Ammonium-mediated reduction in salicylic acid content and recovery of plant growth in Arabidopsis siz1 mutants is modulated by NDR1 and NPR1.

The siz1 mutants exhibit high SA accumulation and consequently severe dwarfism. Although siz1 mutants exhibit growth recovery upon exogenous ammonium supply, the underlying mechanism remains unknown. Here, we investigated the effect of ammonium on SA level and plant growth in SA-accumulating mutants. The growth of siz1-2 and siz1-3 mutants was recovered to wild-type (WT) levels upon exogenous ammonium supply, but that of siz1-3 ndr1 (non-race-specific disease resistance 1) and siz1-3 npr1 (non-expressor of pathogenesis related gene 1) double mutants was unaffected. The SA level was decreased b

Mediation of Flower Induction by Gibberellin and its Inhibitor Paclobutrazol: mRNA and miRNA Integration Comprises Complex Regulatory Cross-Talk in Apple.

Guaranteeing successful flowering is very important in economic plant species, especially apple (Malus domestica Borkh.), which is difficult to induce to flower. However, the gene expression and networks involved in flowering have not been totally characterized. Here, we employed mRNA and microRNA (miRNA) sequencing to understand the different responses to gibberellin- and its inhibitor paclobutrazol- (PAC) mediated flower induction. Significant opposite cytological and morphological changes were observed in treated terminal buds, which led to a reduced flowering rate under gibberellin and an

Isolation of a novel ABC-transporter gene from soybean induced by salicylic acid.

This paper reports on the identification and characterization of a new ATP-binding cassette (ABC) transporter which was identified as a salicylic acid-induced gene from soybean (Glycine max cv. Williams 82) in a subtractive suppression hybridization approach. A fragment of an ABC-transporter gene was used to isolate a full-length cDNA clone for this gene with a length of 4750 bp. The encoded protein has a length of 1447 amino acids and is composed of two similar repeat units typical of full-size ABC transporters. The sequence displays a close relationship to plant pleiotropic drug resistance (

A Pangenome Analysis of the RPD3/HDA1-Type Histone Deacetylase (HDAC) Family in Musaceae Unveils MaHDAC3 as an Epigenetic Repressor of Fruit Ripening.

Histone deacetylases (HDACs) are critical epigenetic modulators of fruit ripening. In plants, the RPD3/HDA1 subfamily is the most evolutionarily dynamic and functionally diverse group within the HDAC superfamily. To decipher how the RPD3/HDA1 subfamily coordinates fruit ripening, we conducted a pangenomic investigation of these members across 18 Musaceae genomes. A total of 199 RPD3/HDA1-type members were identified, with 20 representatives unveiling a distinct subcellular landscape. Evolutionary analyses revealed a dichotomy within the subfamily: Class-I expanded via multiple duplications, wh

The ZmmiR169o/ZmNF-YA13 module orchestrates maize kernel texture by fine-tuning starch biosynthesis and endosperm architecture.

BACKGROUND: Kernel texture is a key agronomic trait determining grain quality and storage stability in maize. The ratio of vitreous endosperm is the principal metric for kernel texture, and elucidating the key genes governing its formation is vital for maize breeding. Although microRNAs (miRNAs) play important regulatory roles in seed development, their functions in controlling grain quality traits, especially endosperm texture, remain poorly understood. RESULTS: Here, we systematically dissect the regulatory role of the ZmmiR169o/ZmNF-YA13 module in maize kernel texture. Our results show tha

Molecular regulation of plant endoreduplication: core mechanisms and their upstream control.

Endoreduplication, or the endocycle, is a widespread developmental program in plants, in which cells repeatedly replicate their nuclear genome without cell division, leading to endopolyploidy. Unlike endomitosis or chemically induced polyploidization, this process arises from an intrinsic reconfiguration of the cell cycle machinery that uncouples G1/S progression from M-phase entry. A useful conceptual framework for this regulation is the "endocycle oscillator", a periodic control logic that supports repeated rounds of DNA replication while bypassing mitosis. Studies in animal systems, particu

Breaking the bloom: Exploring the genetic pathways in crop plants from primordia formation to seed development.

Genetic regulation of fertility and sterility is a key determinant of reproductive success, seed formation, and hybrid breeding efficiency in crop plants. Modern genetics has encouraged us to analyze the genetic and molecular pathways of plant crops and discuss the respective roles of genes. The concept of modern genetics is so vast, and it has opened new doors in scientific research, like the introduction and analysis of several genes that direct plant developmental processes toward either fertility or sterility. Normal plant development depends on coordinated genetic regulation of reproducti

TSlWRKY6 mediates H2S signaling and SlGRF1-SlGIF2 module to coordinately regulate plant growth, fruit yield, and ripening in tomato.

Hydrogen sulfide (H2S), a gaseous signaling molecule, retards plant senescence and fruit ripening via protein persulfidation. However, the molecular mechanism underlying its coordination of fruit ripening with plant growth and development remains undefined. This study found that H2S could delay tomato fruit ripening and leaf senescence, and H2S defective mutant sllcd1 shows premature fruit ripening and stunted growth. Overexpression of SlWRKY6, which could be persulfidated by H2S, promotes fruit ripening and reduces fruit number. Molecular assays demonstrated that SlWRKY6 directly binds to the

Metabolic Enzyme MeHNL11 Regulates MeCAS1b Transcription for Cyanide Reutilization in Response to Nitrate Deficiency in Cassava.

Cassava (Manihot esculenta Crantz) exhibits exceptional tolerance to infertile soils and contains abundant cyanogenic glucosides (CGs). Previous research has indicated that CGs can serve as a significant reservoir of organic nitrogen in plants. However, the extent to which its high-CG content contributes to efficient nitrogen utilisation and adaptation to low nitrogen (N) in cassava remains to be further elucidated. This study represents the first identification of MeHNL11 as a bifunctional protein. In response to N deficiency, the hydroxynitrile lyase activity of MeHNL11 promotes the generati

Phosphate starvation response 1 (PHR1): a versatile master regulator shaping plant resilience beyond phosphate deprivation.

In the last two decades, the transcription factor phosphate starvation response 1 (PHR1) and its homologues in Arabidopsis and other plant species have emerged as undisputed master regulators of phosphorus starvation response (PSR). The role of Suppressor of Yeast Gpa1 (SYG1)/PHOsphate metabolism 81 (PHO81)/Xenotropic and Polytropic Retrovirus receptor 1 (XPR1) (SPX) proteins as negative regulators of PHR1 activity, and binding of this transcription factor to the P1BS element of the target phosphate starvation-inducible genes during the regulation of PSR, is well established. Given the central

Insights into hydrogen sulfide defense against black rot of cabbage at morphological, cellular and transcriptional metabolic levels.

Black rot caused by Xanthomonas campestris pv. campestris (Xcc) has led to severe economic losses in cruciferous crops. Hydrogen sulfide (H2S), an important gaseous signaling molecule, is involved in mediating plant defense responses to various adversities. Therefore, we investigated the mechanism of defense regulation against Xcc in cabbage using exogenous H2S (1 mmol/L NaHS, H2S donor) and determined that hydrogen sulfide was effective in controlling black rot. The results showed that Xcc inhibited the antioxidant properties and secondary metabolite synthesis of cabbage, leading to significa

Identification and characterization of yellow stripe-like genes in maize suggest their roles in the uptake and transport of zinc and iron.

BACKGROUND: Yellow Stripe-Like (YSL) proteins are involved in the uptake and transport of metal ions. They play important roles in maintaining the zinc and iron homeostasis in Arabidopsis, rice (Oryza sativa), and barley (Hordeum vulgare). However, proteins in this family have not been fully identified and comprehensively analyzed in maize (Zea mays L.). RESULTS: In this study, we identified 19 ZmYSLs in the maize genome and analyzed their structural features. The results of a phylogenetic analysis showed that ZmYSLs are homologous to YSLs of Arabidopsis and rice, and these proteins are divid

A DUF21 domain-containing protein regulates plant dwarfing in watermelon.

Dwarf or semidwarf plant structures are well suited for intensive farming, maximizing yield, and minimizing labor costs. Watermelon (Citrullus lanatus) is classified as an annual vine plant with elongated internodes, yet the mechanism governing watermelon dwarfing remains unclear. In this study, a compact watermelon mutant dwarf, induced by the insertion of transferred DNA (T-DNA), was discovered. Through resequencing, a gene named domain of unknown function 21 (ClDUF21), located downstream of the T-DNA insertion site, was identified as the candidate gene for the dwarf mutant, and its function

Exogenous application of Eucommia ulmoides β-1, 4-glucanase promotes propagation by increasing the expression of wound healing genes.

Eucommia ulmoides (E. ulmoides) is a valuable gum-producing plant and traditional Chinese medicine. The utilization value of E. ulmoides varies according to the sex of the plant, and due to its perennial characteristics, the identification of male and female is challenging. To meet production demands, gender selection through grafting has been employed. The fusion of rootstock and scion cells in grafted plants can be enhanced by β-1, 4-glucanase, thus improving the grafting survival rate. However, extracting β-1, 4-glucanase in vivo poses difficulties. In this study, the β-1, 4-glucanase gene

CsCBF2 contributes to cold repression of chlorophyll and carotenoid biosynthesis in albino Camellia sinensis cv. Baiye 1.

C-repeat binding factors (CsCBFs) play a pivotal role in regulating cold response in higher plants. Camellia sinensis cv. Baiye 1, a representative albino tea cultivar, has been identified as temperature-sensitive based on long-term observations by tea farmers. However, it remains unclear whether CsCBFs are involved in temperature-mediated albinism and seasonal greening in 'Baiye 1', and the mechanisms by which CBFs regulate cold responses in albino leaves are unknown. In this study, we demonstrate that CsCBF2 suppresses the seasonal greening of albino leaves by inhibiting chlorophyll and caro

Changes in the expression pattern of OsWUS negatively regulate plant stature and panicle development in rice.

WUSCHEL (WUS) and WUSCHEL-RELATED HOMEOBOX (WOX) encode transcription factors and play important roles in regulating the formation and maintenance of shoot and floral meristems. OsWUS have distinct functions in meristem development with slightly tuned expression. However, the mechanisms regulating the specific expression of OsWUS need to be further explored. In this study, an abnormal expression mutant of OsWUS, called Dwarf and aberrant panicle 1 (Dap1) was used. In order to identify the causal gene in Dap1, high-efficiency thermal asymmetric interlaced (hiTAIL)-PCR and co-segregation analysi

Techniques to Study Common Root Responses to Beneficial Microbes and Iron Deficiency.

Iron (Fe) plays a central role in the vital processes of a plant. The Fe status of a plant influences growth and immunity, but it also dictates interactions of roots with soil microbiota through the production of Fe mobilizing, antimicrobial fluorescent phenolic compounds called coumarins. To adapt to low Fe availability in the soil, plants deploy an efficient Fe deficiency response. Interestingly, this Fe deficiency response is hijacked by root-colonizing microbes in the root microbiome to establish a mutually beneficial relationship. In this chapter, we describe how we cultivate plants and m
