HPRT1 Deficiency Induces Alteration of Mitochondrial Energy Metabolism in the Brain.

Alterations in function of hypoxanthine guanine phosphoribosyl transferase (HPRT), one of the major enzymes involved in purine nucleotide exchange, lead to overproduction of uric acid and produce various symptoms of Lesch-Nyhan syndrome (LNS). One of the hallmarks of LNS is maximal expression of HPRT in the central nervous system with the highest activity of this enzyme in the midbrain and basal ganglia. However, the nature of neurological symptoms has yet to be clarified in details. Here, we studied whether HPRT1 deficiency changes mitochondrial energy metabolism and redox balance in murine n

MicroRNAs: tools of mechanistic insights and biological therapeutics discovery for the rare neurogenetic syndrome Lesch-Nyhan disease (LND).

MicroRNAs (miRNAs) are small regulatory RNAs that modulate the translation of mRNA. They have emerged over the past few years as indispensable entities in the transcriptional regulation of genes. Their discovery has added additional layers of complexity to regulatory networks that control cellular homeostasis. Also, their dysregulated pattern of expression is now well demonstrated in myriad diseases and pathogenic processes. In the current review, we highlight the role of miRNAs in Lesch-Nyhan disease (LND), a rare neurogenetic syndrome caused by mutations in the purine metabolic gene encoding

HPRT deficiency: identification of twenty-four novel variants including an unusual deep intronic mutation.

Hypoxanthine phosphoribosyltranferase (HPRT) deficiency is an X-linked disorder of purine salvage that ranges phenotypically from hyperuricaemia to Lesch-Nyhan Syndrome. Molecular testing is necessary to identify female carriers within families as a prelude to prenatal diagnosis. During the period 1999-2010 the Purine Research Laboratory studied 106 patients from 68 different families. Genomic sequencing revealed mutations in 88% of these families, 24 of which were novel. In eight patients, exon sequencing was not informative. Copy-DNA analysis in one patient revealed an insertion derived from

Methylation status of HPRT1 promoter in HPRT deficiency with normal coding region.

UNLABELLED: Lesch-Nyhan syndrome is an X-linked recessive inborn error of metabolism due to a complete deficiency of hypoxanthine-guanine phosphoribosyltransferase (HPRT) activity (OMIM 300322). Partial deficiency of HPRT (OMIM 300323) is characterized by the effects of excess uric acid synthesis and a continuum spectrum of neurological manifestations, without the manifestations of full-blown Lesch-Nyhan syndrome. Both diseases have been associated with mutations in the HPRT1 gene. We have described one Lesch-Nyhan patient and four partial HPRT deficient patients with a normal HPRT1 coding reg

Molecular evidence for new mutation at the hprt locus in Lesch-Nyhan patients.

Hypoxanthine-guanine phosphoribosyltransferase (HPRT; EC2.4.2.8), which functions in the metabolic salvage of purines, is encoded by an X-linked gene in man. Partial HPRT deficiencies are associated with gouty arthritis, while absence of activity results in Lesch-Nyhan syndrome (L-N). L-N patients fail to reproduce and the heterozygous state appears to confer no selective advantage. Thus, Haldane's principle predicts that new mutations at the hprt locus must occur frequently in order for L-N syndrome to be maintained in the population. This constant introduction of new mutations would be expec

Analysis of in vivo mutation in the hprt and tk genes of mouse lymphocytes.

Determining mutant frequencies in endogenous reporter genes is a tool for identifying potentially genotoxic environmental agents and discovering phenotypes prone to genomic instability and diseases, such as cancer. Here we describe a high-throughput method for identifying mouse spleen lymphocytes having mutations in the endogenous X-linked hypoxanthine-guanine phosphoribosyltransferase (Hprt) gene and the endogenous autosomal thymidine kinase (Tk) gene. The selective expansion of mutant lymphocytes is based on the phenotypic properties of Hprt- and Tk-deficient cells. The same procedure can be

Lesch-Nyhan syndrome in Japan.

An epidemiological survey of Lesch-Nyhan (L-N) syndrome in Japan was carried out. In the first survey, questionnaires were mailed to the pediatric departments of the university hospitals, homes for mentally retarded children, and city and county medical associations. This study disclosed 41 patients with L-N syndrome and 48 with suspected L-N syndrome. In the second survey, questionnaires were mailed to the institutes that reported cases or suspected cases of L-N syndrome. In 29 of these cases, hypoxanthine-guanine phosphoribosyltransferase (HPRT) activity in the patients' erythrocytes was mea

Cellular hypersensitivity to UV-A: a clue to the aetiology of actinic reticuloid?

Fibroblasts cultured from six patients with actinic reticuloid (AR) showed striking cytopathic changes and inhibition of RNA synthesis after exposure to near-ultraviolet radiation that had no effect on normal and other photosensitive cell strains, such as those of xeroderma pigmentosum and Bloom syndrome. An abnormal pattern of DNA fragmentation was observed after doses insufficient to cause cytopathic effects. These results suggest a cellular defect in the prevention or repair of some damage caused by free radicals and other photoproducts. In order to explain the pathogenesis of AR, it is pro

[Routine urinary oxypurine assays for the detection of xanthine-oxidase deficiency (author's transl)].

Xanthine-oxidase deficiency should be suspected whenever hypouricaemia coexists with hypouricuria and hyperoxypurinuria. In 18 months. Two new cases of xanthinuria were detected by routine urinary oxypurine assay among 10,000 patients where uricaemia was systematically tested. The xanthine-oxidase deficiency was associated with Hodgkin's disease in one case and with uncomplicated pregnancy in the other.
