Lipids, lipid-modifying drug targets with venous thromboembolism: a mendelian randomization study.

BACKGROUND: The role of lipid-lowering therapies (e.g., statins) during anticoagulation for Venous Thromboembolism (VTE) remains unclear. This study investigated the causal role of lipid traits in VTE and evaluated lipid-lowering drug targets using a Mendelian Randomization (MR) approach. METHODS: Genetic variants associated with lipid traits and lipid-lowering drug targets were extracted from the Global Lipids Genetics Consortium GWAS. Summary statistics for VTE, Deep Vein Thrombosis (DVT), and Pulmonary Embolism (PE) were obtained from UK Biobank. For associated targets, expression quantita…

Lipid Messengers: Mechanisms and Clinical Applications of Exosomal Lipids in Neurodegenerative Diseases.

Neurodegenerative diseases, including Alzheimer's disease and Parkinson's disease, represent an escalating global health burden owing to their complex pathophysiology and limited therapeutic options. Exosomes, nanoscale extracellular vesicles (30-150 nm) capable of crossing the blood-brain barrier (BBB), have emerged as critical mediators of intercellular communication in the central nervous system. While research has predominantly focused on exosomal proteins and nucleic acids, the functional significance of exosomal lipids in neurodegeneration is increasingly recognized. This review outlines…

Minor lipids with major roles: Viral exploitation of phosphoinositides during RNA replication.

Positive-sense (+)RNA viruses are major pathogens of humans, animals and plants. This review summarizes the complex subversion of the host phosphoinositide cycles for virus replication. Phosphoinositides (PIPs) are generated by tightly regulated activity of dedicated PIP kinases from PI in different subcellular locations. Phosphoinositides frequently act as molecular switches and are master regulators of membrane dynamics. Phosphoinositides are exploited by (+)RNA viruses to build membrane-associated viral RNA replication machinery called viral replication organelles (VROs) inside the infected

Bioactive lipids from grains and their effects on intestinal homeostasis: a review.

Grains are staple foods for a large proportion of the global population and are characterized by high carbohydrate content, with proteins and lipids present in lower amounts. Among these macronutrients, specific bioactive lipids have been reported to exert physiological effects, including reducing blood pressure, decreasing the risk of cardiovascular and cerebrovascular diseases, and promoting intestinal health. Research on bioactive lipids has therefore attracted considerable attention. The intestinal tract is a major site for digestion and absorption and also serves as an important defense b…

Appetite for destruction: dietary lipids tune the ferroptotic threshold of adaptive immunity.

Dietary lipids tune the ferroptosis checkpoint in T cells. PUFA-rich diets promote incorporation of oxidizable PUFAs into T-cell membranes, increasing lipid peroxidation and ferroptosis sensitivity. In contrast, MUFA-rich diets favor incorporation of monounsaturated fatty acids that reduce lipid peroxidation and confer ferroptosis resistance. This lipid-dependent balance establishes a ferroptosis checkpoint that regulates T-cell survival and shapes adaptive immune responses, including vaccine efficacy, anti-tumor immunity, CAR-T-cell therapy and immune homeostasis. Created in BioRender. Morgan…

Marine Lipids and Alzheimer's Disease: Biochemistry, Bioaccessibility/Bioavailability, Metabolism, and Health Effects.

Due to its high prevalence and significant impact on modern society, Alzheimer's disease (AD) is one of the most important neurodegenerative disorders. It is more common among individuals over the age of 65, and its incidence has increased sharply as a result of rising life expectancy. Several factors have made it challenging to identify an effective treatment for AD. One major difficulty lies in its complexity, as the mechanisms involved in its progression are not yet fully understood. Nevertheless, the role of diet and lipids has been highlighted by numerous studies, underscoring their poten…
