Long-term potentiation of synaptic transmission in the hippocampus induced by a bee venom peptide.

Several neurotoxins have been isolated from bee venom. One of these, the mast cell degranulating peptide (MCD), releases histamine from mast cells and on central administration produces arousal at low concentrations and convulsions at higher doses. These effects are mediated through specific high-affinity binding sites which are concentrated in cortical structures, notably the hippocampus. This structure appears to be the source of changes in the electrocorticogram that follow injections of MCD into the cerebral ventricle, and which induce a quasi-permanent hippocampal theta rhythm in the moti…

Histamine sensitive sites in hippocampus: their probable role on prolactin release in male rats.

The effect of histamine (HA), 3-methyl-histamine (3MHA), HA antagonists or drugs interfering with HA synthesis, microinjected into the hippocampus (HPC), on prolactin (PRL) secretion were studied in rats. Three experiments were performed. In Experiment 1, increasing doses of HA or 3-MHA (9-90 nmol) were microinjected stereotaxically into the ventral HPC of adult male rats. In Experiment 2, 135 nmol of pyrilamine (PYR, an H1-HA-antagonist) or ranitidine (RAN, an H2-HA-antagonist) were administered locally into the ventral HPC. Fifteen min later, the rats were microinjected again with 45 nmol of…

Increased release of [3H]acetylcholine in vitro from the mouse hippocampus by a convulsant barbiturate.

The effects of the convulsant barbiturate, 5-(2-cyclohexylidene-ethyl)-5-ethyl barbituric acid (CHEB), on the spontaneous release of [3H]acetylcholine (ACh) from mouse hippocampal slices in an in vitro superfusion system have been evaluated. The pattern of the release of [3H]ACh by a single treatment with CHEB or an elevated potassium concentration was similar, with the peak release occurring in the same fraction. A maximally effective concentration of CHEB (500 microM) caused a 177% stimulation of spontaneous release of [3H]ACh, while 50 mM KCl increased the release by 2100% above the baselin

Patterns of astroglial hypertrophy and neuronal degeneration in the hippocampus of ages, memory-deficient rats.

The brains of aged and young Fischer rats were examined using a modified version of Cajal's gold chloride stain for astrocytes. A sizable loss of pyramidal cells and remarkably hypertrophic astrocytes were found in the hippocampus of the aged animals, while the astrocytic changes were not seen in other forebrain regions. Four major characteristics of the pattern of hippocampal astrogliosis were noted: (1) the astrocytes were often found to be grouped in clusters; (2) grouped astrocytes were commonly seen to have similarly oriented processes; (3) small blood vessels were more often stained in r…

Differential Coding of Associative Memory Information in the Macaque Lateral Prefrontal Cortex and Hippocampus.

The prefrontal cortex (PFC) and hippocampus (HPC) reportedly play crucial roles in the flexible use of stored information according to context. However, it remains unclear whether and how their neural representations differ during the context-guided retrieval. To solve this problem, we examined neural activity in the lateral PFC (lPFC, 470 neurons), medial PFC (mPFC, 322 neurons), and HPC (456 neurons) of three male macaques performing an item-location association memory task. The task required the animals to remember the location of a firstly presented item-cue relative to a background image …

A ventral hippocampus to nucleus accumbens pathway regulates impulsivity in male but not female rats.

Heightened impulsivity is associated with substance abuse disorders, gambling, and obesity. The ventral hippocampus (vHPC) has recently been linked with impulse control, yet the neurobiological and behavioral mechanisms through which this occurs are unknown. A subset of vHPC neurons projects to the nucleus accumbens shell (ACBsh), a brain region known for regulating reward and motivation. Here, we evaluated the role of vHPC-ACBsh signaling in food-directed impulsivity using fiber photometry and transsynaptic chemogenetic and behavioral approaches. Male and female rats were trained in the diffe…

Integrated Proteomics and Metabolomics Analysis in Hippocampus of Rats With T2DM-Associated Cognitive Decline.

Type 2 diabetes mellitus (T2DM) is currently one of the most prominent and global chronic conditions. Cognitive decline is one of the major complications of T2DM, but its precise molecular mechanism remains unclear. Metabolomics and proteomics were combined in this study to investigate alterations in metabolites and proteins in the hippocampus of T2DM rats. KEGG Markup Language (KGML) network analysis was conducted to integrate underlying relationships among differentially expressed metabolites and proteins. 58 significantly differentially expressed metabolites and 61 differentially expressed …

AMPA Receptors in NG2 Glia Differently Affect Signal Transduction in the Hippocampus and Cerebellum.

Gray matter NG2 glia constitute a heterogeneous population of cells whose functions remain incompletely understood. In the hippocampus, Schaffer collaterals activate AMPA receptors (AMPARs) in NG2 glia, giving rise to small excitatory post-synaptic currents (EPSCs). Climbing fibers of the cerebellum also form synapses with NG2 glia, although producing much larger EPSCs. We aimed to identify mechanisms generating these regional differences in the efficacy of neuron-glia synapses. Combined patch-clamp and RT-PCR analyses allowed for determining structural and functional differences of AMPARs exp…

Distinct and coordinated contributions of hippocampus and frontal eye field to novelty exploration and revisitation.

Humans dynamically alternate between exploring novel information and revisiting familiar content, yet the neural mechanisms that separately govern these behaviors remain incompletely understood. In this study, we combined intracranial electrophysiology with eye-tracking to investigate the respective roles of the hippocampus (HP) and frontal eye field (FEF) at the level of individual fixations during a visual working memory task. During novelty exploration, hippocampal high-theta activity increased before fixation onset, suggesting a role in anticipatory attention, while FEF theta oscillations …

Neuronal Activity Rapidly Decreases the Level of p53 Protein in Rat Hippocampus In Vivo.

It was previously shown that the induction of long-term post-tetanic potentiation of synaptic transmission in rat hippocampal slices is accompanied by a rapid decrease in the levels of the transcription factor p53 (tumor suppressor p53). However, it remained unclear whether a similar phenomenon could be observed in vivo. To answer this question, we employed a pentylenetetrazole (PTZ)-induced seizure model in rats. At 20 and 40 min after an intraperitoneal injection of PTZ, hippocampal p53 levels were significantly reduced compared to the intact controls. These findings validate the data previo

Temporal and Spatial Gene Expression Dynamics in Neonatal HI Hippocampus with Focus on Arginase.

BACKGROUND: Hypoxic-ischemic (HI) brain injury triggers a dynamic, multi-phase response involving early microglial efferocytosis followed by extracellular matrix (ECM) deposition and scar formation. Arginase-1 (ARG1), a key enzyme in tissue repair, is implicated in both processes, yet its role in neonatal microglia remains poorly defined. We characterize ARG1-linked pathways in neonatal microglia, identifying distinct efferocytic and fibrotic phases post-HI. METHODS: HI was induced in P9 mice using the Vannucci model, and brains were collected at 24 h (D1) and 5 days (D5). Spatially resolved

Lead neurotoxicity in experimental models: A systematic review on effects on the cerebrum, cerebellum, and hippocampus.

The extensive use of heavy metals, such as lead, has resulted in environmental damage and numerous health problems in many parts of the world. Reports indicate that lead is the most significant toxic heavy metal with adverse impacts on several body systems. Primarily, lead targets the nervous system and causes a variety of neurological, motor, and behavioral deficits in humans and laboratory animals. Although several reports demonstrate the toxicity of lead, there is a paucity of comprehensive and updated reviews to highlight various histopathological findings on the effects of lead and its me

Mesoscale connectivity of the human hippocampus and fimbria revealed by ex vivo diffusion MRI.

The human hippocampus is essential to cognition and emotional processing. Its function is defined by its connectivity. Although some pathways have been well-established, our knowledge about anterior-posterior connectivity and the distribution of fibers from major fiber bundles remains limited. Mesoscale (250 μm isotropic acquisition, upsampled to 125 μm) resolution MR images of the human temporal lobe afforded a detailed visualization of fiber tracts, including those that related anterior-posterior substructures defined as subregions (head, body, tail) and subfields (cornu ammonis 1-3, dentate…

A mouse model of early sporadic tau pathology induces neurogenic plasticity in the hippocampus.

Alzheimer's disease varies by sex but is broadly characterized by widespread neurodegeneration and the accumulation of insoluble amyloid plaques and neurofibrillary tangles. However, at the earliest stages of the disease cell death and pathological tau are localized to the entorhinal cortex. In particular, the lateral entorhinal cortex, and its functions in object-related memory, are among the most vulnerable in aging. Notably, the entorhinal cortex projects directly to the dentate gyrus subregion of the hippocampus, where neurogenesis proceeds throughout adult life. Immature, adult-born neuro…

Dissociable contributions of the hippocampus and orbitofrontal cortex to representing task space in a social context.

The hippocampus (HC) and the orbitofrontal cortex (OFC) jointly encode a map-like representation of a task space to guide behavior. It remains unclear how the OFC and HC interact in encoding this map-like representation, though previous studies indicated that both regions have different functions. We acquired the functional magnetic resonance imaging data under a social navigation task in which participants interacted with characters in a two-dimensional "social space." We calculate the social relationships between the participants and characters and used a drift-diffusion model to capture the…

Stress-induced NLRP3 inflammasome activation and myelin alterations in the hippocampus of PTSD rats.

Inflammatory and myelin changes may contribute to the pathophysiology of post-traumatic stress disorder (PTSD). The NOD-like receptor (NLR) family, pyrin domain-containing protein 3 (NLRP3), a brain inflammasome, is activated in the hippocampus of mice with PTSD. In other psychiatric disorders, NLRP3 expression has been associated with axonal myelination and demyelination. However, the association between NLRP3 and myelin in rats with PTSD remains unclear. Therefore, this study aims to investigate the relationship between the NLRP3 inflammasome and myelin in the hippocampus of rats with PTSD. …

Nonlinear analysis of neuronal firing modulated by sinusoidal stimulation at axons in rat hippocampus.

INTRODUCTION: Electrical stimulation of the brain has shown promising prospects in treating various brain diseases. Although biphasic pulse stimulation remains the predominant clinical approach, there has been increasing interest in exploring alternative stimulation waveforms, such as sinusoidal stimulation, to improve the effectiveness of brain stimulation and to expand its application to a wider range of brain disorders. Despite this growing attention, the effects of sinusoidal stimulation on neurons, especially on their nonlinear firing characteristics, remains unclear. METHODS: To address
