Cardiovascular and intestinal responses to oxidative and nitrosative stress during prolonged magnesium deficiency.

In rodents with dietary magnesium deficiency (Mg deficiency), hypomagnesemia, occurs leading to a rise in circulating substance P from neuronal tissues to trigger systemic inflammatory stress in cardiac and intestinal tissues. Sustained elevations of substance P may result from impaired neutral endopeptidase (NEP) activity due to reactive oxygen and reactive nitrogen species. Associated increase in intestinal permeability includes infiltration of WBC and endotoxemia, which can further amplify the systemic inflammatory response that leads to impaired contractile function associated with up-regu…

Effect of magnesium deficiency on intracellular ATP levels in human lens epithelial cells.

Cataractous lenses have an altered distribution of the intracellular ionic environment, and the lens ionic imbalance with increased levels of calcium (Ca2+) and sodium (Na+), coupled with decreased levels of magnesium (Mg2+) and potassium (K+), is related to cataract development in human senile cataracts. We previously found that the decrease of ATP in lenses caused lens ionic imbalance, and probably decrease in ATPase function. In this study, we investigated the effect of Mg2+ deficiency on cataract progression using human lens epithelial (HLE) cells. Expression levels of inducible nitric oxi

Magnesium deficiency in patients with recent myocardial infarction and provoked coronary artery spasm.

This study sought to clarify the relationship between magnesium (Mg) deficiency and coronary artery spasm provoked by pharmacologic agents in patients with a recent acute myocardial infarction (AMI). Twenty-three consecutive patients suffering from AMI were investigated with a Mg retention test (Mg: 0.1 mmol/kg for 4 h) in both the acute phase (within I week (3+/-2 days) of onset) and the subacute phase (3-4 weeks (24+/-6 days) of the onset). Early coronary arteriography was performed in all patients. Coronary stenosis in the infarct-related artery was less than 90% in all patients in the suba

Effect of acute magnesium deficiency (MgD) on aortic endothelial cell (EC) oxidant production.

Magnesium deficiency (MgD) has been associated with production of reactive oxygen species, cytokines, and eicosanoids, as well as vascular compromise in vivo. Although MgD-induced inflammatory change occurs during "chronic" MgD in vivo, acute MgD may also affect the vasculature and consequently, predispose endothelial cells (EC) to perturbations associated with chronic MgD. As oxyradical production is a significant component of chronic MgD, we examined the effect of acute MgD on EC oxidant production in vitro. In addition we determined EC; pH, mitochondrial function, lysosomal integrity and ge…

Serum and red blood cell magnesium, copper, and zinc content in G6PD deficiency.

Sixty-nine G6PD-deficient and 67 normal non-deficient Chinese adult males were investigated for blood hemoglobin concentration and for serum and red cell content of magnesium, copper, and zinc. There was no significant difference in blood hemoglobin content. Both the serum magnesium and copper content were found to be significantly lower in the G6PD-deficient subjects compared to that in the non-deficient control. Serum zinc content and red cell content of all the three metals were not significantly altered in G6PD deficiency.

Superoxide anion in polymorphonuclear leukocytes of hairless rats suffering from magnesium-deficiency dermatitis.

Clinical and histological studies were made on magnesium-deficiency dermatitis produced on a new strain of hairless rats. Superoxide anion generation in polymorphonuclear leukocytes (PMNs) and the inhibitory effect of some antioxidants and anti-inflammatory agents against the elicited dermatitis were also examined. From the detection of increased superoxide production and effective actions of inhibitors including superoxide dismutase, cepharanthine, 4-4'-diamono-diphenylsulphone and 5,4,11,14-eicosatetraynoic acid (ETYA), it is indicated that superoxide anion or related activated oxygen may be

Disturbances of vitamin D metabolism and action during clinical and experimental magnesium deficiency.

It has been known since the 1920s that magnesium is influential in calcium homeostasis. In the 1970s it was documented that parathyroid secretion and activity may be impaired in magnesium deficiency. In the past two decades a variety of studies have indicated alterations in circulating vitamin D metabolites in humans, although these observations are not entirely consistent. Animal studies have not consistently demonstrated impaired vitamin D metabolism during relatively brief periods of magnesium deprivation, despite in vitro magnesium dependence of 1 alpha-hydroxylase activity. Studies of the…

Hepatic gluconeogenic enzymes, plasma insulin and glucagon response to magnesium deficiency and fasting.

Three experiments were conducted to assess the effects of magnesium deficiency on the activities of hepatic glucose-6-phosphatase (G6Pase), fructose 1,6-bisphosphatase (FDPase) and phosphoenolpyruvate carboxykinase (PEPCK). Experiment 1 was designed to determine if magnesium deficiency interfered with the gluconeogenic response to fasting. Rats were fed either a control (C) or magnesium-deficient (MD) diet for 12 days. One-half of each group of rats was fasted for 24 hours prior to death. Hepatic enzyme activities, plasma and liver magnesium, and whole blood glucose were measured. Activities o…

Cell population kinetics and biochemical changes in the rat stomach during magnesium deficiency.

Cell populations of rat stomach have been counted following varying (4-60 days) periods of magnesium-deficient diet and compared to a control group. The activity of beta-glucuronidase and the serotonin concentration were assayed in magnesium-deficient and control rats within four weeks. In the rat stomach the magnesium deficiency produces a numerical decrease in mucous cells, especially marked after 3 and 4 weeks. At this time, the activity of beta-glucuronidase decreases significantly. The concentration of serotonin increases at an earlier time and this increase coincides with the onset of th…

Dual-Functional Magnesium-Doped Zeolitic Imidazolate Framework-8 Nanoparticles for Rice Blast Control and Restoration of Magnesium-Deficient Photosynthesis.

Conventional chemical control of rice blast often leads to increased pathogen resistance and environmental challenges. Therefore, developing efficient and eco-friendly alternatives has become a key focus in rice blast management. In this study, magnesium-doped zeolitic imidazolate framework-8 nanoparticles (Mg-ZIF-8 NPs) were developed to deliver multiple agronomic benefits, including potent antimicrobial activity, enhanced photosynthetic efficiency and improved plant immunity. The Mg-ZIF-8 NPs, with an average size of approximately 314.50 nm, induced reactive oxygen species production in Magn

The magnesium-specific uptake and translocation transporters ZmMGT10 and MGR6 are upregulated not only by magnesium deficiency but also by high potassium concentrations in maize.

The antagonism between potassium (K+) and magnesium (Mg2+) is the primary cause of Mg2+ deficiency worldwide. Recent studies have demonstrated that the suppressive effect of K+ on Mg2+ uptake is significantly reduced as the K+/Mg2+ ratio increases, as both cations share non-specific cation channels. Concomitantly, the relative root/shoot translocation of Mg2+ increased. In contrast, there are indications that elevated tissue [K+] impedes the primary physiological functions of Mg2+. In this study on Zea mays L., the involvement of the only known specific Mg2+ uptake transporter ZmMGT10 and that

Thermostability of the PYL-PP2C Heterodimer Is Dependent on Magnesium: In Silico Insights into the Link between Heat Stress Response and Magnesium Deficiency in Plants.

Magnesium deficiency increases the susceptibility of plants toward heat stress. The correlation between magnesium levels and stress response has been studied at the physiological level; albeit, the molecular explanation to this relationship remains elusive. Among diverse pathways implicated in the heat stress, the abscisic acid (ABA) signal modulates the heat stress response by magnesium dependent phosphatases (PP2Cs). Exclusively, sequestration of PP2Cs by ABA receptors (PYLs) in the heterodimer form activates the stress response through ABA responsive transcription factors. In this study, th…

Normalization of magnesium deficiency with magnesium-L-Threonate alleviates bladder overactivity in cyclophosphamide-induced cystitis: Mechanisms of inflammatory modulation and barrier restoration.

Bladder Pain Syndrome/Interstitial Cystitis (BPS/IC) is a chronic urological syndrome predominantly affecting women, with an incidence of 2.7-6.5%. It is characterized by bladder-related pain and lower urinary tract symptoms, with inflammation and urothelial barrier disruption as primary mechanisms. Our previous research demonstrated that normalization of magnesium deficiency with Magnesium-L-Threonate (L-TAMS) alleviates mechanical allodynia, depressive-like behaviors, and memory deficits in cyclophosphamide (CYP)-induced cystitis by suppressing neuroinflammatory responses. In the present stu…

Association of Magnesium Deficiency and Reduction in Blood Pressure After Chemotherapy in Previously Hypertensive Cancer Patients: The Role of Chemotherapy and Magnesium Levels.

Background and Objectives: A commonly observed phenomenon in outpatient oncological patients is the appearance of hypotension not attributab … l vascular fragility induced by oncological drugs, leading to reduced vascular resistance. Although magnesium deficiency is generally linked to hypertension, chemotherapy-related shifts in magnesium levels due to impaired renal handling may play a role. These findings may help improve the understanding of blood pressure regulation in oncology patients.

Protective effect of coexistent thiamine deficiency upon the experimental cardiomyopathy associated with acute magnesium deficiency in the Syrian golden hamster.

Repeated efforts to induce beriberi heart disease by experimental thiamine deficiency (B1d) have failed in many species. To test the hypothesis that magnesium deficiency (Mgd) might be the cofactor necessary for heart failure, 10-week-old Syrian golden hamsters were divided into four groups-control (C), B1d, Mgd, and combined MgB1d-and were fed the diets ad libitum for 3 weeks. On day 21, animals were studied under intraperitoneal pentobarbital anesthesia (50 mg/kg). Electrocardiograms were taken and right and left ventricular pressures were measured by transthoracic needle puncture. Cardiac o…

The Concurrent Association of Magnesium and Calcium Deficiencies with Cognitive Function in Older Hospitalized Adults.

Background/Objectives: Hypomagnesemia and hypocalcemia are common conditions among older adults that may contribute to cognitive decline. However, most of the existing research has focused primarily on dietary intake rather than the actual serum levels of these nutrients or examined them separately. This study aims to investigate the relationship between hypomagnesemia, hypocalcemia, and the concurrent presence of both deficiencies in relation to cognitive performance among seniors. Methods: A total of 1220 hospitalized patients aged 60 and older were included in the analysis. The participants

Cardiolipin deficiency leads to the destabilization of mitochondrial magnesium channel MRS2 in Barth syndrome.

Barth syndrome (BTHS) is a debilitating X-linked cardio-skeletal myopathy caused by loss-of-function mutations in TAFAZZIN, a cardiolipin (CL)-remodeling enzyme required for the maintenance of normal levels of CL species in mitochondrial membranes. At present, how perturbations in CL abundance and composition lead to many debilitating clinical presentations in BTHS patients have not been fully elucidated. Inspired by our recent findings that CL is essential for optimal mitochondrial calcium uptake, we measured the levels of other biologically important metal ions in BTHS mitochondria and found
