The nucleoid-associated protein subunit HupA positively regulates the Pqs system and pyocyanin production in Pseudomonas aeruginosa.

Pseudomonas aeruginosa, a ubiquitous opportunistic pathogen, coordinates virulence gene expression primarily through three quorum-sensing (QS) systems: Las, Rhl, and Pqs. Despite the functions of QS systems in P. aeruginosa being relatively well-documented, the regulatory mechanisms modulating their activities remain largely unexplored. Here, we isolated and characterized a Las-null strain, PA_HN008, which overproduces pyocyanin (PYO) at the late growth stage due to its active RhlI-RhlR-PqsE pathway. Leveraging PYO production as a phenotypic marker, we conducted a genome-wide screening for ups

FlgJ cell-wall hydrolyzing activity enhances, but is not required, for flagellum assembly in Salmonella enterica.

Penetration of the peptidoglycan (PG) layer by the nascent flagellar rod is a critical step in basal body assembly and has long been attributed to the acetylglucosaminidase activity of the flagellar rod cap protein FlgJ. Previous work in Salmonella enterica suggested that occasional preexisting openings in the PG layer allow some flagella to assemble in the absence of the FlgJ enzymatic activity. More recently, studies in Bacillus subtilis demonstrated that membrane mobility of nascent flagellar structures enables rod penetration without dedicated PG hydrolysis. Here, we revisited the requirem

Resilient Communities: The Role of Biofilms in Salmonella enterica Ecology, Persistence and Pathogenesis.

Salmonella enterica is a major global foodborne pathogen whose success across environmental, agricultural and host-associated niches is closely linked to its ability to form biofilms. Importantly, biofilm formation capacity varies substantially among S. enterica serovars, with broad-host-range serovars such as Salmonella typhimurium and Salmonella enteritidis typically exhibiting robust curli- and cellulose-based biofilms, while host-restricted serovars and recently evolved invasive lineages may display reduced or altered biofilm phenotypes. In this review, we synthesise current knowledge on t

Photosynthesis in Synechocystis sp. PCC 6803 is not optimally regulated under very high CO2.

One strategy for CO2 mitigation is using photosynthetic microorganisms to sequester CO2 under high concentrations, such as in flue gases. While elevated CO2 levels generally promote growth, excessively high levels inhibit growth through uncertain mechanisms. This study investigated the physiology of the cyanobacterium Synechocystis sp. PCC 6803 under very high CO2 concentrations and yet stable pH around 7.5. The growth rate of the wild type (WT) at 200 µmol photons m-2 s-1 and a gas phase containing 30% CO2 was 2.7-fold lower compared to 4% CO2. Using a CRISPR interference mutant library, we i

Response to Nutrient Stress in the Industrial Model Bacterium Cupriavidus necator: A Thermal Proteome Profiling (TPP) Investigation.

The facultative chemolithoautotroph Cupriavidus necator is capable of heterotrophic growth on diverse carbon sources, or of autotrophic growth using CO2 fixation with H2 as an energy source. Under stress conditions, it produces biodegradable polyesters (polyhydroxyalkanoates, PHAs) as a storage material occupying a high proportion of the total biomass. This metabolic versatility means that C. necator is under intense study for sustainable biotechnology processes; however, a relative lack of understanding of the overall regulatory architecture has limited its application. The major mechanisms b

Demethylation of methylguanidine by a stepwise dioxygenase and lyase reaction.

Guanidine-responsive riboswitches control genes that enable either detoxification or assimilation of guanidino compounds. In Vreelandella boliviensis and other halophilic bacteria, genes encoding the guanidine carboxylase pathway are found in a guanidine riboswitch-regulated operon, along with two uncharacterized genes annotated as 2-oxoglutarate (2-OG/Fe(II))-dependent dioxygenase family protein and hypothetical protein, respectively. Here we show that the 2-OG/Fe(II)-dependent dioxygenase efficiently hydroxylates methylguanidine. The resulting N-(hydroxymethyl)guanidine constitutes an unexpe

Positive feedback regulation between RpoS and BosR in the Lyme disease pathogen.

In Borrelia burgdorferi, the causative agent of Lyme disease, differential gene expression is primarily governed by the alternative sigma factor RpoS (σS). Understanding the regulation of RpoS is crucial for elucidating how B. burgdorferi is maintained throughout its enzootic cycle. Our recent studies have shown that the homolog of Fur/PerR repressor/activator BosR functions as an RNA-binding protein that controls the rpoS mRNA stability. However, the mechanisms regulating BosR, particularly in response to host signals and environmental cues, remain largely unclear. In this study, we uncovered

Exogenous α-linolenic acid acts as a negative regulator of Cronobacter sakazakii structural flagellar proteins.

AIM: This work aims to report on the mechanism of action by which Cronobacter sakazakii virulence is impacted by α-linolenic acid (ALA), a C18:3 fatty acid. METHODS AND RESULTS: To elucidate this, two concentrations of ALA (250 and 1000 µmol L-1) were added exogenously to C. sakazakii 29 544 in tryptic soy broth. Quantitative proteomic analysis using label-free mass spectrometry showed significantly different proteomic profiles of treated and control C. sakazakii samples. Across both ALA treatments, a total of 11 flagellar proteins were identified as reduced in abundance and showed a dose-dep

Overexpressing the ClpC AAA+ unfoldase accelerates developmental cycle progression in Chlamydia trachomatis.

Chlamydia is an obligate intracellular bacterium that undergoes a complex biphasic developmental cycle, alternating between the smaller, infectious, non-dividing elementary body (EB) and the larger, non-infectious but dividing reticulate body. Due to the differences between these functionally and morphologically distinct forms, we hypothesize protein degradation is essential to chlamydial differentiation. The bacterial Clp system, consisting of an ATPase unfoldase (e.g., ClpX or ClpC) and a proteolytic component (e.g., ClpP), is critical for the physiology of bacteria through its recognition,

Infection-associated gene regulation of L-tartrate metabolism in Salmonella enterica serovar Typhimurium.

UNLABELLED: Enteric pathogens such as Salmonella enterica serovar Typhimurium experience spatial and temporal changes to the metabolic landscape throughout infection. Host reactive oxygen and nitrogen species non-enzymatically convert monosaccharides to alpha hydroxy acids, including L-tartrate. Salmonella utilizes L-tartrate early during infection to support fumarate respiration, while L-tartrate utilization ceases at later time points due to the increased availability of exogenous electron acceptors such as tetrathionate, nitrate, and oxygen. It remains unknown how Salmonella regulates its g

Implication of the σE Regulon Members OmpO and σN in the ΔompA299-356-Mediated Decrease of Oxidative Stress Tolerance in Stenotrophomonas maltophilia.

Outer membrane protein A (OmpA) is the most abundant porin in bacterial outer membranes. KJΔOmpA299-356, an ompA C-terminal in-frame deletion mutant of Stenotrophomonas maltophilia KJ, exhibits pleiotropic defects, including decreased tolerance to menadione (MD)-mediated oxidative stress. Here, we elucidated the underlying mechanism of the decreased MD tolerance mediated by ΔompA299-356. The transcriptomes of wild-type S. maltophilia and the KJΔOmpA299-356 mutant strain were compared, focusing on 27 genes known to be associated with oxidative stress alleviation; however, no significant differe

Euo is a developmental regulator that represses late genes and activates midcycle genes in Chlamydia trachomatis.

In this study, we developed a correlative approach that combined DNA immunoprecipitation-seq and RNA-seq analyses to define the regulon of the Chlamydia trachomatis transcription factor Euo. We confirmed the proposed role of Euo as a transcriptional repressor of late chlamydial genes but also showed that Euo activates transcription of a subset of midcycle genes and autoregulates its own expression via negative feedback. This study validates and expands the role of Euo as an important developmental regulator in C. trachomatis. In addition, this genome-wide correlative approach can be applied to

The Outer Surface Protease, SepM, Is Required for blp Locus Activation in Three of the Four Most Common Pherotypes of Streptococcus pneumoniae.

Streptococcus pneumoniae (pneumococcus) is an important human pathogen that primarily resides in the nasopharynx. To persist in this polymicrobial environment, pneumococcus must compete with other members of the bacterial community. Competition is mediated in part by the action of the blp locus, which encodes a variable array of bacteriocins and their associated immunity proteins. The locus is controlled by a two-component regulatory system that senses the extracellular concentration of the peptide pheromone, BlpC. There are four major pherotypes of BlpC that can be found in most pneumococcal

A Novel XRE-Type Regulator Mediates Phage Lytic Development and Multiple Host Metabolic Processes in Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a Gram-negative opportunistic pathogen, the leading cause of acute and chronic infections in immunocompromised patients, frequently with high morbidity and mortality rates. The xenobiotic response element (XRE) family proteins are the second most common transcriptional regulators (TRs) in P. aeruginosa. However, only a few XRE-like TRs have been reported to regulate multiple bacterial cellular processes, encompassing virulence, metabolism, antibiotic synthesis or resistance, stress responses, and phage infection, etc. Our understanding of what roles these XRE-like sma

Effect of Zinc on Microcystis aeruginosa UTEX LB 2385 and Its Toxin Production.

Cyanobacteria harmful algal blooms (CHABs) are primarily caused by man-made eutrophication and increasing climate-change conditions. The presence of heavy metal runoff in affected water systems may result in CHABs alteration to their ecological interactions. Certain CHABs produce by-products, such as microcystin (MC) cyanotoxins, that have detrimentally affected humans through contact via recreation activities within implicated water bodies, directly drinking contaminated water, ingesting biomagnified cyanotoxins in seafood, and/or contact through miscellaneous water treatment. Staphylococcus aureus Preferentially Liberates Inorganic Phosphate from Organophosphates in Environments where This Nutrient Is Limiting.

Phosphate is an essential nutrient that Staphylococcus aureus and other pathogens must acquire from the host during infection. While inorganic monophosphate (Pi) is the preferred source of this nutrient, bacteria can also obtain it from phosphate-containing organic molecules. The Pi-responsive regulator PhoPR is necessary for S. aureus to cause infection, suggesting that Pi is not freely available during infection and that this nutrient must be obtained from other sources. However, the organophosphates from which S. aureus can obtain phosphate are unknown. We evaluated the ability of 58 phosph

Metal-responsive RNA polymerase extracytoplasmic function (ECF) sigma factors.

In order to survive, bacteria must adapt to multiple fluctuations in their environment, including coping with changes in metal concentrations. Many metals are essential for viability, since they act as cofactors of indispensable enzymes. But on the other hand, they are potentially toxic because they generate reactive oxygen species or displace other metals from proteins, turning them inactive. This dual effect of metals forces cells to maintain homeostasis using a variety of systems to import and export them. These systems are usually inducible, and their expression is regulated by metal senso

MS2-Affinity Purification Coupled With RNA Sequencing Approach in the Human Pathogen Staphylococcus aureus.

Staphylococcus aureus is a Gram-positive major human pathogen involved in a wide range of human infectious diseases (from minor skin infections to septicemia, endocarditis or toxic shock syndrome). The treatment of S. aureus infections is very challenging due to the emergence of multiple antibiotic-resistant isolates. The high diversity of clinical symptoms caused by S. aureus depends on the precise expression of numerous virulence factors and stress response pathways, which are tightly regulated at every level (transcriptional, posttranscriptional, translational, and posttranslational). Iron and Fur in the life cycle of the zoonotic pathogen Vibrio vulnificus.

In this study, we aimed to analyze the global response to iron in the broad-range host pathogen Vibrio vulnificus under the hypothesis that iron is one of the main signals triggering survival mechanisms both inside and outside its hosts. To this end, we selected a strain from the main zoonotic clonal-complex, obtained a mutant in the ferric-uptake-regulator (Fur), and analyzed their transcriptomic profiles in both iron-excess and iron-poor conditions by using a strain-specific microarray platform. Among the genes differentially expressed, we identified around 250 as putatively involved in viru

Multiple mutations lead to MexXY-OprM-dependent aminoglycoside resistance in clinical strains of Pseudomonas aeruginosa.

Constitutive overproduction of the pump MexXY-OprM is recognized as a major cause of resistance to aminoglycosides, fluoroquinolones, and zwitterionic cephalosporins in Pseudomonas aeruginosa. In this study, 57 clonally unrelated strains recovered from non-cystic fibrosis patients were analyzed to characterize the mutations resulting in upregulation of the mexXY operon. Forty-four (77.2%) of the strains, classified as agrZ mutants were found to harbor mutations inactivating the local repressor gene (mexZ) of the mexXY operon (n = 33; 57.9%) or introducing amino acid substitutions in its produc
