Measles encephalomyelitis--clinical and immunologic studies.

We studied 19 patients with postinfectious encephalomyelitis complicating natural measles-virus infections, and our results support the hypothesis that this demyelinating disease has a pathogenesis similar to that of experimental allergic encephalomyelitis. Early myelin destruction was demonstrated by the presence of myelin basic protein in cerebrospinal fluid, and lymphocyte proliferative responses to myelin basic protein were found in 8 of 17 patients tested. A lack of intrathecal synthesis of antibody against measles virus suggests that measles encephalomyelitis may not be dependent on viru…

[Congenital german measles encephalitis (author's transl)].

German measles is an infections exanthematous disease, long regarded as completely harmless. The clinical symptoms are banal and often go unnoticed. In complete contrast is the severe malformation of the embryo after infection early on in pregnancy, first discovered by Gregg in 1941. The virus infects the embryo via the placenta and either causes death or impairs the differentiation of embryonic tissues, causing malformation. What is understood today under the congenital German measles syndrome is considerably more extensive and includes, in addition to malformation, growth retardation, premat…

Cell-mediated immune reactions in measles.

Cell-mediated reactions in measles cases, in direct contacts and healthy children were tested. Indices of phytohaemagglutinin-stimulated blastic transformation and leukocyte migration inhibition were evaluated. Positive reactions were found in the first and second weeks after manifestation of clinical symptoms. The application of glucocorticosteroids in clinical complications resulted in delayed and reduced leukocyte migration inhibition. Studies on seronegative contacts suggested that positive cell-mediated reactions may appear before manifestation of clinical symptoms.

Radioimmunoassay of measles virus nucleocapsid antigen.

A double antibody competitive binding radioimmunoassay (RIA) was developed as a tool for investigating the involvement of measles virus in persistent virus infections. The assay employs 125I-labelled measles virus nucleocapsids as the labelled antigen. Nucleocapsids were purified from cytoplasmic extracts of virus-infected Vero cells, treated with trypsin to prevent clumping and iodinated to a specific activity of about 15 microCi/microgram. Electrophoretic analysis of iodinated trypsin-treated nucleocapsids revealed only labelled polypeptide with a relative mol. wt. (Mr) 40 000, indicating th…

Persistent measles infection in malnourished children.

Thirty malnourished and 25 well-nourished children were studied six to 31 days after the onset of a measles rash. Evidence of the virus was found in 40% of the malnourished children but in none of the well-nourished controls. Giant cells were found in the nasal secretions of five out of 17 malnourished children and measles antigen was detected in the lymphocytes of eight out of 28. The malnourished children showed depressed cell-mediated immunity to measles and candida antigens and a low response to meningococcal vaccine. Fifteen died from intercurrent infections. Malnutrition was thought to h…

Does measles really predispose to tuberculosis?

In recent years, textbooks and reviews on mechanisms involved in cellular immunity have emphasized that measles adversely influences tuberculous lesions. The medical literature was critically investigated for the basis of this claim. All field studies reviewed possessed strong deficiencies in the pertinent data and the methods of their collection. These deficiencies rendered invalid conclusions concerning the ability of measles to activate tuberculosis. Indirectly, there are also no adequate epidemiologic data to support the concept that measles suppresses tuberculous immunity when it suppress…

Revaccination against measles--a pilot study.

(1) During the first ten years after primary vaccination with live measles virus vaccine, a slow decline in the level of HAI antibody titres has been demonstrated. The decline after more attenuated vaccines was at the same gradient, but at lower titres.(2) During the same period after primary vaccination by inactivated (; killed ') followed by live - K + L - measles vaccine, a much steeper decline in the level of HAI antibody titres was demonstrated.(3) The GMT may not reach the lowest standard titres for over 40 years after the least attenuated vaccine, and 20-25 years after more attenuated v…

Experimental measles encephalitis: a genetic analysis.

The encephalitogenic potential of nine temperature-sensitive mutants of measles virus was determined in newborn golden Syrian hamsters. The parental virus produced acute encephalitis without any prior adaptation. Six of the mutants were attenuated, two were virulent, and one was associated with hydrocephalus with acute onset. The attenuated mutants, blocked before measles virus antigen and ribonucleic acid synthesis at 39 C, were all members of one complementation group. The virulent temperature-sensitive mutants, defective in hemolysin antigen synthesis at 39 C, were members of a second compl…
