Plant-inspired gallolamine catalytic surface chemistry for engineering an efficient nitric oxide generating coating.

UNLABELLED: A novel concept of generating therapeutic gas, nitric oxide (NO) via catalytic phenolic-amine "gallolamine" surface chemistry is developed. The concept is realized using plant polyphenol, gallic acid, and a glutathione peroxidase-like organoselenium compound cystamine or selenocystamine through one-step phenol-amine molecular assembling process. The resulting NO-generating coating with phenolic-cystamine or -selenocystamine framework showed the ability for long-term, steady and controllable range of NO release rates being unparalleled with any existing NO-releasing or NO-generating…

Development of active bioplastic films from sodium alginate/polyvinyl alcohol-polyethylene glycol copolymer incorporating syringic acid for postharvest mushroom preservation.

The development of biodegradable active bioplastic films has gained tremendous attention as a sustainable alternative to plastic based packaging. In this study, active bioplastic films were developed using sodium alginate (SA) and polyvinyl alcohol-polyethylene glycol (PVA-PEG) copolymer, incorporating syringic acid (SG) for mushroom preservation. The films were prepared via solvent casting, and the addition of SG (0.25% w/v) significantly (p < 0.05) improved tensile strength and surface hydrophobicity. The scanning electron microscopy and 3D profilometry of the finalized film revealed uniform

Restoring mucosal barrier homeostasis by in situ formation of a living-synthetic therapeutic coating.

The mucosal barrier consisting of physicochemical, immune, and microbial components is the first line of defense against external stimuli. Breakdown of the mucosal barrier causes the occurrences of various diseases, while methods capable of multifacetedly restoring mucosal barrier functions have been rarely reported. Here, we describe the restoration of the physicochemical, immune, and microbial homeostasis of the mucosal barrier by in situ formation of a living-synthetic therapeutic coating (LSTC). Through metal-phenolic complexation and π-π stacking interactions, ethyl gallate can chelate Bi

The eNOS-induced leonurine's new role in improving the survival of random skin flap.

In reconstructive and plastic surgery, random skin flaps are commonly utilized to treat skin abnormalities produced by a variety of factors. Flap delay procedure is commonly used to reduce flap necrosis. Due to the limitations of various conditions, the traditional surgical improvement can't effectively alleviate the skin flap necrosis. And leonurine (Leo) has antioxidant and anti-inflammatory effects. In this study, we researched the mechanism underlying the influences of varied Leo concentrations on the survival rate of random skin flaps. Our results showed that after Leo treatment, tissue e

Leonurine alleviates ferroptosis in cisplatin-induced acute kidney injury by activating the Nrf2 signalling pathway.

BACKGROUND AND PURPOSE: Evidence indicates that ferroptosis plays a key role in acute kidney injury induced by cisplatin. The Nrf2/NRF2 pathway regulates oxidative stress, lipid peroxidation and positively regulates cisplatin-induced acute kidney injury, but its effect along with the alkaloid leonurine, found in motherwort, on ferroptosis after such acute kidney injury remains unclear. EXPERIMENTAL APPROACH: The anti-ferroptotic effects of Nrf2 and leonurine were assessed in a mouse model of cisplatin-induced acute kidney injury. In vitro, the effects of leonurine on erastin- and RSL3-induced

The Size Flexibility of Ferritin Nanocage Opens a New Way to Prepare Nanomaterials.

Ferritins are ubiquitous iron storage proteins where Fe(II) sequestration prevents not only its spontaneous oxidation to Fe(III) but also production of toxic free radicals. Recently, scientists have subverted these nature functions and used ferritin cage structures of nanometer dimensions for encapsulation of guest molecules such as anti-cancer drugs or bioactive nutrients based on pH induced ferritin disassembly and reassembly property. However, prior to this study, ferritin nanocage was required to disassemble only under harsh pH conditions (≤2.0 or ≥11.0), followed by reassembly at near neu

The effect of the calcium ion antagonist 8-(N,N-diethylamino)-octyl-3,4,5-trimethoxybenzoate on malignant hyperpyrexia-susceptible porcine skeletal muscle.

In malignant hyperpyrexia susceptible (MHS) porcine skeletal muscle, a low concentration (100 mumol/l) of the calcium ion antagonist 8-(N,N-diethylamino)-octyl-3,4,5-trimethoxybenzoate (TMB-8) inhibited KCl-induced contractures, but potentiated contractures induced by halothane, caffeine and succinylcholine. Higher concentrations of TMB-8 (333 mumol/l to 1 mmol/l) contracted MHS muscle, but had little effect on muscle tension in control preparations. Treatments which inhibit excitation-contraction coupling abolished TMB-8-induced hyper-reactivity in MHS muscle. TMB-8 (50 mumol/l and 1 mmol/l)

Calcium requirements for increased complement receptor expression during neutrophil activation.

It has recently been shown that human neutrophils rapidly increase surface expression of membrane receptors for C3b (CR1) and C3bi (CR3) in response to chemoattractants and other stimuli. In the present studies, we used monoclonal antibodies and flow cytometry to assess the role of Ca2+ in this process. Stimulation with ionophore A23187 in the presence of 1.2 mM Ca2+ increased CR1 330% and CR3 650% compared with unstimulated cells at 37 degrees C. Because this indicated that increasing the cytosolic free Ca2+ caused increased receptor expression, we examined the role of Ca2+ in the response to

Thiopurine methyltransferase. Aromatic thiol substrates and inhibition by benzoic acid derivatives.

Thiopurine methyltransferase (TPMT) catalyzes the S-methylation of thiopurine and thiopyrimidine drugs. If potent TPMT inhibitors were available, studies of the regulation and properties of this drug-metabolizing enzyme would be facilitated. Each of a series of benzoic acid derivatives tested was found to inhibit purified human kidney TPMT. Concentrations required to inhibit TPMT by 50% ranged from 20 microM for 3,4-dimethoxy-5-hydroxybenzoic acid to 2.1 mM for acetylsalicylic acid. Inhibition was noncompetitive or mixed with respect to both S-adenosyl-L-methionine, the methyl donor for the en

Studies on survival of bacteria: rhythmic response of microorganisms to freeze-drying additives.

Various materials were mixed with suspensions of Serratia marcescens and other organisms. Samples were removed and frozen at intervals after mixing; the number of cells that survived both freeze-drying and exposure to air varied rhythmically as a function of time between mixing and freezing. When assayed before or immediately after drying there were essentially no fluctuations. The response was evident only when these dried samples were exposed to air. In a typical experiment, the number of cells surviving in the sample frozen 30 sec after adding propyl gallate was at least 10 times that in sa
