A role for carbonic anhydrase in early eye morphogenesis.

The carbonic anhydrase inhibitor methazolamide was used to test the hypothesis that carbonic anhydrase activity plays a role in the early morphogenesis of the chick eye. Methazolamide was administered topically to eyes of 4-7-day-old chick embryos in shell-free culture. Either continuous application of drug solution with a miniosmotic pump or a single daily application of drug resulted in significant reduction in embryonic eye growth relative to sham-treated controls.

Hydrostatic distention and pharmacological treatment of epinephrine-induced microarterial spasm.

The efficacy of using hydrodistention and vasodilatory drugs to relieve spasm in arteries was investigated. The femoral arteries of 64 rabbits (divided into five groups) were placed in spasm by the topical application of epinephrine (1 mg/ml). In group A (controls) vasospasm was induced without further treatment. In group B vasospasm was induced, and the arteries were hydrodistended with normal saline. In three additional groups vasospasm was induced and either 10% lidocaine hydrochloride (group C), verapamil hydrochloride (group D), or chlorpromazine hydrochloride (group E) was applied. Thirt

Rapid correction of severe lactic acidosis with massive isotonic bicarbonate infusion and simultaneous ultrafiltration.

Patients with severe lactic acidosis and oliguria who receive large amounts of sodium bicarbonate may develop fluid overload and hyperosmolarity. We infused massive amounts of isotonic sodium bicarbonate and simultaneously removed the excess sodium and water with ultrafiltration if 2 patients with lactic acidosis. The first patient received 1,125 mmol of bicarbonate over 4.5 h with a rise in pH from 7.00 to 7.36 and in HCO3 from 3.5 to 15.7 mmol/l. The second received 968 mmol of bicarbonate over 5.25 h with a rise in pH from 7.00 to 7.27 and in HCO3 from 5.3 to 14 mmol/l. Blood pressure remai

Methylxanthines and intestinal drug absorption.

1. Jejunal loops of anaesthetized rats were filled with buffered isotonic solutions of tritiated water (HTO), urea, antipyrine and salicylic acid at pH 6-8. The venous outflow and the appearence rate of the substances in the intestinal venous blood were determined. Blood pressure was kept constant by adjustable supply of blood from donor rats throughout the experiment. 2. The absorption of urea, antipyrine and salicylic acid was, in concentrations from 0.001 to 1.0 mg/ml found to be directly proportional to the intraluminal concentration. 3. Theophylline and caffeine (2 mg/ml), when injected i…

Does a balanced colloid decrease perioperative blood loss in paediatric cardiac surgery: A double-blinded randomized controlled trial?

BACKGROUND: Unbalanced fluid solutions cause metabolic acidosis and could be associated with impaired coagulation and increased blood loss. OBJECTIVE: To investigate whether the use of a balanced colloid compared with a saline colloid for peri-operative fluid therapy in children undergoing cardiac surgery is associated with decreased blood loss and exposure to blood products. DESIGN: Double-blinded randomised controlled trial. SETTING: Tertiary children's hospital from 2013 to 2016. PATIENTS: Children older than 29 days and younger than 3 years admitted for cardiac surgery with cardiopulmo

The Impact of Three Different Wash Solutions on Autotransfusion Products.

Many blood conservation techniques and strategies have been implemented to aid in decreasing the use of allogenic blood utilization during pediatric cardiothoracic surgery. Use of techniques, such as acute normovolemic hemodilution, retrograde autologous prime, venous autologous prime, and autotransfuion, may lead to a decrease in the need for allogenic blood products. Autotransfusion has become a standard of care for all cardiothoracic surgical procedures requiring cardiopulmonary bypass (CPB). Although widely used, there is still debate over which wash solution will produce the most physiolo

A critical appraisal of intravenous fluids: from the physiological basis to clinical evidence.

Fluid management has been a vital part of routine clinical care for more than 180 years. The increasing number of available fluids has generated controversy about the optimal choice of resuscitation fluid. In this review, we provide a critical overview of the different fluids available, their composition, the relevant physiology as well as the published evidence on clinical outcomes to guide their use. Commonly used infusion fluids include semisynthetic colloids and crystalloids; the latter comprises both normal saline (NaCl 0.9%) and the more chloride-restricted 'balanced' crystalloids. Despi

Hypothermic preconditioning of donor organs prior to harvesting and ischaemia using ice-cold intravenous fluids.

To promote organ transplantation and viability, hypothermia has been applied as a protective agent for decades. Current management of organ preservation includes hypothermia as a component of static storage. In rare cases, hypothermic perfusion is initiated in the donor organs prior to harvesting but this requires invasive perfusion techniques. Therefore, hypothermic organ protection is currently achieved only after organ retrieval and onset of ischaemic injury cascades. The relevant mechanisms of cellular and organ damage involve ischaemia-reperfusion injury and apoptosis. In this hypothesis,

Influence of the bathing medium on the contractile responses of the rabbit urinary bladder.

This study examined contractile responses of the in vitro rabbit whole-bladder preparation to field stimulation, bethanechol and KCl in Tyrode's solution and minimum essential medium (MEM). We found frequency-dependent increases in intravesical pressure in bladders incubated in Tyrode's solution and MEM. However, bladders incubated in MEM consistently responded with greater increases in intravesical pressure compared to those incubated in Tyrode's solution. Similarly, there were frequency-dependent increases in the rate of pressure generation in bladders incubated in Tyrode's solution and MEM,

Orogastric determinants of drinking in rats: interaction between absorptive and peripheral controls.

Water intake of water-deprived rats, drinking with absorption prevented by pyloric occlusion, was compared with that of rats drinking normally. Drinking without absorption, under the control of only orogastric factors, was not graded in response to varied deprivation (Experiment 1). Orogastric controls effectively inhibited intake after absorbed water, but not isotonic saline, preloads (Experiment 2). The extent of orogastric inhibition was directly related to absorbed preload volume (Experiment 3). With large absorbed water preloads (9 ml), intakes with and without absorption were equal. Norm

Comparison of microdialysis sampling perfusion fluid components on the foreign body reaction in rat subcutaneous tissue.

Microdialysis sampling is a commonly used technique for collecting solutes from the extracellular space of tissues in laboratory animals and humans. Large molecular weight solutes can be collected using high molecular weight cutoff (MWCO) membranes (100kDa or greater). High MWCO membranes require addition of high molecular weight dextrans or albumin to the perfusion fluid to prevent fluid loss via ultrafiltration. While these perfusion fluid additives are commonly used during microdialysis sampling, the tissue response to the loss of these compounds across the membrane is poorly understood. Ti

Ringer's malate solution protects against the multiple organ injury and dysfunction caused by hemorrhagic shock in rats.

Malic acid, in the form of its anion malate, is a key intermediate in the major biochemical energy-producing cycle known as the citric acid or Krebs cycle. In this study, the authors investigated the protective effect of a novel crystalloid solution of Ringer's malate following fluid resuscitation of hemorrhagic shock using a rat model. Under general anesthesia, Sprague-Dawley male rats were subjected to 60 min of hemorrhagic shock (40 mmHg for 60 min) followed by crystalloid resuscitation. Groups were as follows: (1) sham shock, (2) normal saline, (3) Ringer's lactate, and (4) Ringer's malate

Myocardial protection in heart transplantation using blood cardioplegia: 12-year outcome of a prospective randomized trial.

BACKGROUND: Blood cardioplegia yields a lower prevalence of right heart failure, arrhythmias, and myocardial ischemia early after heart transplantation (HTx). Because depolarizing (high [K(+)]) cardioplegic solutions may alledgedly cause endothelial damage, the 12-year outcome of a prospective randomized trial was reviewed. METHODS: Between January 1997 and March 1998, 47 consecutive patients received crystalloid (Group 1, n = 27) or blood cardioplegia (Group 2, n = 20). The groups were similarly matched: recipient age (54 ± 11 vs 55 ± 7 years, p = 0.9), sex (89% vs 90% males, p = 0.9), diagn

Swelling of the collagen-keratocyte matrix of the bovine corneal stroma ex vivo in various solutions and its relationship to tissue thickness.

AIM: The mammalian corneal stroma, like some other connective tissues, can absorb fluid, swell and become oedematous. Since studies on the corneal stroma have been carried out with different types of preparations and solutions, inter-study comparisons are very difficult. A study was thus undertaken on a standardised preparation to assess the relative magnitude of this swelling and its relationship to thickness of the preparations. METHODS: From selected recent post-mortem eyes of adult cattle, stroma preparations were cut from the central part of the cornea. These preparations were immersed i
