The cognitive cost of age-related hearing loss.

BACKGROUND: Age-related hearing loss (ARHL) or presbycusis is a bilateral sensorineural hearing impairment associated with aging of the structures of the inner ear and auditory pathways and is the most common cause of acquired hearing loss in older adults. Presbycusis is a disabling condition that affects communication and quality of life, and has been associated with poorer cognitive performance. This study aimed to examine the association between ARHL, cognitive screening performance, and hearing-related quality of life in older adults. METHODS: This single-center prospective observational

Novel, pathogenic insertion variant of GSDME associates with autosomal dominant hearing loss in a large Chinese pedigree.

Nonsyndromic hearing loss (NSHL) is a genetically diverse, highly heterogeneous condition characterised by deafness, and Gasdermin E (GSDME) variants have been identified as directly inducing autosomal dominant NSHL. While many NSHL cases associated with GSDME involve the skipping of exon 8, there is another, less understood pathogenic insertion variant specifically found in Chinese pedigrees that causes deafness, known as autosomal dominant 5 (DFNA5) hearing loss. In this study, we recruited a large Chinese pedigree, conducted whole-exome and Sanger sequencing to serve as a comprehensive clin

A novel missense mutation in SMPX causes a rare form of X-linked postlingual sensorineural hearing loss in a Chinese family.

BACKGROUND: X-linked deafness-4 (DFNX4) caused by the functional loss of the SMPX gene is one form of nonsyndromic hearing loss with postlingual onset. This study aimed to investigate the cause of X-linked inherited sensorineural nonsyndromic hearing loss in a four-generation Chinese family and to explain the reason for extremely different hearing phenotypes between the proband and other family members. METHODS: Whole-exome sequencing (WES) and co-segregation analysis were used to identify the pathogenic variants. Furthermore, methylation differences among the androgen receptor genes were uti

Central plasticity and dysfunction elicited by aural deprivation in the critical period.

The acoustic signal is crucial for animals to obtain information from the surrounding environment. Like other sensory modalities, the central auditory system undergoes adaptive changes (i.e., plasticity) during the developmental stage as well as other stages of life. Owing to its plasticity, auditory centers may be susceptible to various factors, such as medical intervention, variation in ambient acoustic signals and lesion of the peripheral hearing organ. There are critical periods during which auditory centers are vulnerable to abnormal experiences. Particularly in the early postnatal develo

Novel heterozygous mutations in the otogelin-like (OTOGL) gene in a child with bilateral mild nonsyndromic sensorineural hearing loss.

Hearing loss is a common disease, of which genetic factors are the main cause. The incidence of mild or moderate postlingual deafness in children is not high, and the impact on life and learning is not as severe as that of prelingual deafness. This leads to insufficient attention to the disorder in the clinic. To date, only a few disease-causing genes have been reported. This report describe a case of novel heterozygous mutations in OTOGL that causes nonsyndromic mild sensorineural hearing loss. Basic information, imaging examinations, audiological examination, and vestibular function tests of

Alteration of brain resting-state networks and functional connectivity in prelingual deafness.

BACKGROUND AND PURPOSE: Early hearing loss causes several changes in the brain structure and function at multiple levels; these changes can be observed through neuroimaging. These changes are directly associated with sensory loss (hearing) and the acquisition of alternative communication strategies. Such plasticity changes in the brain might establish a different connectivity pattern with resting-state networks (RSNs) and other brain regions. We performed resting-state functional magnetic resonance imaging (rsfMRI) to evaluate these intrinsic modifications. METHODS: We used two methods to cha

Missense variant in LOXHD1 is associated with canine nonsyndromic hearing loss.

Hearing loss is a common sensory deficit in both humans and dogs. In canines, the genetic basis is largely unknown, as genetic variants have only been identified for a syndromic form of hearing impairment. We observed a congenital or early-onset sensorineural hearing loss in a Rottweiler litter. Assuming an autosomal recessive inheritance, we used a combined approach of homozygosity mapping and genome sequencing to dissect the genetic background of the disorder. We identified a fully segregating missense variant in LOXHD1, a gene that is known to be essential for cochlear hair cell function an

A clinical and audiological investigation of osteogenesis imperfecta.

Fifty-six patients with osteogenesis imperfecta underwent clinical and audiological assessment. They completed questionnaires regarding their physical and otological disabilities, and attended various centres for audiological testing. It was found that 31 patients had a hearing loss. Hearing loss began in the second and third decades as a conductive loss, and progressed to a mixed loss. Eight patients, found in all age groups, had a pure sensorineural loss in one or both ears. Patients with osteogenesis imperfecta congenita suffered more fractures, became more physically disabled and had more

VEGFA165 gene therapy ameliorates blood-labyrinth barrier breakdown and hearing loss.

Millions of people are affected by hearing loss. Hearing loss is frequently caused by noise or aging and often associated with loss of pericytes. Pericytes populate the small vessels in the adult cochlea. However, their role in different types of hearing loss is largely unknown. Using an inducible and conditional pericyte depletion mouse model and noise-exposed mouse model, we show that loss of pericytes leads to marked changes in vascular structure, in turn leading to vascular degeneration and hearing loss. In vitro, using advanced tissue explants from pericyte fluorescence reporter models co

Cataract and hearing loss in a population-based study: the Beaver Dam studies.

PURPOSE: Cataract and hearing loss are each common at older ages and together may influence the ability to function independently and affect the sense of well-being. For these reasons, we sought to estimate the risk factors for comorbidity of age-related cataract and hearing loss. METHODS: A cross-sectional evaluation of a population-based cohort of older adults was conducted in Beaver Dam, Wisconsin, for presence of age-related cataract and hearing loss. The study evaluation included taking standardized medical histories, measuring blood pressures, and obtaining blood specimens. Standardized

Feasibility Assessment of an Intelligent Agent to Assist Preserving Healthy Hearing.

Hearing loss is a rising health concern. The World Health Organization estimates that over 1.5 billion people worldwide are currently experiencing hearing loss. This estimate is projected to rise to 2.5 billion by 2050. Hearing loss is often linked with other health conditions, such as dementia, exacerbating its impact. Chronic hearing loss often worsens over time. Early intervention and effective management are therefore important. One of the first intelligent agents designed to provide personalised advice to help preserve the hearing of individuals experiencing mild hearing loss is presented

Age-related changes in auditory perception. Hearing loss in the elderly: aging ear or aging brain?

Hearing loss in the elderly is a very common disease: it is estimated to affect up to a third of the population aged 65 years or more, and 50% of people over 75 years old. There is a growing amount of data concerning the association between hearing loss and cognitive decline. Various possible mechanisms at the basis of this association have been proposed, such as the "common cause hypothesis," the "cascade hypothesis," and the "cognitive load hypothesis."Critically reviewing the data is essential to highlight the features of the relationship between hearing loss and cognitive decline. Age-related changes in excitatory and inhibitory intra-cortical circuits in auditory cortex of C57Bl/6 mice.

A common impairment in aging is age-related hearing loss (presbycusis), which manifests as impaired spectrotemporal processing. Aging is accompanied by alteration in normal inhibitory (GABA) neurotransmission, and changes in excitatory (NMDA and AMPA) synapses in the auditory cortex (ACtx). However, the circuits affected by these synaptic changes remain unknown. Mice of the C57Bl/6J strain show premature age-related hearing loss and changes in functional responses in ACtx. We thus investigated how auditory cortical microcircuits change with age by comparing young (∼ 6 weeks) and aged (>1 year

[Causes of inner ear deafness; a critique of therapy (author's transl)].

In spite of recent criticisms, the diagnosis of inner ear hearing impairment is based on both pure tone thresholds and suprathreshold tests. The acoustico-facial reflex is useful only when used in conjunction with the suprathreshold tests.--Noise-induced deafness is generally confined to the higher frequencies of sound, and only exceptionally involve the middle frequencies as well. Low-tone hearing impairment may represent a partial symptom of Meniere's Disease as well as functional hearing loss. Zoster oticus produces a sensory hearing impairment but not a neural one, as might be expected fro
