Ivermectin and moxidectin against soil-transmitted helminth infections.

Ivermectin and moxidectin, two macrocyclic lactones, are potent antiparasitic drugs currently registered and mainly used against filarial diseases; however, their potential value for improved soil-transmitted helminth (STH) control has been acknowledged. This review provides insights on recent studies evaluating the efficacy of ivermectin and moxidectin as single or coadministered therapy against human soil-transmitted helminthiases (including Strongyloides stercoralis infections) and on pharmacokinetic/pharmacodynamic parameters measured in treated populations. Furthermore, we discuss current…

Recent pharmacological advances in the repurposing of artemisinin drugs.

Artemisinins are a family of sesquiterpene lactones originally derived from the sweet wormwood (Artemisia annua). Beyond their well-characterized role as frontline antimalarial drugs, artemisinins have also received increased attention for other potential pharmaceutical effects, which include antiviral, antiparsitic, antifungal, anti-inflammatory, and anticancer activities. With concerted efforts in further preclinical and clinical studies, artemisinin-based drugs have the potential to be viable treatments for a great variety of human diseases. Here, we provide a comprehensive update on recent…

Base-Free Iron Catalyzed Transfer Hydrogenation of Esters Using EtOH as Hydrogen Source.

Herein, we report on the use of the iron pincer complex Iron-MACHO-BH, in the base-free transfer hydrogenation of esters with EtOH as a hydrogen source. More than 20 substrates including aromatic and aliphatic esters and lactones were reduced affording the desired primary alcohols and diols with moderate to excellent isolated yields. It is also possible to reduce polyesters to the diols with this method, enabling a novel way of plastic recycling. Reduction of the renewable substrate methyl levulinate proceeds to form 1,4-pentanediol directly. The yields are largely governed by the equilibrium …

Presentation of three cases with phyto contact dermatitis caused by Ranunculus and Anthemis genera.

In addition to being used especially in a systematic way, herbal treatments are preferred topically by the elderly population in some rheumatological and dermatological diseases. Although alternative medicine treatments may have beneficial effects, certain plants are known to cause common contact dermatitis. Protoanemonin and sesquiterpene lactones, which are found in such plant types as respectively Ranunculus and Anthemis, often cause contact dermatitis. In this article, three cases of phyto contact dermatitis, which are the result of topical use of plant types of Ranunculus ve Anthemis as a…

Hypurpurins A-P, furanolabdane diterpenoids from the aerial parts of Hypoestes purpurea and their bioactivities.

Sixteen previously undescribed furanolabdane diterpenoids, designated hypurpurins A-P (1-16), along with two known analogs, hypopurins B and C (17 and 18), were isolated from the aerial parts of Hypoestes purpurea. The chemical structures of 1-16 were elucidated through comprehensive spectroscopic analyses, including HR-ESI-MS, NMR, UV, and IR spectroscopy. The absolute configurations of these new compounds were further determined by electronic circular dichroism and single-crystal X-ray diffraction analysis. Structurally, compounds 1 and 2 are unprecedented furanolabdane diterpenoids bearing

Mechanism and optimization of supramolecular complexation-enhanced fluorescence spectroscopy for the determination of SN-38 in plasma and cells.

Quantitative detection of two different forms of SN-38 in biological samples is, currently, cumbersome and difficult. A revisit to the mechanism of supramolecular complexation-enhanced fluorescence spectroscopy helps to optimize the determination of SN-38 in plasma and the cellular pharmacokinetics in A549 cells based on the supramolecular complexation. Firstly, the inclusion mechanism dominated by thermodynamic constants was determined by measuring kinetic/thermodynamic parameters (kon , koff , ΔG, ΔH, ΔS). On this basis, the best effect of fluorescence sensitization was optimized through scr

Chemical tools for selective activity profiling of bacterial penicillin-binding proteins.

Penicillin-binding proteins (PBPs) are membrane-associated proteins involved in the biosynthesis of peptidoglycan (PG), the main component of bacterial cell walls. These proteins were discovered and named for their affinity to bind the β-lactam antibiotic penicillin. The importance of the PBPs has long been appreciated; however, specific roles of individual family members in each bacterial strain, as well as their protein-protein interactions, are yet to be understood. The apparent functional redundancy of the 4-18 PBPs that most eubacteria possess makes determination of their individual roles

Specificity and inhibitory mechanism of andrographolide and its analogues as antiasthma agents on NF-κB p50.

Andrographolide (1) is a diterpenoid lactone with an α,β-unsaturated lactone group that inhibits NF-κB DNA binding. Andrographolide reacts with the nucleophilic Cys62 of NF-κB p50 through a Michael addition at the Δ(12(13)) exocylic double bond to form a covalent adduct. Using computer docking, site-directed mutagenesis, and mass spectrometry, the noncovalent interactions between andrographolide and additional binding site residues other than Cys62 were found to be essential for the covalent incorporation of andrographolide. Furthermore, the addition reaction of andrographolide on Cys62 was hi

Mercapturic acid conjugates of 4-hydroxy-2-nonenal and 4-oxo-2-nonenal metabolites are in vivo markers of oxidative stress.

Oxidative stress-induced lipid peroxidation leads to the formation of cytotoxic and genotoxic 2-alkenals, such as 4-hydroxy-2-nonenal (HNE) and 4-oxo-2-nonenal (ONE). Lipid-derived reactive aldehydes are subject to phase-2 metabolism and are predominantly found as mercapturic acid (MA) conjugates in urine. This study shows evidence for the in vivo formation of ONE and its phase-1 metabolites, 4-oxo-2-nonen-1-ol (ONO) and 4-oxo-2-nonenoic acid (ONA). We have detected the MA conjugates of HNE, 1,4-dihydroxy-2-nonene (DHN), 4-hydroxy-2-nonenoic acid (HNA), the lactone of HNA, ONE, ONO, and ONA in

Design, synthesis, and antifungal activity of 14-aryloxy/amide substituted andrographolide derivatives.

In order to find more compounds with antifungal activities, a series of new compounds were designed and synthesized using the natural product of diterpenoid lactones, andrographolide, as the parent nucleus. Target compounds were identified by 1H NMR, 13C NMR and/or HR-MS. The bioassay results demonstrated that compound 4d exhibited inhibition rates of over 60% against six fungal strains at 100 µg/mL, and the EC50 value of 4d against P. piricola was only 9.09 µg/mL. SEM test showed that 4d could damage the mycelia of P. piricola. obviously. Moreover, 4d had no effect on cowpea seed germination,…

Titanium-Catalyzed Exhaustive Reduction of Oxo-Chemicals.

Catalytic reduction of carboxylic acids and derivatives all the way to their corresponding methyl-compounds is very rare and still challenging for homogeneous transition-metal catalysis. Herein, we report an unprecedented and general catalytic exhaustive reduction of carboxylic functional group straightforwardly to a methyl group. This reaction was achieved using earth-abundant and readily available titanium as a catalyst. Our system has broad functional group tolerance and works for various other types of oxo-chemicals such as alcohols, aldehydes, ketones, lactones, and carboxylates (>100 exa…

Inhibition of Vibrio cholerae biofilm by AiiA enzyme produced from Bacillus spp.

Vibrio cholerae is the causative agent of water-borne diarrheal disease, cholera. The formation of biofilm favors survival and persistence of V. cholerae in the aquatic environment and also inside the host. AHL lactonase (AiiA), a metallo-beta-lactamase produced by Bacillus spp., blocks quorum sensing in Gram-negative bacteria by hydrolyzing N-acyl-homoserine lactones (AHLs). In the present investigation, AiiA-mediated inhibition of V. cholerae biofilm was studied. Two novel alleles of aiiA-encoding genes from Bacillus spp. were expressed in E. coli, and the results demonstrated that AiiA enzy…

The PmlI-PmlR quorum-sensing system in Burkholderia pseudomallei plays a key role in virulence and modulates production of the MprA protease.

Burkholderia pseudomallei is the causative agent of melioidosis, an often fatal infection of humans and animals. The virulence of this pathogen is thought to depend on a number of secreted proteins, including the MprA metalloprotease. We observed that MprA is produced upon entry into the stationary phase, when the cell density is high, and this prompted us to study cell density-dependent regulation in B. pseudomallei. A search of the B. pseudomallei genome led to identification of a quorum-sensing system involving the LuxI-LuxR homologs PmlI-PmlR. PmlI directed the synthesis of an N-acylhomose
