Development of amorphous solid dispersion of delamanid, an extremely water-insoluble drug, by hot melt extrusion applying acid-base supersolubilization (ABS) principle.

Delamanid, an antituberculosis drug used for multidrug-resistant tuberculosis, has extremely poor water solubility and a high melting point of ∼195 °C. The current commercial product, Deltyba®, shows incomplete dissolution and food-dependent bioavailability. This study is the first to report the preparation of amorphous solid dispersions (ASD) of delamanid by hot-melt extrusion (HME), made possible by applying the acid-base supersolubilization (ABS) principle using malic acid. Malic acid increased the aqueous solubility of delamanid to 46 mg/mL at pH 1.8, approximately 660 times higher than it

Patagonian red wines: selection of Lactobacillus plantarum isolates as potential starter cultures for malolactic fermentation.

The aim of this study was to evaluate fifty-three Lactobacillus plantarum isolates obtained from a Patagonian red wine, molecularly identified and typified using RAPD analysis, in order to select starter cultures for malolactic fermentation (MLF). The results obtained suggest a considerable genetic diversity, taking into account that all L. plantarum isolates were obtained from one cellar and one vintage. Based on the capacity to tolerate a concentration of 14 % ethanol in MRS broth for 2 days, eight isolates were selected for the subsequent analysis. The incidence of various wine stress facto

UVB irradiation alters the activities and kinetic properties of the enzymes of energy metabolism in rat lens during aging.

Ultraviolet (UV) radiation is one of the major risk factors of cataract (loss of eye-lens transparency). The influence of UVB radiation (300 nm; 100 microW cm-2) on the activity and apparent kinetic constants (Km and Vmax) of rat lens hexokinase (HK;EC 2.7.1.1), phosphofructokinase (PFK; EC 2.7.1.11), isocitrate dehydrogenase (ICDH; EC 1.1.1.41) and malate dehydrogenase (MDH; EC 1.1.1.37) of energy metabolism has been investigated by irradiating the lens homogenate of three- and 12-month-old rats. In the three-month-old group specific activities of HK and PFK are reduced by 56 and 43%, respect

Pathway of glucose catabolism in Caulobacter crescentus.

Glucose when present as a sole organic carbon source in a mineral salts medium is dissimilated by Caulobacter crescentus ATCC 15252 (strain CB-2) by the Entner-Doudoroff pathway throughout the culture cycle (exponential, transition, and stationary phase). Most of the available glucose that is present at the onset of exponential growth is assimilated by the cells during the transition phase or the period associated with stalk cell development. Swarmer cell development is minimized during this phase. During this same period the pH drops from 6.1 to 4.9 as a result of an abundant excretion of ace

Selective fluorescence-based sensing of short-chain alcohols in food samples using an eco-friendly synthesis of egg white protein-malic acid stabilized copper nanoclusters.

Short-chain alcohols are widely used in food and industrial products, yet their uncontrolled presence can compromise food quality and human health, highlighting the need for rapid and reliable detection methods. In this work, a cost-effective and sustainable synthesis of stable copper nanoclusters (CuNCs) using egg white and malic acid was developed. The CuNCs exhibited significant fluorescence enhancement through the aggregation-induced emission enhancement effect in the presence of short-chain alcohols, enabling sensitive detection in aqueous solutions with low detection limits. Based on thi

Molluscicidal activity and physiological toxicity of Macleaya cordata alkaloids components on snail Oncomelania hupensis.

In order to search new local plant molluscicides for the control of the vectors of schistosomiasis, leaves of Macleaya cordata (Willd) R. Br. were used to extract and separate alkaloid components by thinner acid method and column chromatography, and the molluscicidal effect of alkaloid components against snail Oncomelania hupensis was determined by bioassay. The results showed that 7 alkaloid components (AN1-7) were obtained after extracting and separating alkaloids from the leaves of M. cordata, where AN2 was found being the most toxic against snail O. hupensis with 48h LC50 and LC90 values o

Quinone mediated electron transport system in the filarial parasite Setaria digitata.

Setaria digitata, a cattle filarial parasite, is known to have peculiarities such as hydrogen peroxide (H2O2) production, cyanide insensitivity, absence of cytochromes and presence of quinones. Estimation of mitochondrial H2O2 with different substrates and inhibitors showed that salicylhydroxamic acid (SHAM), the alternative oxidase inhibitor, inhibited the H2O2 production maximally. Based on the inhibitory studies with rotenone, antimycin A, o-hydroxydiphenyl, SHAM and 2 thenoyltrifluoroacetone, a mechanism for the electron transport is proposed. Quinone Q8 seems to have a central role, hence

The effect of changes in the respiratory metabolism upon genome activity. A correlation between induced gene activity and an increase in activity of a respiratory enzyme.

The in vitro regression of experimentally induced chromosome puffs was investigated in explanted salivary gland chromosomes of Drosophila hydei. It was observed that the regression of the puffs 2-32A, 2-36A, 2-48C, and 4-81B is accelerated if substrates for the respiratory metabolism are supplied to the cells. A similar effect can be produced by addition of KCN or oligomycin to medium in which intact salivary glands are incubated. The acceleration of puff regression by these substances occurs not only if the puff-inducing stimulus is removed but as well under conditions in which the stimulus i

Formation of alpha-hydroxyglutaric acid by Aspergillus glaucus.

Aspergillus glaucus, cultured on sodium propionate-mineral salts medium, incorporates (14)C-glyoxylate into labeled alpha-hydroxyglutaric acid within 30 sec. Mycelial extracts retain this biosynthetic capacity, which is destroyed by heating. Propionyl-2-(14)C-coenzyme A also in incorporated into labeled alpha-hydroxyglutaric acid by these mycelial extracts, but to a more limited extent. (14)CO(2) evolution studies, employing differentially labeled (14)C-propionate, indicate C-1 is oxidized by the mold before C-2, and C-2 before C-3. These findings suggest the involvement of alpha-hydroxyglutar

Salecan improves diet-induced obesity and hepatic steatosis by regulating citrate cycle intermediates.

Salecan, a naturally occurring β-glucan, represents an approved novel food ingredient in China. Although its numerous health benefits are acknowledged, the underlying mechanisms addressing obesity and hepatic steatosis remain uncertain. This investigation integrated a mouse model of diet-induced obesity with clinical observations in obese children to investigate the effects and underlying mechanisms of Salecan. In the animal model, obese mice were administered Salecan by gavage for 8 weeks. Salecan significantly promoted weight loss, reduced blood glucose levels, and improved insulin resistanc

1H NMR Approach for Evaluating the Effects of a Natural Detergent on Olive Trees Infected by Xylella fastidiosa subsp. pauca.

X. fastidiosa subsp. pauca (Xfp) is the etiological agent of "Olive Quick Decline Syndrome" (OQDS). Cellina di Nardò (Olea europaea L., Oleaceae), one of the major Salento cultivars, is highly susceptible to Xfp, usually showing acute symptoms after infection. NuovOlivo® a plant-derived formulation made with vegetal oils and water infusion from multi botanical species has been reported as effective against OQDS in plants affected by Xfp. A non-targeted 1H NMR (Nuclear Magnetic Resonance) fingerprinting approach, with unsupervised and supervised analysis, was applied to observe the possible cha

RNA-Seq Profiling Identifies Temperature-Responsive Genes in Germinating Urediniospores of Puccinia striiformis f. sp. tritici Race CYR34-8.

Wheat stripe rust (yellow rust), a devastating disease caused by the phytopathogen Puccinia striiformis f. sp. tritici (Pst), poses a major threat to global wheat (Triticum aestivum L.) production. Under ongoing climate warming, the highly virulent Pst race CYR34 has become predominant in China and exhibits increased tolerance to elevated temperatures. Although temperature is known to influence Pst urediniospore germination, the molecular mechanisms underlying temperature sensitivity during this process remain poorly understood. In this study, we combined histological examinations with transcr

Sip4p Regulates the Transcription of Enzymes for Gluconeogenesis in Nakaseomyces glabratus.

To survive in glucose poor niches within the host, the opportunistic pathogen Nakaseomyces glabratus must assimilate alternative carbon sources. In many yeasts, pathways for alternative carbon metabolism are regulated at the transcriptional level by Zn2Cys6 transcription factors, a class of proteins that are unique to fungi. Here, we investigated the transcriptional regulation of alternative carbon metabolism pathways by the Zn2Cys6 transcription factors Cat8p, Sip4p, and Rds2p in the opportunistic pathogen N. glabratus. Phenotypic screens of transcription factor knockout strains grown on alte

Regulation of airway fumarate by host and pathogen promotes Staphylococcus aureus pneumonia.

Staphylococcus aureus is a leading cause of healthcare-associated pneumonia, contributing significantly to morbidity and mortality worldwide. As a ubiquitous colonizer of the upper respiratory tract, S. aureus must undergo substantial metabolic adaptation to achieve persistent infection in the distinctive microenvironment of the lung. We observed that fumC, which encodes the enzyme that converts fumarate to malate, is highly conserved with low mutation rates in S. aureus isolates from chronic lung infections. Fumarate, a pro-inflammatory metabolite produced by macrophages during infection, is
