosilver Fluoride and Xylitol-coated Calcium Phosphate Fluoride Varnishes Using Atomic Force Microscopy and DIAGNOdent®: An In Vitro Study.

BACKGROUND: In recent years, noninvasive caries treatments have drawn increased attention from the scientific community. Fluoride varnishes are topical agents that protect the teeth from caries, as they have a prolonged effect on the duration of fluoride and tooth enamel contact. AIM: To utilize DIAGNOdent® and Atomic Force Microscopy (AFM) and compare the effectiveness of sodium fluoride, nanosilver fluoride, and xylitol-coated calcium phosphate fluoride varnishes on remineralization. MATERIALS AND METHODS: To create artificial carious lesions, 30 samples of prepared enamel were placed for
