Motor effects of deep brain stimulation correlate with increased functional connectivity in Parkinson's disease: An MEG study.

Deep brain stimulation (DBS) of the subthalamic nucleus (STN) is an established symptomatic treatment in Parkinson's disease, yet its mechanism of action is not fully understood. Locally in the STN, stimulation lowers beta band power, in parallel with symptom relief. Therefore, beta band oscillations are sometimes referred to as "anti-kinetic". However, in recent studies functional interactions have been observed beyond the STN, which we hypothesized to reflect clinical effects of DBS. Resting-state, whole-brain magnetoencephalography (MEG) recordings and assessments on motor function were obt…

Non-linear transfer characteristics of stimulation and recording hardware account for spurious low-frequency artifacts during amplitude modulated transcranial alternating current stimulation (AM-tACS).

Amplitude modulated transcranial alternating current stimulation (AM-tACS) has been recently proposed as a possible solution to overcome the pronounced stimulation artifact encountered when recording brain activity during tACS. In theory, AM-tACS does not entail power at its modulating frequency, thus avoiding the problem of spectral overlap between brain signal of interest and stimulation artifact. However, the current study demonstrates how weak non-linear transfer characteristics inherent to stimulation and recording hardware can reintroduce spurious artifacts at the modulation frequency. T

Estimation of the reaction times in tasks of varying difficulty from the phase coherence of the auditory steady-state response using the least absolute shrinkage and selection operator analysis.

Quantitative estimation of the workload in the brain is an important factor for helping to predict the behavior of humans. The reaction time when performing a difficult task is longer than that when performing an easy task. Thus, the reaction time reflects the workload in the brain. In this study, we employed an N-back task in order to regulate the degree of difficulty of the tasks, and then estimated the reaction times from the brain activity. The brain activity that we used to estimate the reaction time was the auditory steady-state response (ASSR) evoked by a 40-Hz click sound. Fifteen heal

From the editors: Epilepsia's survey on the necessity of the Wada test and intracranial electrodes for cortical mapping.

OBJECTIVE: From April 4 to August 1, 2014, Epilepsia conducted an online survey seeking opinions related to the use of the Wada test and intracranial electrodes in localizing cortical functions for epilepsy surgery patients. This study reports the findings of that poll. METHODS: The survey consisted of six questions. Two questions addressed: (1) If a Wada test was always necessary for patients with temporal lobe epilepsy; and (2) if magnetoencephalography (MEG) and functional magnetic resonance imaging (fMRI) data could replace intracranial electrodes in localizing cortical motor-sensory and …

MEG premotor abnormalities in children with Asperger's syndrome: determinants of social behavior?

Children with Asperger's syndrome show deficits in social functioning while their intellectual and language development is intact suggesting a specific dysfunction in mechanisms mediating social cognition. An action observation/execution matching system might be one such mechanism. Recent studies indeed showed that electrophysiological modulation of the "Mu-rhythm" in the 10-12Hz range is weaker when individuals with Asperger's syndrome observe actions performed by others compared to controls. However, electrophysiological studies typically fall short in revealing the neural generators of this

Meg recordings of patients with CNS disorders before and after external magnetic stimulation.

Magnetoencephalographic (MEG) recordings of patients with CNS disorders were obtained using a whole-head 122-channel magnetometer SQUID and analyzed using Fourier statistical analysis. External transcranial magnetic stimulation in the order of pico Tesla (pTMS) was applied to the above patients with proper characteristics (magnetic intensity 1-7.5 pT; frequency: 8-13 Hz) which were obtained with MEG recordings prior to pTMS. The MEG recordings after the application of pTMS showed a rapid attenuation of the high abnormal activity followed by an increase in the number of the low frequency compon

Neural dynamics for facial threat processing as revealed by gamma band synchronization using MEG.

Facial threat conveys important information about imminent environmental danger. The rapid detection of this information is critical for survival and social interaction. However, due to technical and methodological difficulties, the spatiotemporal profile for facial threat processing is unknown. By utilizing magnetoencephalography (MEG), a brain-imaging technique with superb temporal resolution and fairly good spatial resolution, Synthetic Aperture Magnetometry (SAM), a recently developed source analysis technique, and a sliding window analysis, we identified the spatiotemporal development of …

Comparison of clonidine to sleep deprivation in the potential to induce spike or sharp-wave activity.

OBJECTIVE: This study aimed to investigate previously observed side effects, i.e. increased epileptic activity during clonidine medication. The safety and effectiveness of clonidine as spike inducing agent compared to sleep deprivation were tested. METHODS: Patients suffering from drug-resistant localization related epilepsy took part in 3 magnetoencephalography (MEG) sessions. One session was recorded without any activating measures. The other two sessions were either performed after sleep deprivation or after medication with clonidine. Target parameter was the number of spikes or sharp-wave…

Functional anatomy of motor recovery after early brain damage.

Functional magnetic resonance imaging and transcranial magnetic stimulation were used to examine a 34 year-old right-handed patient, who, at the age of 6 years, had experienced sudden right hemiplegia, seizures, and stupor during a bout of measles encephalitis, followed by incomplete distal right motor recovery. Morphological MRI showed massive unilateral enlargement of the left ventricle, associated with extreme thinning of the white and gray matter, with partial preservation of the pyramidal tract. Functional MRI and transcranial magnetic stimulation revealed reorganization of the motor corti

Neuroplasticity of auditory cortex after stape surgery for otosclerosis: a magnetoencephalographic study.

Aim of the present study was to investigate the tonotopic reorganization of the primary auditory cortex in otosclerotic patients following functional stapedioplasty. Characteristics of auditory cortex activation have been evaluated in a series of 10 otosclerotic patients before and after surgery. In these patients, a magnetoencephalographic recording of evoked magnetic fields has been performed by means of tone-burst monoaural stimulation with frequency octaves between 250 and 2000 Hz. Brain topography of cortex response generators (wave N100m) in patients with otosclerosis has been compared w

Analysis of pathways mediating preserved vision after striate cortex lesions.

This study investigated the neural substrates of preserved visual functioning in a patient with homonymous hemianopsia and Riddoch syndrome after a posterior cerebral artery stroke affecting the primary visual cortex (area V1). The limited visual abilities of this patient included above-chance verbal reports of movement and color change as well as discrimination of movement direction in the hemianopic field. Functional magnetic resonance imaging showed that motion and color-change stimuli presented to the hemianopic field produced activation in several extrastriate areas of the lesioned hemisp

Cortical responses to object-motion and visually-induced self-motion perception.

We investigated the spatiotemporal cortical dynamics during the perception of object-motion and visually-induced self-motion perception in six normal subjects, using a 143-channel neuromagnetometer. Object-motion specific tasks evoked early transient activity over the right temporooccipital cortex, while self-motion perception, or vection, additionally was followed by sustained bilateral activity in the temporoparietal area. The specific signal distributions suggest to represent the different perceptual modes of object-motion and self-motion sensation.

Representation of the ear in human primary somatosensory cortex.

We studied 13 healthy subjects with a multichannel magnetoencephalography (MEG) system to investigate the somatotopic representation of the ear in the primary somatosensory cortex (SI). We stimulated three parts of the left ear: the helix, the lobulus, and the tragus. The somatosensory-evoked magnetic fields (SEFs) were successfully measured in 7 of 13 subjects. Short-latency responses were analyzed using both single dipole and multidipole models (brain electric source analysis, BESA). From the single dipole model, the equivalent current dipole (ECD) following the helix stimulation was estimat…

Activation of the human occipital and parietal cortex by pattern and luminance stimuli: neuromagnetic measurements.

We compared cortical reactivity to pattern and luminance stimuli by recording evoked responses and spontaneous brain rhythms from 10 subjects with a whole-scalp neuromagnetometer. Hemifield patterns (black-and-white checkerboards) elicited strong contralateral transient activation of the occipital V1/V2 cortex, maximum at 65-75 ms, followed by sustained activation during the 2 s stimulus. Responses to hemifield luminance stimuli also had an occipital component, but they were dominated by activation of the medial parieto-occipital sulcus (POS) 60-70 ms later. The POS region was equally well act

Noninvasive detection of cerebral plasticity in adult human somatosensory cortex.

After upper limb deafferentation, adult macaques exhibit substantial reorganization of cortical somatosensory topography with enlargement of cortical areas responsive to facial stimuli. In the present study non-invasive magnetic source imaging technology has been used to map in detail the bilateral somatosensory homunculi in four neurologically normal controls and two upper arm amputees. Bilateral homuncular maps of normals and of the unaffected hemisphere of both amputees showed a wide hand area. The affected hemisphere of both amputees showed marked intrusion of facial representations into t

Landau-Kleffner syndrome: epileptic activity in the auditory cortex.

The Landau-Kleffner syndrome (LKS) is characterized by electroencephalographic spike discharges and verbal auditory agnosia in previously healthy children. We recorded magnetoencephalographic (MEG) spikes in a patient with LKS, and compared their sources with anatomical information from magnetic resonance imaging. All spikes originated close to the left auditory cortex. The evoked responses were contaminated by spikes in the left auditory area and suppressed in the right--the latter responses recovered when the spikes disappeared. We suggest that unilateral discharges at or near the auditory c

Frequency-Resolved Cortical Functional Connectivity Across the Adult Lifespan.

The operation of the human brain relies on functional networks enabled by inter-areal oscillatory synchronization between neuronal populations. Although disruptions in this functional connectivity are associated with brain disorders, evidence on its healthy age-dependent variation and behavioral relevance remains limited. Utilizing magnetoencephalography (MEG) recordings from 576 adults aged 18-87 years, we investigated the evolution of resting-state functional connectivity (rs-FC) across the healthy adult lifespan. We observed age-related, frequency-specific changes in widespread cortical net…

Resting-State Network Transitions in Temporal Lobe Epilepsy: Insights From MEG-Based Dynamic Functional Connectivity.

Temporal Lobe Epilepsy (TLE), a common form of focal epilepsy, is associated with recurrent seizures originating in the temporal lobe, often leading to cognitive and psychological impairments. This study explores dynamic functional connectivity (dFC) patterns in TLE patients compared to Healthy Controls (HC) using resting-state Magnetoencephalography (MEG) data. dFC, which captures the temporal variability of brain networks, was analyzed across eight frequency bands (delta, theta, alpha, beta, low gamma, mid gamma, high gamma, and broadband) in 21 TLE patients and 21 HC. Nine dFC metrics, incl…

Large-scale neural network correlates of response inhibition associated with antidepressant response in major depressive disorder.

BACKGROUND: A fundamental impairment of major depressive disorder (MDD) is response inhibition, which may serve as a predictor for inadequate responses to antidepressants. Nevertheless, the neurophysiological connections between treatment outcomes and response inhibition are not yet fully understood. METHODS: This study involved 149 participants, consisting of 77 healthy controls (HCs) and 72 patients with MDD. All individuals undertook a Go/No-go task while undergoing magnetoencephalography (MEG) recording. Those patients with MDD showing at least a 50% decrease in symptom severity (the shor…
